Özet
Bu çalışmada; ülkemizde yumruları salep olarak kullanılan bazı türlerin (Serapias vomeracea
(Burm.f.) Briq., Ophrys apifera Huds. ve Anacamptis pyramidalis (L.) Rich) vejetatif ve asimbiyotik
üretim olanakları araştırılmıştır. Doku kültürü yöntemi ile in vitro koşullarda S. vomeracea ve A.
pyramidalis türlerine ait tohumlar EBP besi ortamında çimlenirken O. apifera’da çimlenme
gözlemlenememiştir. Çimlenen ve gelişen fidelerin aklimatizasyon çalışması yapılmıştır, fakat bitkiler
15-45 günde hızla solarak canlılığını kaybetmiştir. Tarla ve sera şartlarında yumru dikimi ile üretiminden
fenolojik, morfolojik ve kimyasal bileşenlerine ait veriler kaydedilmiştir. Tarla koşullarında üretimi için
türlerin doğal florası olan Samsun’da deneme kurulmuş olup bitkisel ve kimyasal özellikleri kaydedilmiş
ayrıca Ankara şartlarında gölgelendirme serasında türlerin adaptasyonu ve bazı fenolojik gözlemleri
alınmıştır. Denemede son birkaç yıldır kullanılan sök ve yerine yeniden dik olarak adlandırılan işlem
uygulanmıştır ve tarla şartlarında salep yumru sayısının çoğaltılması için çiçeklenme başlangıcında
kullanılabileceği ama salep yumru ağırlığının artırılmasına etkisi gözlemlenmemiştir. Çiçeklenme
başlangıcı, % 50 çiçeklenme dönemi ve çiçeklenme sonunda ‘sök-dik’ yöntemi uygulanarak tesadüf
bloklarında bölünmüş parseller deneme desenine göre üç tekerrürlü olarak yürütülmüştür. Denemeden,
A.pyramidalis, O.apifera, S. vomeracea, tür sıralamasına göre bitkisel özelliklerine ait elde edilen
değerler: bitki boyları 37.91-39.25-47.08 cm, yaprak sayıları 7.5-5.92-5.83 adet/bitki, 5.83-7.5 adet/bitki,
çiçek sayıları 77.25-7.33-9.25 adet/bitki, yavru yumru sayıları 1.9-1.7-1.6 adet/bitki, yumru ağırlıkları
5.92-5.14-5.32 g/bitki, yumru büyüklükleri 2.4-1.8-1.8 cm3
’dür. Kullanılan üç farklı salep orkidesi
türünün ortalama nişasta oranı % 11.41-38.9, müsilaj oranı %23.9-26., nem oranı % 7.06-8.22, ham
protein oranı % 5.45-6.45, toplam makro besin elementi %0.27-0.53, toplam mikro besin elementi 166-
203 Ppb arasında tespit edilmiştir.
In this study, vegetative and asymbiotic production possibilities of some species (Serapias vomeracea (Burm.f.) Briq., Ophrys apifera Huds. and Anacamptis pyramidalis (L.) Rich) whose tubers are used as salep in our country were investigated. Under in vitro conditions with tissue culture method, while seeds of S. vomeracea and A. pyramidalis species germinated in EBP medium, no germination was observed in O. apifera. The acclimatization study of the germinated and developed seedlings was conducted, but the plants quickly withered in 15-45 days and lost their vitality. The data on the phenological, morphological and chemical components of tuber planting and production of these salep species under field and greenhouse conditions were recorded. For the production under field conditions, the experiment was established in Samsun, which is the natural flora of the species, and its herbal and chemical properties were recorded. In addition, the adaptation of the species and some phenological observations were taken in the shading greenhouse under Ankara conditions. In the experiment, the process called uproot and upright, which has been used for the last few years, was applied and it could be used at the beginning of flowering to increase the number of salep tubers under field conditions, but no effect on increasing the weight of salep tubers was observed. In the beginning of flowering, in the middle of the flowering period (50 % flowering) and at the end of the flowering, the 'removal-upright' method was applied in randomized blocks according to the divided plots experimental design with three replications. The values obtained for the vegetative characteristics of A.pyramidalis, O.apifera, S. vomeracea species in the experiment, respectively: plant heights 37.91-39.25-47.08 cm, number of leaves 7.5-5.92-5.83 /plant, 5.83-7.5 units/plant, flower numbers 77.25-7.33-9.25 units/plant, juvenile tuber numbers 1.9-1.7-1.6 units/plant, tuber their weight is 5.92-5.14-5.32 g/plant, tuber size is 2.4-1.8-1.8 cm3. The ratios of starch, crude protein, mucilage, moisture, macro and micro nutrients in the composition of the species were determined. Average starch ratio of three different salep orchid species was detected between 11.41% and 38.9%, mucilage ratio 23.9-26%, moisture ratio 7.06-8.22%, crude protein ratio 5.45-6.45%, total macronutrient 0.27-0.53%, total micronutrient 166-203 Ppb.
