Öğe Effects of diurnal and ontogenetic variability on essential oil composition of oregano (Origanum vulgare var. hirtum)(ASIAN JOURNAL OF CHEMISTRY, 2008) Guemuescue, Ahmet; Ipek, Arif; Sarihan, Ercuement O.; Guerbuez, Bilal; Kaya, M. Demir; Arslan, NesetThis study was carried out to determine the effects of diurnal and stage of harvesting on the essential oil components of oregano (Origanum vulgare var. hirtum). The samples were collected at three different stages of flowering (before and after flowering) and six different times (06:00, 09:00, 12:00, 15:00, 18:00 and 21:00 h) with drying under shade or sunshine. Essential oil was obtained by hydrodistillation method and analyzed with GC-MS. Results showed that the main compound of O. vulgare var. hirtum was thymol. The harvest stage affected the compounds of essential oil such that p-cymene increased at full flowering while gamma-terpinene decreased. Drying methods and harvesting time showed a minor effect on volatile compounds. The highest variation in composition of essential oil in relation to harvest stage was determined in p-cymene and gamma-terpinene. It was concluded that the harvest stage along with drying under shade should be firstly considered.Öğe Evaluation of selected poppy (Papaver somniferum L.) lines by their morphine and other alkaloids contents(SPRINGER, 2008) Guemuescue, Ahmet; Arslan, Neset; Sarihan, Ercument OsmanThe alkaloid analysis was performed in the laboratory of the alkaloid factory located at Bolvadin province of Turkey in 2000 on selected 99 poppy lines, which were selected with respect to their morphological and agronomic characteristics from around 1,000 poppy lines provided by Agriculture faculty, Ankara University. Our results showed that the ratio of the alkaloids, morphine, thebaine, codeine, papaverine and noscapine ranged from 0.110 to 1.140%, 0.005 to 0.134%, 0.005 to 0.27%, 0.001 to 0.440% and 0.006 to 0.418%, respectively, in the selected 99 poppy lines. According to their alkaloid content, 15 lines had significantly higher morphine, three lines had higher noscapine, and one line had higher papaverine levels.Öğe In vitro micro-propagation of endangered ornamental plant-Neotchihatchewia isatidea (Boiss.) Rauschert(ACADEMIC JOURNALS, 2008) cue, Ahmet Guemue; Coecue, Sati; Uranbey, Serkan; Pek, Arif; kan, Mikail Cali; Arslan, NesetThe ornamental plant, Neotchihatchewia isatidea, is an endangered species of Turkey and threatened by complete extinction in the future. Therefore, in vitro multiplication of this species can be valuable for commercial production and germplasm conservation. Immature embryos of N. isatidea were cultured for initiation on Murashige and Skoog medium ( MS) supplemented with N-6- benzylamino- purine ( BAP) and -naphthaleneacetic acid ( NAA). Shoot primordia were visible within 5 - 6 weeks and the shoot primordia later developed into normal shoots 10 - 12 weeks after the culture initiation on calli developed from immature embryos. Shoot tips were also excised from developed plantlets for direct shoot organogenesis and cultured on MS shoot induction medium supplemented with BAP ( 0.5, 1.0 and 2.0 mg/ l), kinetin ( KIN) ( 0.5, 1.0 and 2.0 mg/ l) and thidiazuron ( TDZ) ( 0.05, 0.10 and 0.50 mg/ l). Direct multiple shoots from shoot tips developed in most media tested. High shoot multiplication ( 3.73), high rooting ( 53 %) number of root per shoot ( 3.66) and survival ratio ( 46.6 %) were achieved.Öğe Researches on Heterosis on Yield and Yield Components of Some Poppy Hybrid (Papaver somniferum L.) Lines(ANKARA UNIV, FAC AGR, 2008) Gumuscu, Ahmet; Arslan, NesetThe aim of this study is to improve some poppy materials which both the seed yield and capsule yield are higher and its morphine ratio is higher level than the average ratio of the country. The current sown varieties that on the poppy cultivated areas are selected according to the wishes of the producers. The seven lines were crossed reciprocal that were selected in first year of the study which was carried out two years. The parent lines were taken with their hybrids to yield experiments in the second year. Yield components of hybrid and parent lines were compared and investigated for heterosis and heterobeltiosis between these lines. Heterosis and heterobeltiosis values of some important traits of the material have been given below: for seed yield per plant changed between -13.42% to 37.14% and -22.48% to 36.29%; for capsul yield -33.92% to 45.39% and -40.83% to 34.67%; for seed yield -32.05% to 45.89% and -38.34% to 43.22%; for morphine content -24.21% to 44.62% and -32.44% to 32.68%; for morphine yield -37.63% to 100.39% and -54.68% to 95.21%, respectively.
