Konya ve Afyon bölgesindeki ticari tavuk işletmelerinde marek hastalığının histopatolojik ve immunohistokimyasal yöntemlerle araştırılması

Özet
Bu çalışmada Afyonkarahisar ve Konya illeri çevresinde bulunan yumurtacı tavuk işletmelerinde Marek Hastalığının patolojk ve immunohistokimyasal yöntemlerle araştırılması amaçlandı. Çalışmada 11 farklı işletmeden 5 – 17 haftalık, 150 tavuk kullanıldı. Patolojik ve immunohistokimyasal araştırmalar için perifer sinirler, karaciğer, dalak, böbrek, kalp, akciğer, bursa Fabricius, beyin, bezli mide, deri ve bağırsak parçaları alındı. İmmunsitokimyasal incelemeler için perifer kandan alınan frotiler ile karaciğer, dalak ve böbreklerden alınan tuşe preparatlar indirekt immunperoksidaz yöntemiyle boyandı. Histopatolojik incelemelerde viseral organladaki lezyonlar hafif (+), orta (++) ve şiddetli (+++) olmak üzere ayrı ayrı skorlandı. N. ischiadicus lezyonları ise A-Tipi, B-Tipi ve C-Tipi lezyon olarak skorlandı. Klinik olarak bacak, kanat ve boyunda felçler ile balerin oturuşu gözlendi. Makroskobik bulgular çoğunlukla iç organlarda gözlenen tümöral oluşumlardan ibaretti. Tümöral oluşumlardan dolayı organların birkaç kat büyüklüğe ulaştıkları, aynı zamanda solgun ve açık renkte oldukları gözlendi. Bu tümöral yapıların tek ya da çok sayıda ve sarımsı – beyaz renkli odaklar şeklinde oldukları dikkati çekti. Mikroskobik incelemelerde ise iç organlarda genelde lenfositler, lenfoblastlar ve tek tük plazma hücrelerinden oluşan tümöral yapılar belirgindi. Daha şiddetli olgularda diffuz dağılımlı tümöral oluşumlar dikkati çekti. N. ischiadicusta 11 olguda A-tipi lezyonu gözlenirken, 18 olguda B-Tipi lezyon ve 89 olguda C-Tipi lezyon gözlendi. Klinik ve patolojik bulgularla 150 hayvandan 145'sinde Marek Hastalığı bulguları tespit edilmiştir. İmmunohistokimyasal boyamalarda 60 olguda çoğunlukla intrasitoplazmik, daha az da intranüklear ve serbest pozitif reaksiyonlar gözlendi. İmmunositokimyasal boyamalarda ise 119 olgudan perifer kanda pozitif reaksiyon belirlenmemesine rağmen, dalak ve karaciğer tuşe preparatlarında 29 olguda pozitif reaksiyon gözlendi. Klinik, makroskobik ve mikroskobik bulgular beraber değerlendirildiğinde Marek Hastalığı'nın uygulanan bilinçli aşılamalar sayesinde Afyonkarahisar ve Konya illerinde hafif şiddetli seyrettiği ortaya konmuştur. Hastalık teşhisi için klinik, makroskobik ve mikroskobik bulgular teşhis koymaya yardımcı birer yöntemi oldukları kanısına varılmıştır.
In this study, it was aimed to investigation of Marek's Disease with pathological and immunohistochemical methods in laying hen coops in vicinity of Afyon and Konya. In the study, 5 to 17 weeks aged 150 chickens were used which obtained from 11 different coops. Samples were taken from peripheral nerves, liver, spleen, kidney, heart, lung, bursa of Fabricius, brain, proventriculus, skin and intestines for pathological and immunohistochemical investigations. Collected samples from peripheral blood smears and touch preparations of liver, spleen and kidney were stained with indirect immunoperoxidase method for immunocytochemical investigations. In histopathological investigations, lesions in the visceral organs were scored as mild (+), moderate (++) and severe (+++). N. ischiadicus lesions were scored as A-Type, B-Type and C-Type. Leg, wing, neck strokes and fowl paralysis were seen in clinically. Mostly, macroscopic findings were consisted of tumor formation of the visceral organs. The organs reach a few times the size due to the tumor formation and they also observed to be paled and light colored. Drew attention to be unique or many of these tumor structures and yellowish – white colored form of focus. In microscopic examination of internal organs were pronounced that the tumor structures consist of usually lymphocytes, lymphoblasts and some plasma cells. Diffusely distributed tumor formation was noticed in more severe cases. In N. ischiadicus A-Type nerve lesion in 11 cases, B-Type lesion in 18 cases and C-Type lesion in 89 cases were observed. Marek's Disease signs were detected in 145 of 150 animals by clinical and pathological findings. Mostly intracytoplasmic, less intranuclear and free immunopositive reaction observed in 60 cases in immunohistochemical staining. In the liver and spleen touch preparations positive reactions were seen in 29 of 119 cases, although not identified positive reaction in peripheral blood in immunocytochemical staining. In recent years has been demonstrated that Marek's Disease has a mild severity prognosis in Afyon and Konya provinces owing to conscious immunizations, when considered in conjunction clinical, macroscopic and microscopic findings. It has been concluded that clinical, macroscopic and microscopic methods helps to diagnose.
