Konak Galleria mellonella L. ve endoparaziroid Pimpla turionellae L.'nın hemolenf protein bileşimine parazitlemenin etkileri

Özet
Bu çalışmada, konak Galleria mellonella L. ve endoparazitoid Pimpla turionellae L.'nin larval ve pup gelişim evrelerindeki hemolenf proteinleri ile parazitlemenin konak hemolenf proteinlerine etkileri kolorimetrik ve sodyum dodesil sülfat-poliakrilamid jel elektroforez (SDS - PAGE) yöntemleri ile araştırılmıştır. Kolorimetrik analizlerinde, konak ve endoparazitoid türün her ikisinde de larval evreden pup evresine doğru hemolenf protein düzeyinin arttığı, pup evresinde ise ergin evreye doğru azaldığı tesbit edilmiştir. Parazitlenmiş konakta, parazitlenmemiş kontrollere göre hemolenf protein düzeylerinde ilk saatte azalma olurken, ilerleyen saatlerde artışın olduğu saptanmıştır. III SDS - PAGE analizlerinde ise, G. mellonella ve P. turionellae 'de her bir türe özgü ve ortak proteinlerin bulunduğu görülmüştür. Protein profilleri, larval evreden pup evresine doğru farklılıklar göstermiştir. 33 kD ve >205 kD'luk proteinlerinin her iki türün larval evresine spesifik olduğu görülmüştür. G. mellonella' da. 73, 75, 83 ve 88 kD'luk ve P. turionellae' 'da ise 65, 69, 74 ve 83 kD'luk depo proteinleri saptanmıştır. Parazitlenmenin hemolenf protein profilleri üzerine etkileri araştırıldığında ise, parazitlerime ile 15.5, 17.5, 67 ve 82 kD'luk proteinlerin arttığı ve 18, 23, 26, 34, 36, 42, 46 ve >205 kD'luk proteinlerinin ise azaldığı görülmüştür. Parazitlerime ile konak hemolenf proteinlerinin etkilendiği her iki yöntemde de ortaya çıkmıştır.
In this study, hemolymph proteins in the developmental stages of larvae and pupae of host Galleria mellonella and its endoparasitoid Pimpla turionellae and the effects of parasitism on hemolymph proteins were investigated by the colorimetric and sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) methods. In colorimetric analysis, hemolymph protein levels in both host and endoparasitoid increased from the larval to pupae stages, while it decreased from pupae to adult stage. In parasitized host, unlike unparasitized controls, a decrease at the hemolymph protein levels was seen during the first hour and in advancing hours an increase was observed. In SDS-PAGE analysis, it was seen that some proteins were common in both G. mellonella and P. turionellae and some specific to each species. Protein profiles V showed differences from larvae to pupae stages. It was seen that 33 kD and >205 kD molecular weight proteins were specific at the larval stages of both G. mellonella and P. turionellae. 73, 75, 83 and 88 kD storage proteins in G. mellonella and 65, 69, 74 and 83 kD in P. turionellae were found. When the effects of parasitism on hemolymph protein profiles were examined, it was observed that levels of 15.5, 17.5, 67 and 82 kD proteins increased and 18, 23, 26, 34, 36, 42, 46 and >205 kD proteins decreased. The both methods showed that host hemolymph proteins were affected by parasitization.
