Öğe Evaluation of h?D-1 and h?D-2 levels in saliva of patients with oral mucosal diseases(University of the West Indies, 2013) Kucukkolbasi H.; Kucukkolbasi S.; Ayyildiz H.F.; Dursun R.; Kara H.Objective: This study aimed to determine a possible correlation between oral mucosal disease and salivary concentrations of the antimicrobial peptides human beta-defensin-1 (h?D-1) and human betadefensin- 2 (h?D-2). Method: The present work focussed on the establishment of a reversed phase-high performance liquid chromatography (RP-HPLC) procedure to quantify human beta-defensins (h?D-1 and h?D-2) in saliva samples of patients with oral diseases such as lichen planus (n = 10), Behçet (n = 10) and recurrent apthous stomatitis (n = 10). Results: Linear calibration range for h?D-1 and h?D-2 defensins was 1.67-200 ?g mL-1 and 3.13 - 100 ?g mL-1 with R2 values of 0.9998 and 0.996, correspondingly. The concentration of beta-defensins in saliva was determined by comparing the peak areas of eluted h?D-1 and h?D-2 with that of their standards. The variation of the amount of beta-defensins was evaluated by comparisons of the results obtained from the patients with oral mucosal diseases before and after treatments and the control subjects. The limit of detection (LOD) and limit of quantification (LOQ) were found to be 1.62 ?g mL-1 and 5.39 ?g mL-1 for h?D-1 and 0.94 ?g mL-1 and 3.13 ?g mL-1 for h?D-2, respectively. Conclusion: The salivary beta-defensin concentration was significantly higher in patients with oral mucosal diseases than in healthy volunteers; furthermore, in patients with oral mucosal diseases, the concentration was significantly higher before treatment than after treatment.Öğe Immunohistochemical analysis of Ki-67 in dental follicle of asymptomatic impacted third molars(Medknow Publications, 2014) Kucukkolbasi H.; Esen A.; Erinanc O.H.Introduction: Dental follicle (DF) is an ectomesenchymal tissue that surrounds the developing tooth germ and contains precursor cells for cementoblasts, periodontal ligaments and osteoblasts. Radiographically, the DFs are seen as semicircular radiolucencies around unerupted teeth. However, if the DFs are larger than 2.5 mm, they are considered to be a pathological change. Aims and Objectives: The purpose of this study is to assess the cell proliferation activity of DF surrounding an asymptomatic impacted third molar teeth using the Ki 67 proliferation marker and to evaluate the variation of cell proliferation depending on the age factor. Materials and Methods: Forty-four specimens of DFs associated with impacted mandibular third molars fully covered by mucosa or bone were surgically removed from 44 patients. The patients were divided into 2 age groups. Twenty of forty-four DFs were between 18 and 29 years (Group 1) and 24 were 30 years and above (Group 2). Ki-67 immunostaining was evaluated in epithelial component of the DFs. Results: Ki 67 expression was found to be 60% in Group 1 and 75% in Group 2. Statistically significant differences were found among the two groups in both the basal layer and the supra-basal layer. Conclusion: This study shows that DFs have more proliferative potential in older people as compared to the young and squamous metaplasia may be an early sign of developing lesions of odontogenic origin. Therefore, clinicians should be aware that histopathological changes could be found in DFs without clinical and radiographic alterations.Öğe Reconstruction of an unusual orocutaneous fistula by using a bilobed flap.(2008) Dolanmaz D.; Kucukkolbasi H.; Isik K.Orocutaneous fistulas or cutaneous sinus tracts of dental origin are pathologic communications between the cutaneous surface of the face and the oral cavity. They are relatively uncommon and may be easily misdiagnosed. In this report, management of an orocutaneous fistula of dental origin, which endured for 20 years, by using an extraoral bilobed skin flap, an intraoral buccal rotational mucosal flap and allogenic bone graft, is presented.
