| dc.description.abstract | Gastrik kanserler yüksek ölüm oranı ile dünyada en yaygın malignensilerden biridir. Etiyolojisi kesin olarak bilinmemesine rağmen gastrik karsinogenezden birçok çevresel ve genetik faktörler sorumlu tutulmaktadır. Genetik faktörlerin incelendiği birçok çalışmada tümör baskılayıcı genlerdeki değişikliklerle kanser gelişimi arasında ilişki bulunduğu belirtilmektedir. Mevcut çalışmada tümör baskılayıcı genlerden olan PTEN geni (ekzon 2, ekzon 3 ve ekzon 6) ile çalışma grubumuzda bulunan gastrik kanserli olgular arasında ilişki bulunup bulunmadığı PZR/TİUP metodu ve DNA dizi analizi kullanılarak incelendi. İnceleme gastrik adenokarsinom tanısı almış 47 hastanın parafin bloklardaki tümörlü dokuları, kontrol grubu olarak aynı hastaların normal dokuları, ayrıca aynı gruptaki 10 hastanın periferik venöz kan örnekleri kullanılarak yapıldı. Çalışma sonucunda PTEN geni intron 1'de DNA dizi analizi sonucunda hastalara ait tümör ve normal dokuların tamamında heterozigot formda TNP (Tek Nükleotid Polimorfizmi) bulunmuştur. TNP geri yönlü analizde intronik bölgede bulunan 8402202 bç pozisyonunda, 401. Nükleotide ve C/T genotipinde bulunmaktadır. Mevcut bölge önceden tanımlanmış olup referans numarası rs1903858'dir. Çalışmamızda kullanılan diğer ekzon 3 ve ekzon 6'da ise herhangi bir değişiklik tespit edilmemiştir. Sonuç olarak PTEN geni rs1903858 varyantı gibi ekzon, intron ve diğer kodlanmayan bölgelerdeki TNP'ler GK'lerin gelişminde etkili olabilir. | en_US |
| dc.description.abstract | Gastric cancer (GC) is one of the most frequent malignancies in the world with high mortality rates. Although its etiology is not clearly known, many genetics and environmental factors can be hold responsible for gastric carcinogenesis. Many previous researches support that there is a relation between mutations in tumor suppressor genes and cancer progress. The present research has been examined whether there isany relationship between the gastric cancer cases in our working group and PTEN gene (exon 2, exon 3 and exon 6) from tumor suppressor genes family by using PCR/SSCP method(s) and DNA sequence analysis. DNA isolation was performed through using paraffin-embedded tumor tissues of 47 patients diagnosed as gastric carcinoma and paraffin-embedded normal tissues of the same patients as control group, also peripheral venous blood samples of 10 patients from the same patient group. As a result, at the end of the sequence analysis, heterozygote form of SNP was found in PTEN gene intron 1 of the patients? tumor and normal tissues. The SNP (Single Nucleotide Polymorphism) was found in the position of 8402202 base pair, 401st nucleotide and in C/T genotype. This region was defined before, and reference number is rs 1903858. There was no change determined in the regions of exon 3 and exon 6 used in this study. In conclusion, like the gene of PTEN- rs 1903858, Exon, Intron and SNPs which are in other non codable regions can be effective in progress of GCs. | en_US |
