Öğe Isolation of Ornithobacterium Rhinotracheale From Chickens and Turkeys(SCIENTIFIC TECHNICAL RESEARCH COUNCIL TURKEY, 2002) Erganiş, Osman; Ateş, Mehmet; Hadimli, H. H.; Çorlu, MehmetA total of 251 tracheal swabs and a total of 102 tracheae, lungs, airsacs, livers and spleens were collected from commercial layer, broiler and broiler parent stock flocks with respiratory infection. From turkey flocks, 96 tracheal swabs and 5 tissue samples from the trachea, lung, airsac, liver and spleen were also taken. Cultivations on Blood Agar supplemented with 51 10%sheep blood, on Chocolate Agar and on MacConkey Agar were incubated at 37degreesC for 24-72 hours in the presence of 10%CO(2)Gram staining was carried out for the smooth and non-hemolytic colonies 1-3 mm in diameter and for those showing grayish pigmentation after approximately 48 hours of culture. Cram negative pleomorphic rods were considered to be Ornithobacterium rhinotracheale (ORT) and other conventional tests were made as follows: hemolysis, motility, spore formation, growth on MacConkey and TSI agars, catalase, indol, sorbitol, inositol, rafinose, trehalose, xylose, dulcitol, nitrate, methyl red, oxidase, beta-D galactosidase, urease, glucose, galactose, lactose, maltose, fructose and Voges-Proskauer. ORT was isolated from none of the samples taken from turkeys, broiler or broiler parent stock flocks. However, two ORT isolations were achieved from two commercial pullets aged 12 and 15 weeks. One of these isolates was serotyped as "B" by the ACP test while the other was not found to belong to any of the serogroups. Yavuz; Erganiş, Osman; Çorlu, MehmetIn this study, some biochemical characteristics, different antibiotics resistance states and R-factor transmissibility of 51 E. coli strains isolated from calves with diarrhea up to one week age were examined. Biochemical characteristics of the strains were found to be as follows: The rates for glucose, lactose, mannitol dulsitol, arabinose and adonitol fermentation tests were 100 %, 100%, 100%, 54.9 %, 98.1 % and 21.6 % respectively and the rates for urease production, citrate utilization, tryptophane deaminase, H&S, indol and motility tests were 100 %, 0.0 %, 0.0%, 0.0 %, 100%, 0.0 %, 96.1 % and 80.4% respectively. It was observed that only one of the whole strains (1.96 %) possessed a hemolytic activity. According to the results of disk diffusion method, antibiotic resistance rates of E. coli strains were determined as follows 70.5 % for tetracyline, 86.2% for oxytetracycline and neomisin, 98.1 % for erytromisin, 54.9% for ampisilin, 49.0% for streptomisin, 23.4 % for chloramphenicole and 5.9% for nalidixic acide. Of the strains 94.1 per cent were resistant to more than 3 antibiotics. Sheep were examined by clinical procedures and tested with California Mastitis test (CMT). Milk samples taken from sheep with mastitis (CMT scores, +++++) were cultured in aerobic, microaerophlic and anaerobic conditions for bacteriological and mycological examination bacteria isolated and identified were tested against Ampicillin, Streptomycin, Tetracyclin, Penicillin, Kanamycin, Erytromycin, Gentamycin and Chloramphenicol discs (Difco). In this area, in the mammary glands of 13.5 % of the tested sheep, mastitis was detected. Although the rates of clini cal mastitis was 0.75 percent, subclinical mastitis in sheep was 12.75 percent. Microbiologic isolation were succeded from (75.0 %) 81 samples of 108 mastitic milk. But, there was mix infection in 8 samples. The percent values of isolated microorganisms were found as below 57.3 % Staph. aureus, 9.0 % for E. coli, 7.9 % for Bacillus ssp. 5.0 % for B. cereus, 5.0 % for C. pyogenes (Actinomyces pyogenes), 3.4 % for Str. agalactiae, 3.4 % for C. pyogenes (Actinomyces pyogenes), 3.4 % for Str. agalacti- ac, 3.4 % for K. pneumoniae, 3.4 % for Candida albicans, 2.2 % for B. subtilis, 1.1 % for Str. dysgalactiae and 1.1 for Ente- robacter ssp. In antibiotic susceptibility tests, Staph. aureus isolates were found to be sensitive to Tetracyclin, Chloramphenicol and Erytromycin, at the rates of 76.0 %, 58.0 % and 38.0 %, respectively, E. coli strains were sensitive to Streptomycin (100%) and Tetracyclin (100 %), C. pyogenes strains were sensitive at the rates of 100 %, 80.0 % and 80.0 %, for Penicil- lin, Streptomycin and Chloramphenicol, respectively.Öğe Masti?ti?sli? Yavuz; Erganiş, Osman; Çorlu, Mehmet; Ateş, MehmetNineteen strains of Staph. aureus isolated from cows milk with mastitis were tested for their ability to produce coagulase, phosphatase, catalase, B-lactamase, clumping factor, oxydation/fermentation (O/F) and resistance of penicillin and ampicillin. All of the strains have produced B-lactamase and their resistance rates to penicillin and ampicillin were 94.9%, 47.9% respectively. Catalase, phosphatase and O/F producing rate of all Staph. aureus were positive 100%. The Coagulase tests found to be positive at the rate of 89.8% with rabbit plasma, 73.7% with cow plasma, 100% with mixture of rabbit-cow plasma. The rate of positive clumping factors was also found to be 89.8%, 94.9%, 100% with rabbit, cow and mix plasma, respectively. 73.7% of all strains have grown on Mannitol Salt Agar plates. 12 strains produced - golden cooured colonies. All of the 6 strains were intracutaneously inoculated to rabbits reduced necrosis.Öğe Microbiological Examination of the Tracheal Flushing Sample and its Clinical Importance(Selçuk Üniversitesi, 1989 Ocak) Turgut, Kürşat; Erganiş, Osman; Başoğlu, Abdullah; Çorlu, Mehmet; Ok, MahmutSelçuk Üniversitesi Veteriner Fakültesi İç Hastalıklar Kliniğine getirilen 25 buzağıdan mikrobiyolojik muayene için burun sıvabı ve trakeal yıkama örnekleri alındı. Burundan alınan örneklerin 22'sinden çeşitli mikroorganizmalar izole edilirken, trakeal yıkama örneklerinden P. pneumonia (% 40), Staph. aureus (% 20), Klebsiell ssp. (% 13.3), Corynebacterium ssp. (% 6.6), Shigella ssp. (% 6.6), Ps. maltophila (% 6.6), Aspergillus ssp. (% 6.6) saf olarak izole edildi. Etken izolasyonu yapılamayan buzağıların 7'sine önceden değişik antibiyotikler uygulanmıştı. Onbir vakada Linko-spektin (% 47), 9 vakada Gentamisin (% 39) etkili bulundu. Aspergillus ssp. izole edilen bir buzağı Thiabendazole ile tedavi edildi. Bir buzağı tedavi edilemedi. Bu buzağının otopsisinde mikrobiyolojik ve patolojik olarak Tüberküloz olduğu teşhis edildi. Diğer 3 buzağı geniş spektrumlu antibiyotiklerle tedavi edildi. Çalışmanın sonucunda, trakeal yıkama metodunun enfeksiyöz buzağı pneumonilerinin teşhis ve tedavilerinde kolaylıkla ve güvenilir bir şekilde uygulanabileceği kanısına varıldı.Öğe Pnömonili koyun akciğerlerinden izole edilen bakteriler ve antibiyotik duyarlılıkları(2006) Öztürk, Dilek; Çorlu, MehmetBu çalışma ile koyunlarda pnömoniye neden olan bakterilerin izolasyonu, identifikasyonu ve izole edilen bakterilerin antibiyotik duyarlılıklarının belirlenmesi amaçlandı. Konya Et-Balık Kurumu'ndan temin edilen makroskobik olarak pnömoni olabileceği düşünülen 150 koyun akciğerinden bakteriyolojik ekimler yapıldı. Örneklerin 38 (% 25.33)'inden 15 (% 25) Mannheimia haemolytica, 18 (% 30)'inden Mycoplasma spp., 2 (% 3.3)'sinden Pasteurella multocida, 9 (% 15)'undan Streptococcus spp., 10 (% 16.7)'undan Staphylococcus spp., 3 (% 5)'ünden Aeromonas hydrophilla ve birer örnekten (% 1.7) Escherichia coli, Klebsiella pneumoniaeve Pseudomonas spp. toplam 60 bakteri izole ve identifiye edildi. Mycoplasma spp. dışında izole edilen bakterilerin antibiyotik duyarlılıkları disk difüzyon yöntemi ile belirlendi. Üzeri?nde I?ncelemeler(Selçuk Üniversitesi, 1988 Ocak) Çorlu, MehmetIn this study, biochemical, colicinogenic, hemagglutination (HA), mannose sensitive hemagglutination (MSHA) and mannose resistant hemagglutination (MRHA) characteristics of 200 E. coli strains isolated from neonatal healthy and diseased lambs have been examined. There was no significant differences as to the biochemical characteristics of the strains isolated from healthy and diarrhoeic lambs. No hemolitic activity was found in all isolates. In HA tests, using different species of erytrocytes (human, bovine, chicken, guinae-pig and ovine) multiple HA patterns were observed. MRHA, MSHA and HA (-) rates of the organisms originated from healthy and diseased lambs, were found to be 28 %-32 % for MRHA, 27 %-31 % for MSHA and 45 %-37 % for HA (-). There was not significant differences in MRHA, MSHA and HA (-) characteristics of two groups.
