Öğe Alterations of Serum Leptin Levels in Patients With Nonalcoholic Liver Cirrhosis(Lippincott Williams & Wilkins, 2006) Kayaçetin, Ertuğrul; Kısakol, Gürcan; Gönen, Sait; Dikbaş, Oğuz; Güngör, Kağan; Kayaçetin, Serra; Topcu, Cemile; Kaya, AhmetIn liver cirrhosis, nutritional status is quite impaired, and the current data has shown conflicting results in serum leptin levels of cirrhotic patients. In this study, we aimed to evaluate serum leptin levels in nonalcoholic liver cirrhosis and determine its relationship with anthropometric parameters and liver function. Thirty-three patients (13 males, 20 females) with nonalcoholic cirrhosis and 16 (9 males, 7 females) healthy control subjects were recruited for this study. Serum leptin values were measured in morning samples of both groups by enzyme-linked immunosorbent assay. The mean levels of leptin were not different in nonalcoholic cirrhotic patients when compared with the control group (P = 0.478). Serum leptin values were positively correlated with prothrombin (international normalized ratio) (P = 0.008, r = 0.64) and total bilirubin (P = 0.047, r = 0.488) and were negatively correlated with high-density lipoprotein (P = 0.025, r = 0.666) and albumin (P = 0.009, r = 0.615). No correlation was found between leptin levels and severity of the liver disease. These findings suggest leptin levels do not depend on the degree of liver damage in patients with nonalcoholic cirrhosis of the liver.Öğe Diabetli hastalarda Na+/K+, ATPaz gen polimorfizminin araştırılması(Selçuk Üniversitesi Sağlık Bilimleri Enstitüsü, 2006) Topcu, Cemile; Gürbilek, MehmetTip 2 diabetik hastalarda ATP1A1 polimorfizminin Na+/K+-ATPaz aktivite değişimleri ile ilişkili olup olmadığını araştırdık. Aynı zamanda C-peptid seviyelerinin ATP1A1 polimorfizmi ve Na+/K+-ATPaz aktivitesi arasındaki ilişkiyi etkileyip etkilemediğini araştırdık. Tip 2 diabetik hastalarda, Na+/K+-ATPaz aktivitesi kontrol grubuna göre daha düşük, C-peptid seviyeleri daha yüksek bulundu. Enzim aktivitesi ve C-peptid seviyeleri arasında korelasyon bulunmadı. Parametrelere ait ortalama ± standart sapma değerleri; C-peptide seviyeleri kontrol grubu için; 1,51 ± 0,44 ve hasta grubu için 2,11 ± 1,19 , (p< 0,002), Na-K-ATPaz aktiviteleri; kontrol grubu için 2,47 ± 1.30 hasta grubu için; 1,13 ± 0,45 (p< 0,000). Tüm diabetik hastalar ve sağlıklı kontrol grubu kesilmemiş allel için homozigot olarak bulundu.Öğe Effect of deferoxamine on Na+K+ATPase activity after cerebral ischemia in rabbits(2006) Gürbilek, Mehmet; Topcu, Cemile; Aköz, Mehmet; Barışkaner, Hülagü; Üstün, Mehmet E.; Köylü, ÖznurObjective: To study the effects of deferoxamine on tissue sodium-potassium adenosine triphosphatase (Na+-K+ ATPase) activity on cerebral ischemia in rabbits. Methods: We cared for the animals in the Pharmacology Department of the Medical School of Selcuk University in 2004. We used 30, New Zealand, 7-day-old male rabbits in the experiment. We anesthetized all the animals with xylazine hydrochloric acid and ketamine. We divided the rabbits equally into 3 groups. In group 1 (n=10) (sham group), we observed baseline levels, and did not apply ischemia. In group 2 (n=10) (untreated group) we produced cerebral ischemia by clamping the bilateral common carotid arteries for 60 minutes, and in group 3 (n=10), we administered deferoxamine (DFO) 50 mg/kg intravenously immediately after opening the clamps. Results: The Na +-K+ATPase activity increased after DFO treatment (p=0.045). Conclusion: We conclude that Na+-K+ATPase activity in cortical brain tissue was higher in DFO-treated rabbits compared with untreated animals after ischemia.Öğe The efficacy of intrathecal administration of a very low dose potirelin after acute spinal cord injury(ELSEVIER SCI LTD, 2008) Cengiz, Sahika Liva; Ustun, Mehmet Erkan; Topcu, Cemile; Ahmet, A. K.Background and object: The objective of this study was to determine the effect of a very tow dose protirelin in cerebrospinal fluid (CSF) glucose, magnesium and lactate levels after spinal cord trauma (SCT) in rabbits. We also aimed to evaluate whether this very tow dose might induce analeptic effect. Material and methods: Twenty rabbits were divided equally into two groups: group I (n = 10) was the control group, suffered from SCT but received only saline after SCT. Group II (n = 10) (treatment group), received a very low dose of 0.05 mg/kg thyrotropin releasing hormone (TRH), analogue protirelin intrathecally after SCT. The basal CSF glucose, magnesium and lactate levels were recorded in both groups. CSF lactate, glucose and magnesium contents were recorded at the same time (an hour before and after) SCT Serum thyroid stimulating hormone (TSH), freetriiodothyronine (73) and freethyroxine (FT4) were measured in all rabbits before and after SCT. Results: Before spinal cord trauma, there were not any significant differences in glucose, lactate and magnesium Levels between group I and II whereas, after spinal cord trauma in group 11, the significant suppression in elevation of Lactate and glucose depletion (p < 0.05) were observed white no significant suppression was observed in magnesium level (p > 0.05) as compared with group I (Table 3). In respect of serum TSH levels, there were not any significant differences between two groups before and after SCT. Conclusions: This study showed that intrathecal TRH has no analeptic effect on serum TSH, FT3 and FT4 levels but can attenuate the increase of lactate levels following spinal cord trauma. No significant decrease in magnesium level and also suppression of glucose decline in group II, may be related to the neuroprotective effects of TRH. (C) 2008 Published by Elsevier Ltd.Öğe The Investigation of Relationship Between Na+/K+ ATPase Enzyme Activity, ATP1A1 Gene Polymorphism and C-Peptide in Type 2 Diabetic Patients with Neuropathy(TURKISH BIOCHEM SOC, 2009) Topcu, Cemile; Gürbilek, Mehmet; Aköz, Mehmet; Çora, TülinObjectives: Na+/K+-ATPase is encoded by various genes, of which the ATP1A1 gene is expressed predominantly in peripheral nerves and in erythrocytes. We investigated whether the ATP1A1 polymorphism is associated with Na+/K+-ATPase activity modifications in healthy subjects and in type 2 diabetic patients with polyneuropathy. We also tested whether C-peptide level could influence the relationship between ATP1A1 polymorphism and Na+/K+-ATPase activity. Methods: Na+/K+-ATPase polymorphism has been determined by using polymerase-chain reactions and restriction fragment length polymorphism methods. The erythrocyte membrane Na+/K+-ATPase activity measurement was performed with modified Kitao-Hattori method. C-peptide measurements were performed using competitive chemiluminesence enzyme immunoassay method. Results: Na+/K+-ATPase activity was significantly lower in diabetic polyneuropathic patients than in healthy subjects. Correlation between C-peptide levels and Na+/K+-ATPase activity was not detected. All diabetic patients with polyneuropathy and healthy controls were homozygous for the unrestricted allele. Conclusion: Oxidative stress which develops in diabetes causes lipid peroxidation in the membrane and consequently Na+/K+-ATPase activity decreases. Low enzymatic activity leads to decrease in nerve conduction velocity. Therefore, we believe that better regulation of diabetes can abrogate complications and prevent the development of neuropathy.Öğe Is Asymmetric Dimethylarginine a Useful Biomarker in Children With Carbon Monoxide Poisoning?(LIPPINCOTT WILLIAMS & WILKINS, 2019) Yazar, Abdullah; Akin, Fatih; Sert, Ahmet; Ture, Esra; Topcu, Cemile; Yorulmaz, Alaaddin; Ercan, FatihObjective Carbon monoxide poisoning (COP) is the leading cause of mortality and morbidity due to poisoning worldwide. Because children are affected more quick and severely from COP, they may require a longer treatment period, even if carboxyhemoglobin (CO-Hb) and/or lactate levels return to normal. Therefore, a new marker that predicts the duration of treatment and the final outcomes of COP is needed. Methods This case control study was conducted on 32 carbon monoxide-poisoned patients younger than 18 years who had been admitted to pediatric emergency department. The control group included age- and sex-matched 30 healthy children. Blood samples were obtained for analysis of arterial blood gases, CO-Hb percent, methemoglobine, lactate, and asymmetric dimethylarginine (ADMA). Results Asymmetric dimethylarginine levels were significantly increased (P < 0.05) in patients with COP on admission and after the treatment when compared with controls (1.36 [0.89-6.94], 1.69 [0.76-7.81], 1.21 [0.73-3.18] nmol/L, respectively). There was no positive correlation between CO-Hb and ADMA levels on admission and at 6 hours (P = 0.903, r = 0.218, P = 0.231, r = 0.022, respectively). Positive correlation was found between lactate and CO-Hb levels on admission (P = 0.018, r = 0.423). Conclusions This study showed that ADMA levels were still high after 6 hours of 100% oxygen therapy in children with COP, even CO-Hb and/or lactate levels return to normal range. On the basis of these results, we consider that ADMA may be a useful biomarker in patient with COP.
