Özet
Amaç: Çalışmamızın amacı streptozotosinle oluşturulmuş diyabette D vitaminin koruyucu ve tedavi edici etkilerinin gösterilmesidir. Gereç ve Yöntem: Çalışma 50 adet Wistar cinsi dişi rat üzerinde gerçekleştirildi. Ratlar 5 gruba ayrıldı. 1. Grup (Kontrol G), 2. Grup (Diyabet G), 3. Grup (Diyabet+D vitamini Tedavi G) , 4. Grup (Diyabet+D vitamini Önleyici G) ve 5.Grup (D vitamini Kontrol G). Diyabet gruplarına 50mg/kg dozunda Streptozotosin intraperitonal yoldan verildi ve 5.günde 300mg/dl üzeri glukoz seviyeleri ile diyabet tanısı kondu. Grup 4'e D vitamini 12.5 µgr/kg dozunda diyabet tanısı konmadan 14 gün önce başlayıp, sonrasında 14 gün daha devam ederek toplamda 28 gün, grup 3'e ise diyabet tanısı konduktan sonra 14 gün boyunca oral gavaj yoluyla verildi. Çalışmanın sonunda ratlardan ketamin anestezisi altında kardiyak ponksiyonla kan örnekleri alındı. Alınan kanların serum örneklerinde IP-10 düzeyleri ELİZA tekniği ile; TAS, TOS, Paraoksonaz ve glukoz spektrofotometrik yöntemlerle ölçüldü. Bulgular: Diyabet+D vitamini Önleyici grubun (grup 4), glukoz seviyeleri, diyabet grubuna (grup 2) ve Diyabet+D vitamini Tedavi grubuna (grup 3) göre anlamlı derecede düşük bulundu. TAS, TOS, Paraoksonaz aktiviteleri açısından gruplar arası anlamlı fark bulunamadı. IP-10 seviyesi diyabet (grup 2) ve Diyabet+D vitamini Tedavi (grup 3) grubunda kontrol gruplarına göre anlamlı yüksek bulunurken; Diyabet+D vitamini Önleyici (grup 4) grubun IP-10 seviyesi kontrol gruplarına göre istatistikî açıdan farklı değildi. Sonuç: D vitamininin diyabet üzerine koruyucu ve tedavi edici etkisini incelemeye çalıştığımız bu çalışmamızın sonucunda; D vitamininin erken dönemde başlandığında çok daha etkin olduğu ve bunu pankreas adacık hücrelerinden sitokin salınımını azaltarak gerçekleştirdiği düşüncesindeyiz.
Aim: The aim of our study was to investigate preventive and therapeutic effects of vitamin D on streptozotocin-induced diabetes. Materials and Methods: This study was performed on 50 Wistar female rats. The rats were separated into five groups. The groups were formed as follows: Control group (group 1), diabetes group (group 2), diabetes+ vitamin D therapeutic effect group (group 3), diabetes + vitamin D preventive effect group (group 4) and vitamin D control group (group 5). Streptozotocin was intraperitoneally administered at a dose of 50mg/body weight. Blood glucose was measured 5 days after the injection of streptozotocin. The diabetic state was con ? rmed when the glucose concentration exceeded 300mg/dL. Group 4 rats received 12,5µg//w/day vitamin D3 for 14 days before streptozotocin injection and for 14 days after streptozotocin injection via oral gavage. Group 3 received 12,5 µg //w/day vitamin D3 for 14 days after streptozotocin injection via oral gavage. After that period, rats were killed under ketamine anesthesia and blood samples were obtained via cardiac punction. IP-10 was determined with ELISA technique, TAS, TOS, IP-10 and glucose levels were determined with spectrophotometric methods. Findings: Serum glucose level was significantly lower in group 4 than group 2 and group 3. There was no significant difference between serum TAS, TOS, Paraoxonase levels of the groups. Serum Ip-10 levels were higher in group 2 and group 3 than in the control groups. But, there was no significant difference between serum IP-10 levels of group 4 and control groups.Results: According to our study that we tried to investigate preventive and therapeutic effects of vitamin D on streptozotocin-induced diabetes we conclude that early treatment is more effective and this beneficial effect of 1,25-(OH)2D3 is associated with decreased cytokine secretion by the pancreatic islets.
