Antalya'da yetişen Areceae arum dioscorides tohumlarının antioksidan aktivitesi ve toplam fenolik madde tayini

Özet
Bu çalışmada Arum D.'nin antioksidan aktivitesi ve toplam fenolik madde tayini'ni değerlendirmek için toplam fenolik madde tayini, 1,1-difenil-2-pikril-hidrazil (DPPH) radikal giderme aktivitesi, cuprak yöntemi, ß-karoten lineloik asit emülsiyon, indirgeme gücü ve kromatografik analizler GS-MS ve HPLC çalışıldı. Ayrıca ?-tokoferol, troloks, bütilendirilmiş hidroksitoluen (BHT), bütillendirilmiş hidroksianisol (BHA) referans antioksidan bileşikler olarak kullanıldı. Arum D. ekstraktları ile BHA ve BHT standartları şu şekilde DPPH? radikal giderme aktivitesi sergilediler; metanol ekst.>BHT>BHA>aseton ekst.>hekzan ekst. Bu değerler sırasıyla %96, %95, %91, %35, %33 olarak hesaplandı. Arum D. metanol ve aseton 2 mg/mL konsantrasyonunda sırasıyla %78, %52 linoleik asit emülsiyonunun lipit peroksidastonunu inhibe ettiği gözlendi. Referans olan BHT ve BHA sırasıyla %78, %87 linoleik asit emülsiyonunun lipit peroksidastonunu inhibe ettiği gözlendi. Aynı şekilde diğer yöntemlerde benzer özellik göstermiştir. Sonuç olarak bu bitki bir doğal antioksidan kaynağı olarak ümit vericidir.
In this study, total phenolic amount determination technique, free radicals scavenging activity of 1.1-diphenyl-2-picryl-hydarzyl (DPPH), cuprac method, emulsion of ß-carotene lineloic acid, reducing power and chromatographic analyses which are GC-MS and HPLC, methods were worked to evaluate antioxidant activity of Arum D and total phenolic amount. ?-tocopherol, trolox, butylirized hydroxytoulene (BHT), and butylirized hydroxyanisol (BHA) were also used as reference antioxidant compounds. In the free radical scavenging acivity experiments, Arum D extracts and BHA and BHT standards gave results as metanol extracts > BHT standards > BHA standards > acetone extracts > hexane extracts. These values were calculated as 96%, 95%, 91%, 35%, 33% relatively. In the 2 mg/mL concentrations of Arum D methanol and acetone extracts, Arum D methanol and acetone extracts inhibited lipid peroxidation at 78% and 52% relatively in the lineloic acid emulsion experiment. BHT and BHA which were used as reference matters, inhibited lipid peroxidation at 78% and 87% in the same experiment. The similar results were observed after the other methods were applied. As a result, this plant is promising us to be used as a natural antioxidant source.
