Güneydoğu Anadolu Bölgesi’ndeki Koyun ve Keçilerde Anaplasmosisin Prevalansının Belirlenmesi

Özet
Bu çalışma Eylül 2019 tarihinde Güneydoğu Anadolu Bölgesi (Kilis, Gaziantep, Şanlıurfa,
Adıyaman, Diyarbakır, Mardin, Batman, Siirt, Şırnak)’nde koyun ve keçilerde Anaplasma
etkenlerinden Anaplasma ovis ve Anaplasma phagocytophilum’un yaygınlığını belirlemek amacıyla
yapılmıştır.
Güneydoğu Anadolu Bölgesi’ndeki koyun ve keçilerde anaplasmosis etkenlerinin
yaygınlığını belirlemek için yapılan bu çalışmada mikroskobik muayene yöntemi ve Rt-PCR testi
uygulanmıştır. Mikroskobik muayene ile 384 koyun frotisinin 84 (%21,88)'ünde, 384 keçi kan
frotisinin 152 (%39,58)’sinde Anaplasma spp. etkenleri tespit edilmiştir. Koyun ve keçi kan
numunelerinin tamamına DNA ekstraksiyonu yapıldıktan sonra Rt-PCR testi uygulanarak koyun
numunelerinin %89,32(343)’sinde A. ovis, %42,97 (165)’sinde A. phagocytophilum; keçi
numunelerinin %78,91(303)’inde A. ovis, %15,89 (61)’unda A. phagocytophilum tespit edilmiştir.
Çalıma sonucunda Rt-PCR yönteminin mikroskobik muayene yöntemine göre daha kesin ve spesifik
sonuçlar verdiği görülmüştür. Çalınmada, Anaplasma türleri mikroskobik muayene ile keçilerde
koyunlara göre daha yüksek oranda tespit edilmiştir. Çalışmada Anaplasma türleri Rt-PCR ile
koyunlarda keçilere göre daha yüksek oranda tespit edilmiştir.
This study was carried out to determine the prevalence of Anaplasma ovis and Anaplasma phagocytophilum in sheep and goats in Southeastern Anatolia Region (Kilis, Gaziantep, Sanliurfa, Adiyaman, Diyarbakir, Mardin, Batman, Siirt, Sirnak) in September 2019. The microscopic examination method and Rt-PCR test were applied in this study to determine the prevalence of anaplasmosis agents in sheep and goats in the Southeastern Anatolia Region. Anaplasma spp agents were detected in 84 (21,88%) of 384 sheep smears and 152 (39,58%) of 384 goat blood smears by microscopic examination. After DNA extraction was performed on all sheep and goat blood samples, a Rt-PCR test was applied, and A. ovis was detected in 89,32% (343), A. phagocytophilum in 42,97% (165) of sheep samples; A. ovis was detected in 78,91% (303) and A. phagocytophilum in 15,89% (61) of goat samples. As a result of the study, it was seen that the Rt-PCR method gave more precise and specific results than the microscopic examination method. In the study, Anaplasma spp. were detected at a higher rate in goats than sheep by microscopic examination. In the study, Anaplasma spp. were detected at a higher rate in sheep than goats by Rt-PCR.
