    Combination Antioxidant Effect of Alpha-Tocoferol and Erdosteine in Ischemia-Reperfusion Injury in Rat Model

    Introduction Renal ischemia/reperfusion (I/R) which is an important cause of renal dysfunction is inevitable in renal transplantation, surgical revascularization of the renal artery, partial nephrectomy and treatment of suprarenal aortic aneurysms. Aim The purpose of this study was to investigate the efficacy of alpha-tocopherol and erdosteine combination in the reduction in injury induced by ROS in a rat model of renal ischemia-reperfusion. Materials and methods Thirty-six-male Wistar albino rats weighing 200-250 g were utilized for this study. Rats were divided into six groups, and each group was consistent of six rats: (1) sham-operated (control), (2) ischemia group (3) I/R group, (4) I/R/alpha-tocoferol group (5) I/erdosteine group (6). I/R/alpha-tocoferol and erdosteine group. Biochemically tissue MDA, XO and SOD activities, light and electron microscopic findings were evaluated. Results The erdosteine and alpha-tocoferol significantly reversed the effect of protein oxidation and lipid peroxidation induced by I/R shown by the decreased levels of MDA and XO activities. Both MDA and XO levels were found to be lower in group 6 compared to single agent treatment groups, and this was significantly different. All treatment groups showed increased SOD activity, which accounts for their oxidative properties. The mean Paller score of the combination treatment group (group 6) was lower than all groups except the sham group (3.67 +/- 1.2), and this finding was statistically significant (0.05). Our results showed that the antioxidant pretreatment with alpha-tocopherol and erdosteine combination reduced lipid peroxidation of renal cellular membranes in a model of normothermic renal ischemia-reperfusion in rats. Combination of erdosteine and alpha-tocopherol has a synergistic effect of protection against oxidative processes. Long-term use of alpha-tocopherol seems to have a greater effect on the prevention of IR injury. However, further investigations are needed for the clinical applications of our findings.

    Ghrelin, resistin and leptin levels in patients with metabolic syndrome

    Amaç: Bu çalışmada metabolik sendromlu (MS) hastalar ile sağlıklı kontroller arasında açlık ghrelin, leptin ve resistin düzeylerinin karşılaştırılması amaçlanmıştır. Metod: Çalışmaya 21 MS hastası (7 erkek; ortalama yaş, 444) ve 17 sağlıklı kontrol (8 erkek; ortalama yaş 433) alınmıştır. MS tanısı Ulusal Kolesterol Eğitim Programı (NCEP) Erişkin Tedavi Paneli (ATP) III kriterlerine göre tanımlandı. MS karşılama kriterlerinden en az 3’üne sahip olan ve beden kitle indeksi (BKI) 30 kg/m² olan hastalar MS grubuna seçildi. Sağlıklı gönüllüler arasında BKI 30 kg/m² olanlar kontrol grubu olarak seçildi. Plazma ghrelin, leptin ve resistin düzeyleri ELISA yöntemi ile ölçüldü. Bulgular:Ghrelin düzeyleri MS ve kontrol grupları arasında benzerdi. Ghrelin ve Leptin seviyeleri ile BKİ arasında negatif bir ilişki tespit edildi (r-.54, P.01 ve r-.56, p 0.009, sırasıyla). Resistin düzeyleri kontrol grubu ile MS arasında benzer bulundu. Leptin düzeyleri kontrol grubuna göre MS grubunda (3517 ng/ml ve 148 ng/ml, p 0.001) anlamlı derecede daha yüksek bulundu. Leptin düzeyleri ile BKİ arasında pozitif korelasyon vardı (r 0.56; p 0.008). Biz MS grubunda leptin düzeylerinin kontrol grubuna göre daha yüksek olduğunu gösterdik. Ancak, ghrelin ve resistin düzeyleri kontrol grubuna benzerdi. Sonuç: Sonuç olarak biz leptin düzeyleri ile BKİ arasında pozitif bir ilişki olduğunu ve ghrelin seviyeleri ile negatif ilişkili olduğunu gösterdik.

    Relationship of Cellular Oxidant and Antioxidant Status with Disease Activity in Patients with Rheumatoid Arthritis

    In recent years, increasing attention has been given to the role of reactive oxygen metabolites in the pathogenesis of inflammatory diseases such as rheumatoid arthritis (RA). The aim of this study was to elucidate plasma and especially erythrocyte oxidant and antioxidant status in RA patients and to assess the relationship between disease activity scores (evaluated by Disease Activity Score (DAS) 28 as mild, moderate and severe) and antioxidant status. Materials and Methods: Fasting blood samples were obtained from 50 RA patients and 26 control subjects. DAS28 was used to evaluate the activity. Plasma and erythrocyte levels of malondialdehyde (MDA), xanthine oxidase (XO) and superoxide dismutase (SOD) were investigated in both groups.Results: The plasma levels of MDA and XO and erythrocyte levels of MDA and SOD were significantly higher in RA patients than in control subjects. Although increases in plasma MDA levels in mild and moderate and erythrocyte MDA levels in mild activity groups of RA were not significant (p>0.05), plasma MDA levels in severe (p0.001) and erythrocyte MDA levels in moderate and severe activity groups of RA were significantly higher (p<0.001). Whereas plasma SOD levels showed no significant change (p0.241), erythrocyte SOD levels were significantly increased in RA patients (p<0.001). Although increase in erythrocyte SOD activity in mild RA was not significant, it was significant in moderate and severe RA (p0.002 and p<0.001, respectively). Conclusion: These results suggest that increased plasma and erythrocyte MDA levels in RA patients, especially in those with severe activity, indicate an increased oxidative stress due to inflammation. Nonetheless, an increase particularly in erythrocyte SOD activity in moderate and severe RA patients suggests that the cellular antioxidant system might counterbalance the oxidant status in RA patients.

    The Serum Levels of Resistin in Ankylosing Spondylitis Patients: A Pilot Study

    Resistin is a recently described adipokine which is a member of cysteine-rich secretory protein family. Although it has been primarily defined in human adipocytes, it has been identified that its level was higher in mononuclear leukocytes, macrophages, spleen, and bone marrow cells. Because ankylosing spondylitis is an inflammatory disease, it is suspected that upregulation of proinflammatory cytokines is effective in its immunopathogenesis. The aim of our study is to determine the serum resistin levels in patients with AS and to research the relationship with disease activity markers. A total of 30 patients with AS and 30 healthy controls were included in this study. Serum resistin concentrations, erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), Bath AS Disease Activity Index (BASDAI) were evaluated. In results resistin levels in ankylosing spondylitis group were significantly higher than in control group. But, there was no correlation between resistin and ESR, CRP, BASDAI. In conclusion, higher serum resistin levels in patients with AS compared to healthy subjects give clues that resistin could have a role in the pathogenesis of AS.
