Özet
Konya Melike Hatun Pazarındaki satıcılardan 2008-2009 yıllarında satın alınan bazı sebze tohumlarındaki (bamya, biber, domates, ıspanak, kabak, karpuz, kavun, marul, pırasa ve salatalık) fungal floranın tespiti ve tanılanması için blotter ve agar yöntemleri kullanılmıştır. Her iki yöntemle de izole edilen funguslar, genus ve özellikle de tür düzeyinde teşhis edilebilmeleri için x40 büyütmeli ışık mikroskobunda fungal flora bakımından incelenmiştir. Satıcılardan temin edilen örneklerde blotter yöntemine göre, patojen fungus cins veya tür sayısının, agar (PDA) yöntemine göre daha az olduğu belirlenmiştir. Blotter yönteminde en yaygın fungus cins veya türünün bamya tohumunda Pythium spp.; biber tohumunda Sclerotinia spp.; domates ve marul tohumunda Botrytis cinerea; hıyar, kabak ve karpuz tohumunda Penicillium spp.; ıspanak ve pırasa tohumunda Fusarium culmorum ve kavun tohumunda ise, Macrophomina phaseolina olduğu belirlenmiştir. Ayrıca, blotter yöntemine göre, fungal mikroorganizmalar ile en fazla bulaşık olan tohum marul tohumu olarak saptanmış ve bulaşıklık oranı %70.65 olarak bulunmuştur. Benzer şekilde, satıcılardan temin edilen örneklerde en yaygın fungus cins veya türü, bamya, biber, domates, ıspanak, marul ve pırasa tohumlarında, blotter yönteminde olduğu gibi agar (PDA) yönteminde de aynı çıkmıştır. Diğer sebze tohumlarından ise, hıyar, kabak ve kavunda Fusarium solani; ıspanakta Fusarium culmorum; karpuzda Alternaria spp. en yaygın fungus cins veya türü olarak tespit edilmiştir. Bununla birlikte, agar (PDA) yönteminde de blotter yönteminde olduğu gibi fungal mikroorganizmalar ile en fazla bulaşık tohum marul tohumu olarak belirlenmiş ve bulaşıklık oranı %99.46 olarak bulunmuştur.
Blotter and agar (PDA) method was used for some vegetable seeds (okra, pepper, tomato, cucumber, spinach, squash, watermelon, melon, lettuce, leek) bought from sellers of Konya Melike Hatun Bazaar in 2008-2009 to determine and identify fungal flora. Fungi growing both blotter and agar (PDA) method to determine for genus and especially species were investigated in x 40 enlarged light microscope. Less fungi genus or species were isolated from seeds used for blotter method compared to agar (PDA) method. According to blotter method, the most common genus or species was Pythium spp. in okra seeds; Sclerotinia spp. in pepper seeds; Botrytis cinerea in tomato and lettuce seeds; Penicillium spp. in cucumber, squash and watermelon seeds; Fusarium culmorum in spinach and leek seeds and Macrophomina phaseolina in melon seeds. Also, according to blotter method, the most infectious seed with fungal microorganism was found lettuce and its infectious rate was counted as %70.65. Especially, in agar (PDA) method, the most common fungi genus or species was same in okra, pepper, tomato, spinach, lettuce and leek seeds as blotter method. The most common genus or species for other vegetable seeds was Fusarium solani in cucumber, squash and melon; Fusarium culmorum in spinach and Alternaria spp. in watermelon. Moreover, the most infectious seed with fungal microorganism in agar (PDA) method was found lettuce as well blotter method and its infectious rate was counted %99.46.
