Özet
Bu çalışmada Ulex europaeus bitkisinden elde edilen Ulex europaeus aglutinin-I (UEA-I)'in Rh (-) insan kan grubu bağlama özgüllüğü araştırıldı. Bu deney için A Rh (-), B Rh (-), AB Rh (-) ve O Rh (-) kan grubu eritrositleri kullanıldı. Phosphate buffered saline (PBS) ile yıkanan eritrositlerin %3' lük süspansiyonları hazırlandı. 100 ?g/ml lektin solüsyonu PBS ile hazırlanarak başlangıç konsantrasyonu olarak alındı ve UEA-I' in seri dilüsyonu ile Rh (-) insan kan grubu özgüllüğü belirlendi.Yapılan hemaglutinasyon deneyinde UEA-I'in A Rh (-) insan kan grubu eritrositlerini 50 ?g/ml konsantrasyonda, B Rh (-) kan grubu eritrositleirini 25 ?g/ml konsantrasyonda, AB Rh (-) kan grubu eritrositlerini 50 ?g/ml konsantrasyonda, O Rh (-) kan grubu eritrositlerini ise 6.25 ?g/ml konsantrasyonda aglutine ettiği gözlenmiştir. Bu konsantrasyonlara bakılarak, UEA- I'in O Rh (-) grubu insan kanı eritrositlerine karşı spesifik olduğu tespit edildi.
In this study, human Rh (-) blood group binding specificity of UEA-I lectin from Ulex europaeus was researched. For this experiment A Rh (-), B Rh (-), AB Rh (-) and O Rh (-) blood group erytrocytes were used. %3 Erytrocytes solutions were prepared by using Phosphate buffered saline (PBS). 100 ?g/ml lectin solution prepared with PBS, was taken as a begining concentration and human blood group specificity of UEA-I was determined with the serial dilution of lectin. Hemagglutination experiment was observed that the UEA-I agglutinated the %3 A Rh (-) group erythrocytes at the concentration of 50 ?g/ml, B Rh (-) group erythrocytes at the concentration of 25 ?g/ml, AB Rh (-) group erythrocytes at the concentration of 50 ?g/ml and O Rh (-) group erytrocytes at the concentration of 6.25 ?g/ml. It was determined that UEA-I was specific for O Rh(-) human blood group erytrocytes.
