Ankara Tekelerinde Sperma Sulandırıcısına Farklı Dozlarda Eklenen Rock İnhibitörü, Antifreeze Protein III ve Bor’un Sperma Dondurulabilirliği Üzerine Etkileri

Özet
Sunulan tez çalışmasında, Ankara tekesi spermasının dondurma-çözdürme sonrası
spermatolojik parametreleri üzerine Rock inhibitörü Y-27632, Antifreeze Protein III ve Bor’un iki
farklı dozda etkileri araştırılmıştır. Ejakülatlar yetiştirme mevsiminde tekelerden elektroejakülatör
kullanılarak alındı. Alınan ejakülatlardan uygun özellik gösterenler birleştirilerek spermanın
sulandırılması ve dondurulması işleminde kullanıldı. Sulandırıcı grupları, kontrol ve kontrol
sulandırıcısına iki farklı dozda üç farklı katkı maddesinin (Rock inhibitörü Y-27632 5 ve 20 µM;
Antifreeze Protein III 1 ve 4 µg/ml; Bor 0.25 ve 1 mM) ilave edilmesiyle oluşturuldu. 35-37 °C’ta
farklı sulandırıcılarla sulandırılan spermalar payetlere çekildi. Ekilibrasyonu izleyen süreçte sıvı azot
buharında donduruldu ve sıvı azotta saklamaya alındı.
Rock inhibitörünün 5 ve 20 µM (% 71.82±2.16 ve 74.04±1.59) ve Bor’un 0.25 ve 1 mM
dozlarda (% 76.36±1.80 ve 72.08±1.99), kontrol grubuna göre (% 66.15±1.51) çözüm sonu motilitede
iyileştirmeler yaptığı görüldü (P<0.01). Dondurma-çözdürme sonu akrozomal bütünlük ve
mitokondriyel aktivitenin değerlendirilmesine bakıldığında; sulandırıcıya katılan Antifreeze Protein
III ve Bor’un tüm dozlarda koruma sağlamasına rağmen, bu etkinliğin istatistiksel olarak önemli
olmadığı görüldü (P>0.05). Çözdürme sonu DNA hasarında Antifreeze Protein III 1 µg/ml (%
1.23±0.23) ve Bor’un tüm dozlarının (0.25 mM: % 1.83±0.39 ve 1 mM: % 1.18±0.23), kontrol
grubuna (% 3.37±0.70) göre iyileşme sağladığı görüldü (P<0.01). Sunulan tez çalışmasında,
sulandırıcıya katılan kimi katkı maddelerinin çözüm sonu spermatozoa motilitesi ve DNA hasarı
üzerinde önemli oranda koruma sağladığı saptandı.
In the presented thesis, the effects of Rock inhibitor Y-27632, Antifreeze Protein III and Boron at two different doses were investigated on the spermatological parameters of Ankara buck semen after freeze-thawing. Ejaculates were collected from bucks using by an electroejaculator during the breeding season. The ejaculates that showed appropriate characteristics were pooled and used in the dilution and freezing of semen. The extender groups were formed by adding two different doses of three different additives (Rock inhibitor Y-27632 5 and 20 µM; Antifreeze Protein III 1 and 4 µg/ml; Boron 0.25 and 1 mM) to the control, and control extender. The semen were diluted with different extenders at 35-37 °C and loaded into straws. Sperm samples frozen in liquid nitrogen vapor following equilibration were stored at liquid nitrogen. It was observed that 5 and 10 µM (71.82±2.16 and 74.04±1.59 %) of Rock inhibitor and 0.25 and 1 mM doses of Boron (76.36±1.80 and 72.08±1.99 %), compared to the control group (66.15±1.51 %), improved post-thaw motility (P<0.01). Considering the evaluation of acrosomal integrity and mitochondrial activity after freezing-thawing; Although some groups provided protection at all doses, this efficacy was not statistically significant (P>0.05). It was observed that DNA damage was improved by Antifreeze Protein III 1 µg/ml (1.23±0.23 %) and Boron at all doses (0.25 mM: 1.83±0.39 % and 1 mM: 1.18±0.23 %), compared to the control group, (3.37±0.70 %) (P<0.01) following the thawing process. In the presented thesis, it was determined that some additives added to the extender provided significant improvements on the buck spermatozoa motility and DNA damage after thawing.
