Öğe Determination of multiple antioxidant activities of endemic tricholoma anatolicum(2018) Taşkın, Hatıra; Eker, Tülin; Bozok, Fuat; Doğan, Hasan Hüseyin; Büyükalaca, SaadetTricholoma anatolicum H.H. Doğan & Intini is known as the most prized mushroomspecies in Feke region of Turkey. This mushroom species is collected from Cedar(Cedrus libani) forests and therefore is named as Cedar mushroom in this region. It iscollected and consumed by local collectors and also exported to Far East countries suchas Japan by exporting companies. Therefore, it is important to investigate the nutritionaland medical importance of this species. The aim of this study is to reveal the antioxidantactivity of methanol extract in different concentrations (1, 2 and 4 mg/mL) of T.anatolicum H.H. Doğan & Intini collected from Feke district of Adana province ofTurkey in 2015. In this study, it was determined that total phenolic content of thismushroom was 56 mg/kg. 2,2-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide (NO) andreducing power (RP) activities given in trolox (?M) and ferric-reducing antioxidantpower (FRAP) given in FeSO4 (?M) were found as 449, 180, 337 and 2 at the highestconcentration (4 mg/mL), respectively. As a result, it could be suggested that methanolextract of T. anatolicum H.H. Doğan & Intini has significantly antioxidant activity.Öğe Investigation of antioxidant potential and phytotoxic effects of ganoderma lucidum methanol extract(2016) Bozok, Fuat; Eker, Tülin; Sezer, Gökhan; Bozdoğan, Adnan; Doğan, Hasan Hüseyin; Büyükalaca, SaadetThe aim of this study is to reveal allelopathic effects and antioxidant activities of methanolic extract of Ganoderma lucidum in Zorkun plateau (Osmaniye). It was determined that methanolic extract of G. lucidum has significantly phytotoxic effect by increasing the doses (1, 2, 4, 8 mg/mL) on Hordeum vulgare and Triticum aestivum. Total phenol and flavonoid amounts of G. lucidum were 114.55 mg/kg and 8.95 mg/kg, respectively. 2,2-diphenyl-1-picrylhydrazyl (DPPH), scavenging activity (60%, 91% and 92%), reducing power (0.83, 1.43 and 2.23 Abs), and nitric oxide (NO) scavenging activity (48%, 55% and 70%) of this mushroom at the different concentrations (1.25, 2.5 and 5 mg/mL) were determined, respectively. However, H2O2 scavenging activity of the different concentrations (0.0156-0.125 mg/mL) was found as 18%, 56%, 60% and 86%, respectively. Based on these findings, it can suggest that G. lucidum methanolic extract has significantly phytotoxic effect and antioxidant activity. The present study is the first report on the phytotoxic effects and antioxidant activities of G. lucidum which consumed and collected from Amanos Mountains.Öğe A multigene molecular phylogenetic assessment of true morels (Morchella) in Turkey(Academic Press Inc Elsevier Science, 2010) Taşkın, Hatıra; Büyükalaca, Saadet; Doğan, Hasan Hüseyin; Rehner, Stephen A.; O'Donnell, KerryA collection of 247 true morels (Morchella spp.) primarily from the Mediterranean and Aegean Regions of Southern Turkey, were analyzed for species diversity using partial RNA polymerase I (RPB1) and nuclear ribosomal large subunit (LSU) rDNA gene sequences. Based on the result of this initial screen, 62 collections representing the full range of genetic diversity sampled were subjected to multigene phylogenetic species recognition based on genealogical concordance (GCPSR). The 62-taxon dataset consisted of partial sequences from three nuclear protein-coding genes, RNA polymerase I (RPB1), RNA polymerase II (RPB2), translation elongation factor (EF1-a), and partial LSU rDNA gene sequences. Phylogenetic analyses of the individual and combined datasets, using maximum parsimony (MP) and maximum likelihood (ML), yielded nearly fully resolved phylogenies that were highly concordant topologically. GCPSR analysis of the 62-taxon dataset resolved 15 putative phylogenetically distinct species. The early diverging Elata (black morels) and Esculenta Clades (yellow morels) were represented, respectively, by 13 and two species. Because a Latin binomial can be applied with confidence to only one of the 15 species (Morchella semilibera), species were identified by clade (Mel for Elata and Mes for Esculenta) followed by a unique Arabic number for each species within these two clades. Eight of the species within the Elata Clade appear to be novel, including all seven species within the Mel-20-to-31 subclade and its sister designated Mel-25. Results of the present study provide essential data for ensuring the sustainability of morel harvests through the formulation of sound conservation policies.Öğe A New Macrofungi Record for Turkey and Asia with Molecular Characterization: Xerocomellus redeuilhii (Boletales, Basidiomycota)(Selçuk Üniversitesi, Mantarcılık Uygulama ve Araştırma Merkezi Müdürlüğü, 2019) Bozok, Fuat; Assyov, Boris; Taşkın, Hatıra; Büyükalaca, SaadetXerocomellus redeuilhii A.F.S.
