Abstract
Konya bölgesinde, yumurtacı tavuk çiftliklerinde M. gallisepticum ve M. synoviae varlığı bakteriyolojik olarak izolasyon ve identifikasyon, serolojik olarak Enzyme Linked Immunosorbent Assay (ELISA) ve Lam Aglütinasyon Testi (LAT) ve Polimeraz Zincir Reaksiyonu (PZR) ile araştırıldı. Daha önce solunum sistemi problemi geçirmiş ve Konya Veteriner Kontrol Araştırma Enstitü'sünde serolojik olarak CRD enfeksiyonu teşhisi konmuş farklı yaşlarda 15 kümesten 73 tavuğa ait toplam 300 kan örneği toplandı. Ayrıca 73 tavuğun trakealarından, hava keselerinde, akciğerlerinde ve eklem sıvılarından toplam 365 sıvap alındı. Toplam 15 kümesten alınan 300 serum örneğinin 127'si (%43) MG yönünden LAT ile pozitif, 173'ü (%57) negatif, 165'i (%55) MS yönünden LAT ile pozitif, 235'i (%45) negatif olarak değerlendirildi. Aynı 300 örneğin 243'ü (%80) MG yönünden ELISA ile pozitif, 57'si (%20) negatif olarak, 265'i (%88) MS yönünden ELISA ile pozitif, 35'i (%12) negatif olarak değerlendirildi. Toplam 365 sıvap numunesinin sadece 3'ünde etken izole edilerek yapılan identifikasyon çalışmaları ile etkenin MG olduğu tespit edilmiştir. PZR ile ticari yumurtacı kümeslere ait toplam 365 sıvap örneğinin 9'u (%2.46) MG yönünden pozitif, 3'ü (%0.82) MS yönünden pozitif olarak değerlendildi. Sonuç olarak; yumurtacı tavuklarda MG ve MS teşhisinde hem multipleks hem de tekli PZR'ın izolasyondan üstün olduğu ve pratikte kullanılabilecek bir test olduğu ve PZR için alınacak numunelerin mümkün olduğu kadar erken ve nekropsi yapılan hayvanların trakelarından mukozalar kazınarak alınmasını daha iyi sonuç verdiği tespit edilmiştir.
The occurence of Mycoplasma gallisepticum and Mycoplasma synoviae in commercial layer populations in Konya region was investigated by using bacteriological methods, serological methods and Polymerase Chain Reaction (PCR). A total number of 300 blood samples were collected from 15 flocks 73 hens with different ages which has a history of respiratoric problems confirmed by serological test for CRD in Konya Veterinary Control and Research Institute. 365 swap samples from trachea, air sacs, lungs and synoviae were collected from 73 hens. Out of 300 samples from a total of 15 flocks, 127 (%43) samples were found positive for MG and 173 (%57) samples observed negative by LAT, 165 (%55) samples were found positive for MS and 235 (%45) samples observed negative by LAT as well. Out of the same 300 samples, 243 (%80) samples were found positive for MG and 57 (%20) samples observed negative by ELISA, 265 (%88) samples were found positive for MS and 35 (%12) samples observed negative by ELISA. Out of 365 swap samples from 73 hens, 9 (%2.47) samples were found positive for MG and 3 (%0.82) were found positive for MS by PCR. As a conclusion; both the single and the multiplex PCR was found superior than the isolation and identification methods and can be applied for the detection of MG and MS in laying hens and to take the swap samples by scraping the trachea mucosa after the necropsy as soon as the clinical signs appear is recommended as well.
