Biberde anter kültürü uygulamalarının in vitro androgenesis üzerine etkileri

Özet
Bu çalışma genotip, besin ortamları ve inkübasyon koşullarının 12 biber genotipinde (Capsicum annuum L.) anter kültüründe haploid bitki oluşumuna etkilerinin belirlenmesi amacıyla yapılmıştır. Öncelikle tomurcuklar +4 oC'de 1 gün ön soğuk uygulamasına tabi tutulmuş, yüzey sterilizasyonu yapıldıktan sonra 3 farklı ortamda (O1, O2 ve O3) kültüre alınmıştır. Sonuçlar uygulamaların anterlerin gelişimi, embriyo ve bitkicik rejenerasyonu üzerine farklı etkilerin olduğunu göstermiştir. Anter gelişim oranında genotipler açısından en düşük değer % 2.47 ile SÜ-31 (Çarliston tipi) genotipinden elde edilirken, en yüksek değere % 12.79 ile Flinta F1 (sivri biber) sahip olmuştur. Ortamlar açısından en yüksek anter gelişim oranları O2 ortamından elde edilmiş, bu ortamda Dolphin çeşidi % 25.0 ile en yüksek değeri vermiştir. Embriyo sayısı açısından Dolphin 8 adet embriyo ve 5 adet bitki ile en fazla embriyo ve bitkinin elde edildiği çeşit olurken Flinta F1 ve SÜ-34 genotiplerinden embriyo ve bitki elde edilememiştir. Kültüre alınan 2868 anterden 195'i gelişim göstermiş ve gelişim oranı % 6.8 olarak gerçekleşmiştir. Toplamda 37 embriyo ve 18 bitki elde edilmiş frekansları ise sırasıyla % 1.29 ve % 0.63 olmuştur. Stomatal gözlemler sonucunda elde edilen bitkilerden 13 tanesinin haploid olduğu belirlenmiş ve bitkiler % 0.05'lik kolhisin ile dihaploid hale getirilmiştir.
This study was conducted to determine the effects of genotype, media composition and incubation condition on haploid plant formation in anther culture in 12 pepper genotypes (Capsicum annuum L.). Firstly, the buds were kept at +4 oC for 1 day in pre-cold treatment, then they were subjected to surface sterilization process and were cultured in 3 different medium compositions (O1, O2 and O3). The results showed that the treatments had different effects on the development of anthers, embryo and plantlet initiation. In terms of genotypes, the lowest value was obtained from SU-31 (Charleston type) genotype with 2.47%, while Flinta F1 (long pepper) genotype had the highest value with 12.79% in anther growth rate. In terms of media, the highest anther growth rates were obtained from O2 medium, and Dolphin variety produced the highest value with 25.0 %. , Dolphin variety was the prominent genotype which has the most embryos and plantlets with 8 embryos and 5 plants, respectively. Besides, Flinta F1 and SU-34 genotypes did not produce embryos and plants. Of the 2868 anthers taken into the culture, 195 anthers developed and the rate of development was 6.8 %. A total of 37 embryos and 18 plants were obtained, with frequencies of 1.29% and 0.63%, respectively. As a result of the stomatal observations, 13 plants were determined as haploid and the haploid plants were transformed to dihaploid at a dose of 0.05% colchicine.
