Özet
Bu çalışma 2020-2021 yıllarında Üniversite Sanayi iş birliği kapsamında yürütülmüştür.
Bu bağlamda Türkiye’nin ilk tohumculuk şirketlerinden bir olan Beta Ziraat Tic. A.Ş. firması
ile işbirliği yapılmıştır. Firmanın elinde var olan yaklaşık 4000 yazlık kabak genotipinin tohum
yenileme ve kademe ilerlemesi çalışmaları firma tarafından daha öncesinde yapılmış olup bu ön
çalışma ile 344 adet ümit var genotip bulunarak bu tez çalışmasının genetik materyali
oluşturmuştur. Çalışmada yapılmış olan ölçüm ve gözlemler UPOV parametrelerinden
yararlanarak belirlenmiştir. Yüksek lisans tez çalışmasında çalışma iki generasyon sera
koşullarında işbirliği yapılan firma alt yapısı kullanılarak gerçekleştirilmiştir, İki generasyon
kademe ilerlemesi yapılmış agro-morfolojik parametreler saptanmış ve moleküler markörler
yardımıyla hastalık testlemesi yapılmıştır. Yapılan morfolojik ve moleküler karakterizasyon
sonucunda 113 adet S5 kademesinde hatlar elde edilmiştir. Elde edilen S5 kademesinde ki
hatlarda yapılan hastalık testlemesinde toplam 53 adet ZYMV’ye dayanıklı genotip olduğu
saptanmış bunların 43 adeti homozigot dominant dayanıklı (RR) ve 10 adeti heterozigot
dayanıklı (Rr) şeklindedir. Kalan 55 adet genotip homozigot çekinik dayanıksızdır (rr). Yapılan
tüm bu çalışmalar neticesinde 50 adet hastalığa dayanımlı yazlık kabak F1 ıslahı için yarı yol
materyali olarak kullanılabilecek hatlar elde edilmiştir. Ayrıca genotiplerde kümeleme analizleri
yapılarak hem amaca uygun yazlık kabak ebeveyn hatlar, hem de heterotik gruplar
belirlenmiştir.
This study was carried out in 2020-2021 within the scope of University-Industry cooperation. In this context, Beta Ziraat Tic., one of the first seed companies in Turkey. Inc. collaborated with the firm. Seed renewal and stage progression studies of approximately 4000 summer squash genotypes available to the company were carried out, and 344 hopeful genotypes were found with this preliminary study and formed the genetic material of this thesis. Measurements and observations made in the study were determined using UPOV parameters. The master’s thesis study was carried out in greenhouse conditions in two generations using the infrastructure of the collaborating company, agro-morphological parameters were determined and disease tests were carried out with the help of molecular markers. As a result of the morphological and molecular characterization, 113 S5 stage lines were obtained. In the disease testing of the obtained S5 stage lines, a total of 53 ZYM resistant genotypes were found, 43 of which were homozygous dominant resistant (RR) and 10 were heterozygous resistant (Rr).The remaining 55 genotypes were homozygous recessive resistant (rr). As a result of all these studies, 50 lines that can be used as half-way material for disease resistant summer squash F1 breeding were obtained. These measurements and observations are given in detail in the method section. In addition, cluster analyses were performed on the genotypes and both suitable summer squash parental lines and heterotic groups were determined.
