Mast® Antimikrobik Rezistotiplendirme Metodunun Broylerlerden Izole Edilen Arcobacter Butzlerı Suşlarına Uygulanması

| dc.description.abstract | In this study, the applicability of Mast Resistotyping scheme, which was originally developed for the subtyping of Campylobacter species, to Arcobacter butzleri strains isolated from broiler chickens was tested. The resistotyping scheme includes six antimicrobial agents comprising tetracycline (18µg); nalidixic acid (30µg); metronidazole (5µg): 2,3,5, triphenyltetrazolium chloride (1000µg): 5-fluorouracil (80pg) and sodium arsenite (30µg), which are impregnated to paper discs, and the theory of the system is based on determination of the resistance of the bacterial strains to those antibiotics. All strains tested were found sensitive to tetracycline and 2,3,5, triphenyttetrazolium chloride. Sixty-five percent, 15 %, 12% and 9 % of the A. butzleri strains tested gave 66, 26, 06 and 46 code numbers, respectively, and 4 different resistotypes were obtained. Mast Campylobacter Resistotyping scheme was not found very suitable for the sub-typing of A. butzleri strains in this form, since there were few sub-types obtained with this system and the most strains gave the same code numbers (e.g. 22 of the 34 strains gave 66 code numbers). In addition, it is suggested that resistotyping scheme has to be made suitable for the sub-typing of Arcobacter species either by modifying the antimicrobial agents incorporated or by re-adjusting their concentrations. | en_US |
| dc.description.abstract | Bu çalışmada, Campylobacter türlerinin alt tiplendirilmesi amacıyla geliştirilmiş olan Mast rezistotiplendirme metodunun, ticari broyler karkaslarından izole edilen 34 adet Arcobacter butzleri susuna uygulanabilirliği test edildi. Bu rezistotiplendirme metodu, kağıt disklere emdirilmiş: tetracycline (18µg); nalidixic acid (30ug): metronidazole (5ug); 2,3,5, triphenyltetrazolium chloride (1000µg): 5-fluorouracil (80ug) ile sodium arsenite (30µg) olmak üzere 6 adet antimikrobiyal maddeden oluşan ve bakteri suşlarının bu maddelere karşı direncinin saptanması teorisiyle geliştirilen bir alt tiplendirme metodudur. Test edilen suşların tamamı tetracycline ve 2,3,5, triphenyltetrazolium chlorid'e duyarlı bulundu. Mast rezistotiplendirme metodu ile A. butzleri suşlarının % 65, % 15, % 12 ve % 9'u sırasıyla 66, 26, 06 ve 46 kod numaralarını verdi ve 4 ayn rezistotip elde edildi. Elde edilen alt tip sayısının azlığından ve suşların büyük çoğunluğunun (ömeğin 34 suşun 22'si 66 kodunu verdi) aynı kod numarasını vermesinden dolayı, Mast Campylobacter rezistotiplendirme metodunun bu haliyle A. butzleri suşlarının alt tiplendirilmesi amacıyla yeterli olmadığı, kullanılan antimikrobiyal ajanların ya tamamen değiştirilmesi ya da konsantrasyonlarının yeniden ayarlanması gerektiği sonucuna varıldı. | en_US |
