Özet
Bu araştırmada somatik hücre sayımı sonuçlarına göre mastitis olgusu belirlenmiş hayvanlann sütlerinde nitrik oksit düzeyi ile somatik hücre sayısı arasındaki ilişki incelenmiştir. Araştırmada 4-5 yaşlı laktasyon ortasında bulunan, 64 İsviçre Esmeri süt ineğinin her bir meme lobundan alınmış 256 süt ömeği kullanılmıştır. Kalitatif hücre sayım yöntemi kullanılarak elde edilen somatik hücre sayımı (SHS) sonuçlanna göre gruplar oluşturulmuştur. Somatik hücre sayısı misinde 0-200000 adet olan 93 örnek kontrol grubu, SHS mi'sinde 200000 400000 adet olan 46 ömek mastitis şüpheli grubu, 400000 1200000 adet olan 49 ömek 1. grubu, 1200000 2200000 adet olan 33 ömek 2. grubu, 2200000'den fazla olan 35 örnek ise 3. grubu oluşturmuştur. Süt ömeklerinde Griess reaksiyonu temeline dayanan spektrofotometrik yöntem kullanılarak nitnik oksit ve nitrit düzeyleri belirlenmiştir. Nitrik oksit düzeyi sağlıklı grupta ortalama 21,2712,86 ?mol/L olarak tespit edilirken mastitis şüpheli grupta 22,55+3,09 ?mol/L. 1, 2 ve 3 grupta sırasıyla 27,42+2,54 ?mol/L, 30,75+2,62 µmol/L 37,42:6.27 umov, nitrit düzeyleri ise sağlıklı sütlerde 0.5210.35 umol/L olarak belirlenirken mastitis şüpheli sütlerde 0,52+0,43 µmol/L. mastitisin derecesi arttıkça (sırasıyla 1.2 ve 3.grup) 0.99±0,54 ?mol/L, 1,1310,27 µmol/L, 1,34±0,46 umol/L olarak, belinenmiştir. Sağlıklı ve mastitisli sütler arasında nitrik oksit miktarı bakımından bir farklılık bulunmazken, mastitisin derecesi le birlikte anlamlı düzeyde arttığı ve bu üç grubun nitrik oksit düzeylerinin sağlıklı ve mastitis şüpheli sütlerden yüksek ol duğu belirlenmiştır (p<0.05). Nitrit düzeylen ise kan nitrit düzeyleri bakımından sağlıklı ve şüpheli grup arasında anlamlı bir lark bulunmazken bahsi geçen bu gruplar ile mastitisli sütler arasında fark bulunmuştur (p<0.05). Elde edilen bulgular sonucunda mastitisli inek sütlerinde nitrik oksit ile parçalanma ürünlerinden nitrit düzeyinde bir artış ve süt somatik hücre sayısı ile süt nitrik oksit miktarı arasında anlamlı pozitif bir ilişkinin olduğu bulunmuştur.
In this study the relationship between the level of nitric oxide and somatic cell count in the cow milk in the milks of animals with mastitis determined according to the results of somatic cell count. In the study. 256 milk samples received from each breast lobe of Brown Swiss 64 milk cows of 4-5 age and in the middle of their lactation period. During the survey qualitative cell counting method has been used and according to results of somatic cell count (SCC) some groups have been established. 93 samples with 0-200.000 have established the control group, 46 samples with 200.000400.000 have established the trace group, 49 samples with 400.000-1.200.000 have established the 1st group, 33 samples with: 1.200.000- 2.200.000 have established the 2 nd group and 35 samples with more than 2.200.000 somatic cell count in 1 ml have established the 3 th group. In the milk samples received from these groups nitric oxide and nitrite levels of milk have been determined through the spectrophotometric method based on Griess reaction. The average levels of nitnc oxide were established in healthy group: 212712.86 µmo L in trace group 22.55-3,09 umol/L, the degree of mastitis was getting increase they were established as (respectively 1 St, 2 nd and 3rd group) 27,42+2,54 µmol/L 30,75+2.62 µmol/ Land 37,4216.27 µmol/L. While the average nitrite levels were dedected in healthy milk 0,52 0,35 umol/L, in trace group 0.5210.43 umoVL the degree of mastitis was getting increase they were respectively established as (1st, 2 nd and 3 rd group) 0.99±0.54 umol/L 1.13:0,27 umplt and 1.32-0,46 movL Regarding the nitrite levels in blood there was not any difference between control and trace groups. On the other hand there have been some differences observed between these and remaining 1 st, 2 nd and 3 g groups (P<0.05). There was not any difference between the control and trace group with respect to the amount of nitric oxide, it has been spon that other groups have showed significant increases between each other and with respect to control and trace groups (P<0,05).
