Differential identification of cattle Sarcocystis spp. by random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR)

dc.description.abstractMorphological identification of Sarcocystis spp. and the diagnosis of their developmental stages within the host is very difficult. However, recent developments in molecular biology have produced a number of techniques to overcome these difficulties. In this study, the RAPD-PCR (random amplified polymorphic DNA-PCR) technique was used to investigate the molecular biology of Sarcocystis spp. in cattle. In order to isolate the parasite, phenol extraction/ethanol precipitation method was employed to obtain parasitic DNA. After optimisation of the PCR conditions, five primers were selected to amplify the DNA of each Sarcocystis spp. As a result, various Sarcocystis spp. were detected and differentiated using this RAPD-PCR assay in cattle.en_US
