Yüksek yağlı diyetle beslenen ratlarda liraglutit ve metformin tedavisinin metabolik parametreler ile ilişkilerinin değerlendirilmesi

Özet
Giriş: Bu çalışmanın amacı yüksek yağlı diyetle beslenen ratlarda liraglutit ve metformin tedavisinin metabolik parametreler ile ilişkilerinin değerlendirilmesidir. Gereç ve Yöntem: Bu çalışma; 18.01.2023-28.03.2023 tarihleri arasında gerçekleştirildi. Çalışmada ortalama 456g (419-490g) ağırlığındaki 16 haftalık sağlıklı yetişkin Wistar türü albino erkek sıçanlar kullanıldı. Toplam 36 adet Wistar türü erkek sıçan 5 gruba ayrıldı:(1) kontrol grubu (KG), (2) yüksek yağlı diyet grubu (YYD), (3) düşük doz liraglutit grubu (YYD+DDL), (4) yüksek doz liraglutit grubu (YYD+YDL), (5) metformin grubu (YYD+M). Kontrol grubu dışındaki gruplar 10 hafta boyunca yüksek yağlı diyet ile beslenirken deneyin son 4 haftasında 3. gruba düşük doz liraglutit (1,2mg/kg) subkutan yoldan, 4. gruba yüksek doz liraglutit (2,4 mg/kg) subkutan yoldan ve 5. gruba metformin oral yoldan uygulandı. Çalışmanın sonunda (10.haftada) ratların vücut ağırlıkları tartıldı, anestezi altında kan örnekleri alınarak biyokimyasal analiz yapıldı. Hayvanlar sakrifiye edildikten sonra karaciğer diseke edilerek ağırlığı hassas terazi ile tartıldı. Bulgular: Ratlara uygulanan 10 haftalık yüksek yağlı diyetin ardından yüksek yağlı diyet grubunda vücut ağırlığında anlamlı artış görüldü (p<0,05). Vücut ağırlıkları (VA), trigliserit düzeyleri, karaciğer ağırlıkları ve TyG indeks değerleri düşük doz ve yüksek doz liraglutit grubunda, yüksek yağlı diyet grubu ve metformin grubuna göre anlamlı olarak daha düşüktü (p<0,05). Vücut kitle indeksleri (VKİ) düşük doz ve yüksek doz liraglutit grubunda yüksek yağlı diyet grubuna göre anlamlı düzeyde daha düşüktü bununla berber yüksek doz liraglutit grubunda metformin grubuna göre anlamlı düzeyde daha düşüktü(p<0,05). LDL kolesterol düzeyleri yüksek doz liraglutit grubu ve metformin grubunda yüksek yağlı diyet grubuna göre istatistiksel olarak anlamlı derecede düşüktü (p<0,05). Açlık plazma glukozu düşük doz ve yüksek doz liraglutit grubunda metformin grubuna göre istatistiksel olarak anlamlı ölçüde düşüktü (p<0,05). Hs-CRP'nin kontrol grubu, yüksek yağlı diyet grubu, düşük doz liraglutit ve yüksek doz liraglutit grubu metformin grubu ile karşılaştırıldığında metformin grubu diğer tüm gruplara göre anlamlı düzeyde yüksek olarak gözlenmiştir(p<0,05). Sonuç: Yüksek yağlı diyet vücut ağırlığında, lipit düzeylerinde, açlık kan şekeri düzeylerinde ve karaciğer ağırlığında artışa yok açmaktadır. Diyabet tedavisinde kullanılan liraglutit ve metformin lipit düşürücü etkisiyle dislipidemi tedavisine alternatif olabilir.

Materials and Methods: This study; It was held between 18.01.2023 and 28.03.2023. In the study, 16-week-old healthy adult Wistar albino male rats weighing an average of 456g (419-490g) were used. A total of 36 Wistar male rats were divided into 5 groups: (1) control group (KG), (2) high-fat diet group (YYD), (3) low-dose liraglutide group (YYD+DDL), (4) high-dose liraglutide group (YYD+YDL), (5) metformin group (YYD+M). While the groups other than the control group were fed a high-fat diet for 10 weeks, in the last 4 weeks of the experiment, low-dose liraglutide (1.2mg/kg) was administered subcutaneously to the 3rd group, high-dose liraglutide (2.4 mg/kg) was administered subcutaneously to the 4th group, and 5 Group 1 was administered metformin orally. At the end of the study (at the 10th week), the body weights of the rats were weighed, blood samples were taken under anesthesia and biochemical analysis was performed. After the animals were sacrificed, the liver was dissected and weighed with a precision balance. Results: After 10 weeks of high-fat diet applied to rats, a significant increase in body weight was observed in the high-fat diet group (p<0.05). Body weights (VA), triglyceride levels, liver weights and TyG index values were significantly lower in the low-dose and high-dose liraglutide group than in the high-fat diet group and metformin group (p<0.05). Body mass indexes (BMI) were significantly lower in the low-dose and high-dose liraglutide group than in the high-fat diet group, however, they were significantly lower in the high-dose liraglutide group than in the metformin group (p<0.05). LDL cholesterol levels were statistically significantly lower in the high-dose liraglutide group and metformin group compared to the high-fat diet group (p<0.05). Fasting plasma glucose was statistically significantly lower in the low-dose and high-dose liraglutide group compared to the metformin group (p<0.05). When the control group, high-fat diet group, low-dose liraglutide and high-dose liraglutide groups of Hs-CRP were compared with the metformin group, the metformin group was observed to be significantly higher than all other groups (p<0.05). Conclusion: High-fat diet leads to no increase in body weight, lipid levels, fasting blood sugar levels and liver weight. Liraglutide and metformin therapy used in the treatment of diabetes may be an alternative to treatments with their lipid-lowering effectiveness.
