| dc.description.abstract | Metotreksat, bir folik asit antagonisti olup, dihidrofolat redüktaz enzimini inhibe eder ve bunun sonucunda DNA ve RNA sentezini inhibe ederek antiproliferatif etki gösteren bir ajandır. Anti proliferatif özelliği sebebiyle yüksek dozlarda kanser tedavisinde kullanılır. Ayrıca antiinflamatuvar etkisinden dolayı romatoid artrit tedavisinde yaygın olarak kullanılmaktadır. Yüksek doz Metotreksatın toksik etkilere sebep olduğu bilinmektedir. Kanser tedavisinde kullanılan metotreksat; nefrotoksisiteye yol açabilecek yan etkilere sahiptir. Nefrotoksisite glomerüler veya tübüler disfonksiyon, hipertansiyon ve renal endokrin fonksiyonda oluşabilecek bozukluklarla seyredebilir.Metotreksat tayini; dozajı optimize etmeye, toksisite riskini en aza indirmeye ve olası ilaç-ilaç etkileşimlerini araştırmaya yardımcı olmaktadır. Klinikte ilaç düzeyi ölçümünde hassasiyet çok önemli olduğu için kullandığımız yöntemin spesifitesi ve sensitivitesinin yüksek olması gerekmektedir. Bu çalışmada kullanmış olduğumuz yöntem Sıvı Kromatografi-Tandem Kütle Spektrometre (LC-MS/MS)’ de yeni bir ölçüm metodu geliştirdik. Metot validasyon çalışmamızda Metotreksat için 2500-0,305 ng/ml aralığı lineer bulunarak linearite grafiği çizilmiş ve korelasyon katsayısı (r2) değeri 1000-0,975 µg/l aralığında 0,998olarak hesaplanmıştır. LC-MS/MS’de sinyal gürültü oranına göre belirlenmiş olan dedeksiyon limiti 9 ng/ml, kantitasyon limiti 19 ng/ml ‘dir. Gerçekleştirmiş olduğumuz çalışmada serum Metotreksat tayini yapabilmede ortaya konulacak yeni yöntem için CLSI kuralları çerçevesinde metot validasyonu tamamlanmıştır. | en_US |
| dc.description.abstract | Methotrexate is a folic acid antagonist that inhibits the enzyme dihydrofolate reductase and as a result inhibits DNA and RNA synthesis and exhibits an antiproliferative effect. Due to its anti proliferative properties, it is used in cancer treatment in high doses. It is also widely used in the treatment of rheumatoid arthritis due to its antiinflammatory effect. High-dose methotrexate is known to cause toxic effects. Methotrexate used in cancer treatment; has side effects that may lead to nephrotoxicity. Nephrotoxicity may present with glomerular or tubular dysfunction, hypertension and impairments in renal endocrine function. Determination of methotrexate; it can help optimize dosage, minimize the risk of toxicity, and investigate potential drug-drug interactions. Since sensitivity is very important in drug level measurement in the clinic, the specificity and sensitivity of the method we use should be high. In this study, we developed a new measurement method in Liquid Chromatography Tandem Mass Spectrometer (LC-MS/MS) In our method validation study, 2500-0,305 ng / ml range was found linear by linear linear graph for Methotrexate and correlation coefficient (r2) value was calculated as 0,998 in the range of 1000-0,975 µg / l. Detection limit determined according to the signal to noise ratio in LC-MS / MS is 9 ng / ml and the quantitation limit is 19 ng / ml. In the study we have conducted, method validation has been completed within the framework of CLSI rules for the new method to be introduced in determining serum methotrexate. | en_US |
