Ergin ve ergin olmayan O. mykiss karaciger lipazını saflastırma islemleri 3 adımdan olusmustur: Homojenatın hazırlanması, amonyum sülfat çöktürmesi, Diethylamino ethyl (DEAE) selülozik iyon degisim kromatografisi. Saflastırma islemleri boyunca, ergin balıkların karacigerinden elde edilen lipaz enziminin spesifik aktivitesi 2.23±1.59 ?mol/mg protein, verimi %0.037 ve saflastırma katsayısı 2.65±1.94 olarak belirlenmistir. Ergin olmayan balıklarda ise spesifik aktivite 1.86±0.14 ?mol/mg protein, verim %3.24±0.51 ve saflastırma katsayısı 15.50±1.13 bulunmustur.In this study, determination of liver lipase activity of mature and immature Oncorhynchus mykiss has been purposed. Liver lipase (E.C 3.1.1.3) purification process from mature and immature O. mykiss consisted of three steps: Preparation of homogenate, ammonium sulfate precipitation, Diethylamino ethyl (DEAE) cellulose ion exchange chromatography. Through the purification procedure, liver lipase enzyme of mature fish was purified with a yield of 0.037± 0.06% and a purification coefficient of 2.65±1.94, having the spesific activity of 2.23±1.59 ?mol/mg protein.
