dc.description.abstractBu çalışmada Türkiye'deki üç farklı yarasa türünün perifer kan lenfosit oranları ile alfa naftil asetat esteraz (ANAE) ve asit fosfataz (ACP-az) pozitif lenfosit oranlarının işik mikroskobik düzeyde belirlenmesi amaçlanmıştır. Bu amaçla dört Rhinolophus ferrumequinum (Büyük nal burunlu yarasa; 29, 28, Kilis), dört Pipistrellus kuhlii (Beyaz yakalı yarasa; 29, 23, Mersin) ve beş Tadarida teniotis (Kuyruklu yarasa; 49. 18. Şanlıurfa) örneğinden usulüne uygun olarak kan alınmış ve her örnekten altı adet froti hazırlanarak havada kurutulmuştur. Rutin May-Grünwald Giemsa yöntemiyle boyanan iki frotide lenfosit oranları belirlenmiştir. Diğer frotiler fosfat tamponlu gluteraldehit asetonda (-10ºC) 3 dakika tespit edilmiştir. Bu frotilerden ikisine ANAE demonstrasyonu, son ikisine de ACP-az demonstrasyonu uygulanmıştır. ANAE demonstrasyonu sonucunda lenfositler 1-3 arasında değişen sayıda kiremit kirmizisi kahverengi granülden ibaret nokta tarzında pozitivite göstermiştir. Monositler sitoplazmalarında güçlü ve yaygın tarzda pozitivite gösterirken, nötrofiller ise zayıf bir reaksiyon vermiştir. ACP-az demonstrasyonu sonucunda ise bu çalışmada incelenen her üç yarasa türünün lenfositlerinde ACP-az pozitivitesine özgü granüllere rastlanmamıştır. R. ferrumequinum, P. kuhlii ve T. teniotis için ortalama lenfosit oranları sırasıyla h.9426.53, a.58±7.14 ve r.80±3.18, ortalama ANAE pozitif lenfosit oranları ise R.612.55, D.769.07 ve F.29+3.48 olarak belirlenmiştir. Lenfosit oranı bakımından P. kuhlii ve T. teniotis arasındaki fark ile ANAE pozitif lenfosit oranı bakımından R. ferrumequinum ve T. teniotis arasındaki fark istatistiksel olarak önemli düzeyde bulunmuştur (p<0.05).en_US
dc.description.abstractThe aim of this study was to determine peripheral blood lymphocyte (PBL). alpha-naphthyl acetate esterase (ANAE) and acid phosphatase (ACP-ase) positive PBL percentages of three bat species (Rhinolophus ferrumequinum 29, 28, Kilis; Pipistrellus kuhlii 29, 23, Mersin; and Tadarida teniotis 49, 18, Şanliurfa) in Turkey. For this purpose, six smears were prepared and air dried from each animal. Two of them were stained with May Grünwald-Giemsa for the determination of PBL percentages. Four smears were fixed in glutaraldehyde-acetone solutions (pH 4.8) at - 10°C for 3 minutes. Two of the smears were used for ANAE demonstration. The remaining two smears were used for ACP-ase demonstration. ANAE positive lymphocytes displayed a dot like positivity pattern characterized with 1-3 reddishbrown cytoplasmic granules. Monocytes gave a diffuse and strong reaction while neutrophils displayed a weak positive reaction in their cytoplasms for ANAE. However, lymphocytes were not displayed ACP-ase positivity in all bat species investigated. Mean PBL ratios were 68.94+6.53%, 61.58+7.14% and 72.80±3.18%, and mean ANAE positivity ratios of PBL were 52.61±2.55%, 44.76+9.07% and 46.29±3.48% for R. ferrumequinum, P. kuhlii and T. teniotis, respectively. The differences in PBL ratios between P. kuhli and T. teniotis and in ANAE positivity ratios of PBL between R. ferrumequinum and T. teniotis were statistically significant (p<0.05).en_US
