Özet
Bal Kabağı (Cucurbita moschata Duchesne) ve Kestane Kabağı (Cucurbita maxima Duchesne)
önemli sebze türlerimizdendir. Bu türlere ait Batı Anadolu gen kaynakları 2012-2013 yıllarında taranarak
148’i Kestane kabağı, 54’ü Bal kabağı olmak üzere toplam 202 materyal derlenmiştir. Derlenen
genotiplere ait tohumlar 5 adet bitki kendilemede kullanılmak üzere 20 adet olacak şekilde ekilmiştir.
Çalışmada UPOV özellikleri dikkate alınarak, 2013-2015 yılları arasında yaprak, çiçek, meyve, bitki ve
tohumlarda bazı morfolojik ölçüm ve gözlemler yapılmıştır. Mevcut genetik havuz her yıl kendilenerek
S3 kademesine getirilmesinin yanı sıra, belirlenen bazı özellikler açısından tartılı derecelendirmeye tabi
tutulmuştur. Bunun yanı sıra S2 kademsinde selekte edilen genotiplerde moleküler karakterizasyon ve
homojenite testleri yapılmıştır. Çalışmada ilk yıl yapılan tartılı derecelendirme sonucunda toplam 48 adet
Cucurbita maxima Duchesne genotipi ve 29 adet Cucurbita moschata Duchesne genotipi yüksek puan
alarak selekte edilmiştir. Çalışmanın ikinci yılında yapılan tartılı derecelendirme sonucunda 10 adet
Cucurbita maxima Duchesne genotipi ve 4 adet Cucurbita moschata Duchesne genotipi yüksek puan
alarak ümitvar genotipler olarak ortaya çıkmıştır. S1 kademesindeki genotiplerin meyve özelliklerinden
yapılan kümeleme analizleri sonucunda genotipler arasında benzerliklerin olduğu fakat bu genotiplerin
farklı bölgelerden derlendiği belirlenmiştir. Yapılan moleküler analizler sonucunda Bulk1 ve Bulk2
grupları için Simple Matching benzerlik indeksine göre oluşturulmuş olan matrixler programda analiz
edilmiştir. Analiz sonucunda iki matrix arasındaki korelasyonu ifade eden r değeri 0.95623 olarak
hesaplanmış olup, bu değer göz önüne alındığında her iki veri grubu arasındaki korelasyon yüksek olarak
tespit edilmiştir. Elde edilen sonuçlar ışığında, mevcut gen havuzunun belirli bir kademe ilerlemesi
sağlanmış olup, gen havuzunun önemli bir varyasyon göstermesi yeni çeşit adaylarının geliştirilmesi için
önem arz ettiği düşünülmektedir.
Pumpkin (Cucurbita moschata Duchesne) and winter squash (Cucurbita maxima Duchesne) are among our important vegetable species. The Western Anatolian gene sources of these species were scanned in 2012-2013 and a total of 202 materials, 148 of which were winter squash and 54 of which were pumpkin, were collected. Seeds belonging to the compiled genotypes were sown as 20 seeds to be used in 5 plant selfing. In the study, some morphological measurements and observations were made on leaves, flowers, fruits, plants and seeds between 2013 and 2015, taking into account the UPOV characteristics. The genetic pool has been inbred every year and has been graded in terms of certain traits, as well as being brought to the S3 level. In addition, molecular characterization and homogeneity tests were performed on genotypes selected at S2 level. In the study, a total of 48 Cucurbita maxima Duchesne genotypes and 29 Cucurbita moschata Duchesne genotypes were selected with high scores as a result of the weighted grading made in the first year. As a result of the weighted grading in the second year of the study, 10 Cucurbita maxima Duchesne genotypes and 4 Cucurbita moschata Duchesne genotypes were found to be promising genotypes with high scores. As a result of clustering analyzes made from fruit characteristics of genotypes in S1 level, it was determined that there were similarities between genotypes, but these genotypes were compiled from different regions. In the molecular analyzes, the matrices created according to the Simple Matching similarity index for the Bulk1 and Bulk2 groups were analyzed in the program. As a result of the analysis, the r value, which expresses the correlation between the two matrices, was calculated as 0.95623, and considering this value, the correlation between both data groups was determined to be high. In the light of the results obtained, a certain step progress has been achieved in the gene pool, and it is thought that the significant variation of the gene pool is important for the development of new cultivar candidates.
