Sıçanlarda Deneysel Böbrek İskemi-Reperfüzyonunda Naringin’in Aquaporin-1 ve Aquaporin-2 Düzeylerine Etkisi

Özet
Böbrek iskemi reperfüzyonu ciddi bir sağlık problemine yol açabileceğinden dolayı bunun
elektrolit su dengesinde oluşturabileceği olumsuz değişimleri önlemede aquaporinler önemli rollere
sahiptir. Bu çalışmanın amacı deneysel böbrek-iskemi reperfüzyonunda naringin tedavisinin böbrek
korteks ve medulla dokusundaki AQP 1 ve AQP 2 düzeylerine olan etkisini belirlemekti. Araştırma
Selçuk Üniversitesi Deney Hayvanları Araştırma ve Uygulama merkezi ile Necmettin Erbakan
Üniversitesi Deney Hayvanları Araştırma ve Uygulama Merkezinden temin edilen 8-12 haftalık 40 adet
erkek Wistar tipi sıçan üzerinde gerçekleştirildi. Deney grupları şu şekilde oluşturuldu; 1) Kontrol
grubu, 2) Sham kontrol + Vehicle grubu, 3) Böbrek iskemi-reperfüzyonu (Renal İ+R) + Vehicle grubu,
4) Renal İ+R + Naringin (50 mg/kg/gün) grubu (3 gün uygulama), 5) Renal İ+R + Naringin (100
mg/kg/gün) grubu (3 gün uygulama) grubu.
İskemi reperfüzyon gruplarında sol böbreklerde nefrektomi sağ böbreklerde ise 45 dakika
iskemi ve takiben 72 saat reperfüzyon gerçekleştirildi. Naringin reperfüzyonun hemen başında, 24 saat
sonra ve 48 saat sonra intraperitoneal yolla deney hayvanlarına verildi. Deneylerin bitiminde genel
anestezi altındaki hayvanlarda öncelikle kalpten kan alındıktan sonra servikal dislokasyonla hayvanlar
öldürülerek böbrek doku örnekleri alındı. İdrar osmolarite, kan osmolarite, üre ve kreatinin yönünden
böbrek dokuları da AQP 1 ve AQP 2 yönünden Elisa ve PCR yöntemleri ile değerlendirildi.
Böbrek dokularında iskemi plazma ve idrar osmolaritesi ile AQP 1 ve 2 seviyeleri önemli
şekilde düşerken plazma üre ve kreatinin seviyeleri ise önemli oranda artış gösterdi. Ancak naringin
takviyesi meydana gelen bozulmaları belirli oranda düzeltti.
Çalışmanın sonuçları naringinin sıçanlarda tek taraflı nefrektomi ve İ/R ile oluşan böbrek
fonksiyonundaki bozulmalar üzerinde koruyucu etkilere sahip olabileceğini göstermektedir.
Since renal ischemia reperfusion can lead to a serious health problem, aquaporins have important roles in preventing negative changes in electrolyte-water balance. The aim of this study was to determine the effect of naringin treatment on AQP1 and AQP2 levels in renal cortex and medulla tissue in experimental kidney-ischemia reperfusion. The study was carried out on 40 male Wistar type rats, 8-12 weeks old, obtained from Selçuk University Experimental Animals Research and Application Center and Necmettin Erbakan University Experimental Animals Research and Application Center. Experimental groups were formed as follows; 1) Control group, 2) Sham control + Vehicle group, 3) Renal ischemia-reperfusion (Renal İ+R) + Vehicle group, 4) Renal İ+R + Naringin (50 mg/kg/day) group (3 days of administration ), 5) Renal I+R + Naringin (100 mg/kg/day) group (3 days application) group. Nephrectomy in the left kidneys and 45 minutes ischemia in the right kidneys and followed by 72 hours of reperfusion. Naringin was given to the experimental animals by intraperitoneal route at the beginning of the reperfusion, after 24 hours and 48 hours. At the end of the experiments, first of all, blood was taken from the heart in animals under general anesthesia, and then the animals were killed by cervical dislocation and kidney tissue samples were taken. Blood was evaluated in terms of osmolarity, urea and creatinine, urine osmolarity and kidney tissues were evaluated in terms of AQP1 and 2 by Elisa and PCR methods. In kidney tissues, ischemia led to reduced to plasma and urinary osmolality and AQP 1 and 2 levels, while plasma urea and creatinine levels increased significantly. However, naringin supplementation corrected the deterioration to a certain extent. The results of the study show that naringin may have protective effects on the deterioration of renal function caused by unilateral nephrectomy and I/R in rats.
