Lentınula edodes (Berk) pegler 1976'in misel gelişmesine humik maddelerin ve gibberilik asidin etkisinin araştırılması

Özet
Bu çalışmada humik maddelerin ve gibberilik asidin (GA3) Lentinula edodes’in
misel gelişimi üzerine etkisi araştırılmıştır. Besiyeri olarak Malt ekstrakt agar (MEA) ve
patates dekstroz agar (PDA) kullanılmıştır. Ön çalışma olarak besiyerlerine %0, %1,
%3, %5, %7, %9 oranlarında humik madde ve gibberilik asit (GA3) ilavesi yapılmış ve
ön çalışma sonuçlarına göre de denemelerin %0, %0.5, %1, %1.5 oranlarında
yapılmasına karar verilmiştir. Besiyerleri belirtilen oranlarda humik madde ve gibberilik
asit (GA3) eklenerek, otoklavda 121°C’ de 15 dk steril edilmiştir. Daha önceden pastör
fırınında steril edilmiş olan petrilere hazırlamış olduğumuz besiyerleri güvenlik kabini
içinde dökülüp katılaştıktan sonra besiyerlerine misel ekimi yapılmıştır. Ekimi yapılan
petriler etüvde inkübasyona bırakılmıştır. Daha sonra belli aralıklarla miselyum
gelişmeleri alansal olarak çizilmiş ve L. edodes’in misel gelişimi incelenmiştir.
Denemeler 3 tekrarlı olarak yapılmıştır. Deneme sonuçlarına göre yapılan varyans
analizlerinde ortalamalar arasındaki farklar istatistiki olarak önemli bulunmuştur
(P<0.01). %0, %0.5, %1 ve %1.5 oranında humik madde ve gibberilik asit (GA3)
uygulamaları yönünden değerlendirme yapıldığında gelişim miktarı ve katkı maddeleri
oranları arasında yüksek bir korelasyon tespit edilememiş olup, MEA besi ortamında
0.5 ve 1.5’ lik humik madde uygulamasında nispeten yüksek gelişim olduğu
görülmüştür (R² = 0.370).
In this study, the effect of humic substances and gibberellic acid (GA3) on the mycelial growth of Lentinula edodes was investigated. Malt extract agar (MEA) and potato dextrose agar (PDA) were used as growth media. As a preliminary study, 0%, 1%, 3%, 5%, 7% and 9% humic substances and gibberellic acid (GA3) were added to the growth cultures and according to preliminary results, it was decided that the experiments should be performed at 0%, 0.5%, 1% and 2%.The prepared growth cultures was poured into the petri dishes which had previously been sterilized in pasteur oven in the security cabinet and micelles were planted on the solidified growth cultures.Cultivated petroleum was allowed to incubate in the oven.Then, mycelium growth areas were drawn at certain intervals and the micelle development of L. edodes was investigated. The experiments were conducted with 3 replications. According to the analysis of variance using experimental results, the differences between the averages were statistically significant (P <0.01). When considering applications of 0%, 0.5%, 1% and 1.5% humic substances and gibberellic acid (GA3) ratings no significant correlation was found between the amount of growth and additives, and it was observed that there was a relatively high development in the application of 0.5 and 1.5 humic substances in MEA growth culture (R² = 0.370).
