Koksidiyozisli Buzağılarda Bağırsak Hasarının Belirlenmesinde Bazı Bağırsak Biyobelirteçlerinin Değerlendirilmesi

Özet
Bu çalışmanın amacı, koksidiyozisli buzağılarda bağırsak epitel hasarının varlığı ve şiddetinin
belirlenmesinde bağırsağa özgü hasar biyobelirteçlerinin kullanılabilirliğinin değerlendirilmesidir.
Araştırmanın materyalini farklı ırk ve cinsiyette 21-60 günlük yaş aralığındaki 30 adet
koksidiyozisli (deneme grubu) ve 10 adet sağlıklı (kontrol grubu) olmak üzere toplam 40 buzağı
oluşturdu. Hastalığın teşhisi şüpheli buzağılardan alınan dışkının mikroskobik muayenesi sonucu
Eimeria ookistlerinin görülmesi ve klinik bulgular eşliğinde konuldu. Tüm buzağılardan 0. ve 72.
saatlerde antikoagülantlı ve antikoagülantsız kan örnekleri alındı. Antikoagülantlı kan örneklerinden
kan gazları ve hemogram ölçümleri yapıldı. Serum örneklerinden intestinal yağ asidi bağlayıcı protein
(İ-FABP), trefoil faktör-3 (TFF-3), intestinal alkalin fosfataz (İAP), claudin-3 (CLD-3), bağırsak düz
kas aktini (ACTG2) ve interlökin-8 (İL-8) biyobelirteçleri sığıra spesifik enzime bağlı immunosorbent
analiz (ELISA) test kitleri ile ölçüldü. Koksidiyozisli buzağılara toltrazuril (15 mg/kg tek doz) ve
destekleyici tedavi uygulandı. Tedavi uygulanan koksidiyozisli buzağıların tamamı iyileşti.
Koksidiyozisli buzağılarda emme refleksinde yetersizlik, dehidrasyon, tenesmus, kanlı ve
mukoid ishal belirlendi. Koksidiyozisli buzağılarda tedavi öncesi (0. Saat) serum İ-FABP ve CLD-3
düzeyleri sağlıklı buzağılara göre önemli oranda yüksek olduğu tespit edildi (p<0,05). Koksidiyozisli
buzağılarda tedavi sonrası (72. saat) serum TFF-3 ve ACTG2 düzeylerinin sağlıklı buzağılara göre
anlamlı şekilde arttığı belirlendi (p<0,05). Koksidizyozisli buzağıların tedavi sonrası (72. saat) serum
İL-8 düzeyinin tedavi öncesine göre (0.saat) önemli azaldığı saptandı (p<0,05). Koksidizyozisli
buzağıların 0.saatte kan pH’sı, parsiyel oksijen basıncı (pO2), oksijen saturasyonu (sO2), bikarbonat
(HCO3) ve sodyum (Na) düzeylerinde sağlıklı buzağılara göre azalma belirken, Laktat (Lac) düzeyinde
ise artış tespit edildi (p<0,05). Bununla beraber Koksidiyozisli buzağıların 0. saatte total lökosit sayısı
(WBC), granülosit sayısı (Gra), monosit sayısı (Mon), ortalama eritrosit hacmi (MCV), hematokrit
(HCT) ve ortalama korpusküler hemoglobin konsantrasyonu (MCHC) düzeylerinde sağlıklı buzağılara
göre artış tespit edildi (p<0,05).
Sonuç olarak, sunulan bu çalışmada İ-FABP, TFF-3, CLD-3, ACTG2 ve İL-8
biyobelirteçlerinin koksidiyozisli buzağılarda bağırsak epitel hasarının varlığının ve boyutunun
belirlenmesinde yararlı ve güvenilir biyobelirteçler olduğu ortaya konuldu.
The aim of this study was to evaluate the avability of intestine-specific injury biomarkers in determining the presence and severity of intestinal epithelial damage in calves with coccidiosis. The material of the study consisted of 40 calves of different races and sexes, aged between 21- 60 days, 30 with coccidiosis (experimental group) and 10 healthy (control group). The diagnosis of the disease was made in the presence of clinical findings and Eimeria oocysts as a result of microscopic examination of feces taken from suspicious calves. Anticoagulant and non-anticoagulant blood samples were taken from all calves at 0 and 72 hours. Blood gases and hemogram measurements were made from the blood samples with anticoagulant. Intestinal fatty acid binding protein (I-FABP), trefoil factor3 (TFF-3), intestinal alkaline phosphatase (IAP), claudin-3 (CLD-3), intestinal smooth muscle actin (ACTG2) and interleukin-8 from serum samples (IL-8) biomarkers were measured with bovine-specific enzyme-linked immunosorbent analysis (ELISA) test kits. Toltrazuril (15 mg/kg single dose) and supportive treatment were administered to calves with coccidiosis. All of the treated calves with coccidiosis recovered. Insufficient sucking reflex, dehydration, tenesmus, bloody and mucoid diarrhea were determined in calves with coccidiosis. Pre-treatment (0. hour) serum I-FABP and CLD-3 levels in calves with coccidiosis were found to be significantly higher than in healthy calves (p<0.05). It was determined that serum TFF-3 and ACTG2 levels increased significantly in calves with coccidiosis after treatment (72nd hour) compared to healthy calves (p<0.05). It was determined that the serum IL-8 level of calves with coccidiosis after the treatment (72nd hour) decreased significantly compared to the pre-treatment (0.hour) (p<0.05). While blood pH, partial oxygen pressure (pO2), oxygen saturation (sO2), bicarbonate (HCO3) and sodium (Na) levels were decreased in calves with coccidiosis compared to healthy calves, an increase was detected in lactate (Lac) levels (p<0.05). On the other hand, calves with coccidiosis have an increase in total leukocyte count (WBC), granulocyte count (Gra), monocyte count (Mon), mean erythrocyte volume (MCV), hematocrit (HCT) and mean corpuscular hemoglobin concentration (MCHC) levels at 0 hours compared to healthy calves. detected (p<0.05). In conclusion, in the present study, it was revealed that biomarkers I-FABP, TFF-3, CLD-3, ACTG2 and IL-8 are useful and reliable biomarkers in determining the presence and extent of intestinal epithelial damage in calves with coccidiosis.
