Öğe Comparison of culture, Real- time-PCR, ELISA, and histopathological examination methods for identification of Helicobacter pylori(2018) Maçin, Salih; Alp, Alpaslan; Şener, Burçin; Sökmensüer, Cenk; Orhan, Diclehan; Özen, Hasan; Kav, TaylanIntroduction: There are several methods used for the diagnosis of Helicobacterpylori infections, and there is an increasing demand for theuse of non-invasive, more rapid tests. The aim of the present study wasto compare different diagnostic methods.Methods: A total of 87 patients who had undergone esophagogastroduodenoscopywere included in the study. Biopsy samples obtainedfrom these patients were used for culture, real-time polymerase chainreaction (RT-PCR), and histopathological examination. Stool sampleswere also collected from these patients and were tested using the Helicobacterpylori stool antigen (HpSA) kit. Histopathological examinationwas accepted as the gold standard test.Results: H. pylori was identified by histological examination in 77/87(87.5%) patients, whereas it was negative in 10/87 (12.5%) patients. Furthermore,positive results were obtained in 55 (63.2%), 71 (81.6%), and 77(87.5%) patients using the culture method, HpSA analysis, and RT-PCR method,respectively. The sensitivity and specificity of culture, HpSA, and PCRtests were determined as 71.4% and 100%, 87% and 60%, and 97.4% and80%, respectively. Antibiotic susceptibility tests were performed on 48 outof the 55 culture positive samples. Resistance to clarithromycin was foundin 28 (58.3%), metronidazole in 14 (29.2%), and levofloxacin in 4 (8.3%) ofthe isolates. Resistance to amoxicillin and tetracycline was not observed.Conclusion: There are currently several invasive and non-invasive diagnostictests for the detection of H. pylori infections. Each test has someadvantages and disadvantages. The diagnostic method of choice shouldbe easy and applicable to all age groups.Öğe Retrospective Evaluation of Serum Zinc Levels in Patients with Protein-Energy Malnutrition(Selçuk Üniversitesi, 2023 Haziran) Özen, Hasan; Emiroğlu, Halil Haldun; Emiroğlu, Melike; Akdam, Neriman; Yorulmaz, AlaaddinAim: In this study, we aimed to investigate the frequency of serum zinc deficiency in children with protein-energy malnutrition and to show the relationship between the degree of malnutrition and serum zinc level. Methods: Methods: This retrospective study was conducted at the Pediatric Gastroenterology, Hepatology, and Nutrition clinic in Selçuk University Hospital, and included 506 patients aged between 1 and 18 years who were diagnosed with protein-energy malnutrition according to the Waterlow classification. Results: The prevalence of serum zinc deficiency was 61.1% in patients with protein-energy malnutrition. The mean serum zinc levels of patients aged 1-5 were significantly lower than those of patients aged 5-12 and 12-18. There was no difference between malnutrition degrees calculated according to Waterlow criteria and mean serum zinc levels. Although the relationship between the degree of malnutrition calculated according to the BGA scale and the mean serum zinc level was not statistically significant, the mean decrease in serum zinc levels was remarkable in contrast to the increase in the level of malnutrition. Conclusion: It is important to routinely measure serum zinc levels in patients with PEM and to provide zinc supplementation if deficiency is detected.Öğe Sığır leptospirozunun patogenezinde ubiquitin ve 8-hidroksiguanozin’in rollerinin immunohistokimyasal yöntemlerle araştırılması(Selçuk Üniversitesi Veterinerlik Fakültesi, 2010) Karaman, Musa; Özen, Hasan; Erginsoy, Serpil Dağ; Şahin, MitatAmaç: Bu çalışmada, Leptospira spp. ile doğal enfekte sığırların karaciğer ve böbrek dokularında oluşan hasarda ubiquitin ve 8-hidroksiguanozin’in (8-OHG) rolleri araştırılmıştır. Gereç ve Yöntem: Leptospirozlu (n=7) ve sağlıklı kontrol grubu (n=7) hayvanlarda, bu moleküllere spesifik poliklonal antikorlar kullanılarak indirekt immunoperoksidaz metodu ile antijenlerin dokulardaki lokalizasyonları belirlendi. Bulgular: Leptospirozlu hayvanların karaciğer hepatositleri ve böbrek tubul epitellerinde ubiquitin antikoruna karşı intrasitoplazmik immun boyanma tespit edildi. 8-OHG immun boyanmasının karaciğer hepatositlerinde intrasitoplazmik ve/veya intranükleer, böbrek tubul epitellerinde ise çoğunlukla intrasitoplazmik olarak lokalize olduğu gözlendi. Öneriler: Leptospirozun patogenezinde doku hasarı oluşumunda ubiquitin ile 8-OHG’nin rol oynadığı ortaya konularak, bu moleküllere ilişkin mekanizmaları daha detaylı anlayabilmek için ayrıntılı çalışmaların gerekliliği tavsiye edilmektedir.
