Tavşanlarda paklitaksel ve resveratrolun bazı kan parametreleri üzerine etkisi

| dc.description.abstract | Bu çalışmada paklitakselin kan parametreleri üzerine oluşturduğu olumsuz etki ve bunun üzerine resveratrol'ün koruyucu etkisi olup olmadığı, perifer kan ANAE pozitif lenfosit oranı, lökosit formülü, nötrofil çekirdek lob sayısı ve eritrosit çapı üzerine etkileri ile değerlendirildi. Yeni Zelanda ırkı erkek tavşanlar, her grupta 8 adet olacak şekilde 4 gruba ayrıldı. Kontrol grubuna 40 ml serum fizyolojik, paklitaksel grubuna 40 ml serum fizyolojik içerisinde 5mg/kg dozunda paklitaksel, resveratrol grubuna 40 ml serum fizyolojik içerisinde 4 mg/kg dozunda resveratrol, paklitaksel+resveratrol grubuna ise 40ml lik serum fizyolojik içerisinde 5 mg/kg paklitaksel + 4 mg/kg resveratrol uygulandı. Tüm uygulamalar 8 hafta boyunca haftada bir defa intravenöz yolla yapıldı. Paklitaksel nötropeniye sebep olurken total lenfosit ve monosit oranını artırdığı gözlendi. Nötrofil çekirdek loblanma eğrisinde diğer gruplardan farklı olarak sağa kayma gözlenirken ANAE + lenfosit oranında ise azalma gözlendi. Resveratrolün ANAE + lenfosit oranını azalttığı kısmende olsa nötrofil oranını artırırken total lenfosit oranını azalttığı gözlendi. Birlikte ise resveratrolun, paklitakselin ortaya çıkardığı nötropeni ve total lenfosit artışını engellediği ve ANAE + lenfosit oranını artırdığı gözlendi. Sonuç olarak paklitakselin bazı kan parametreleri üzerine olumsuz etkileri olduğu belirlendi. Ayrıca resveratrolun paklitakselin olumsuz etkisine karşı koruyucu etkileri olduğu tespit edildi. | en_US |
| dc.description.abstract | Adverse effects of paclitaxel on blood parameters, consisting the rate of ANAE positive peripheral blood lymphocytes, white cell count, neutrophil core lobe count and erythrocyte diameter, and the protective effects of resveratrol on these parameters are discussed in this study. New Zealand breed male rabbits were divided into 4 groups to be 8 per each group. Drugs were administered to the releated groups as following; 40 ml of normal saline to control group, 5 mg / kg dose of paclitaxel in 40 mL normal saline to paclitaxel group, 4 mg / kg dose of resveratrol in 40 ml normal saline to the resveratrol group, 5 mg / kg paclitaxel + 4 mg / kg in 40 ml normal saline to paclitaxel + resveratrol group. All administrations were done intravenously once a week for 8 weeks. Increase in the total proportion of lymphocytes and monocytes while neutropenia was observed due to paclitaxel. The rightward shift of the lobe curve of the neutrophil nuclei was observed but meanwhile the rate of ANAE positive lymphocytes was declined. Resveratrol was found to reduce the rate of ANAE positive lymphocytes. Although the partly increase in the percentage of neutrophils and the reduction in the total lymphocyte ratio were also observed. In the paclitaxel+resveratrol group it is observed that resveratrol prevents the neutropenia and the increase of lymphocytes due to paclitaxel and supplies the increase of ANAE positive lymphocytes. As a result, it was determined that paclitaxel had a negative impact on some blood parameters. In addition, resveratrol was found to have a protective effect against the harmful effects of paclitaxel. | en_US |
