Ratlarda Streptozotosin (STZ) ile İndüklenen Deneysel Diyabet Modelinde Bazı Hemostatik Parametreler Üzerine Egzersizin Etkileri

Özet
Bu araştırmada streptozotosin (STZ) ile deneysel diyabet oluşturulan sıçanlarda düzenli
egzersiz uygulamasının bazı hemostatik parametreler üzerine etkilerinin belirlenmesi amaçlanmıştır.
Çalışmada, 36 adet yetişkin, sağlıklı ve canlı ağırlıkları birbirine yakın Wistar Albino rat
kullanıldı. Deneme başlamadan önce ratlar Kontrol (K), Diyabet (D), Egzersiz (E) ve Egzersiz+
Diyabet (ED) olmak üzere 4 gruba ayrıldı. Bütün gruplar deneme boyunca standart sıçan yemi ile
beslendi. D ve ED gruplarına 60 mg/kg STZ intraperitoneal enjeksiyon ile tek doz olarak uygulandı. E
ve DE gruplarına 4 hafta boyunca koşu bandında 20 m/dak hızda günlük 45 dak. egzersiz yaptırıldı.
Deneme sonunda deneklerden elde edilen verilerde D ve ED grubundaki insülin değerlerinin
kontrol gruplarına göre anlamlı düzeyde düşük olduğu tespit edildi (p<0.05). Ancak ED grubundaki
insülin düzeyin her ne kadar K ve E gruplarına göre düşük olsada D grubundaki verilere göre belirgin
olarak yüksek olduğu belirlendi (p<0.05). D grubunda serum fibrinojen, D-Dimer, PAI-1, Faktör VIII,
t-PA ve TAT düzeyleri K grubuna göre belirgin bir artış gösterdiği gözlendi (p<0.05). Buna karşın K
grubuna kıyasla D grubunda PT düzeyinde anlamlı düzeyde düşük (p<0.05), ATIII ve aPTT
düzeylerinde ise rakamsal olarak azalma (p>0.05) tespit edildi. Ayrıca K grubuna kıyasla Faktör VIII
ve TAT düzeyleri sadece egzersiz uygulanan ratlarda (E grubu) belirgin olarak yüksekti (p<0.05). E
grubunda diğer parametreler ise (fibrinojen, D-Dimer, PAI-1, t-PA, ATIII, aPTT, PT) K grubundan
elde edilen veriler ile benzer olduğu tespit edildi (p>0.05). Ayrıca egzersizin koruyucu etkinliğini
belirlemek için oluşturulan ED grubunda serum PAI-1 ve Faktör VIII düzeylerinin D grubuna kıyasla
önemli oranda azaldığı belirlendi (p<0.05). ED gruplarından elde edilen söz konusu parametrelere ait
değerlerin E grubuna göre insülin ve PT değerleri dışında, K grubunda ise PAI-1, Faktör VIII ve PT
dışında K ve E gruplarına yaklaşma eğiliminde veya benzer olduğu gözlendi.
Sonuç olarak, bu çalışmada deneysel diyabet oluşturulan sıçanlarda alternatif bir tedavi
metodu olarak düşünülen egzersizin; insulin ve bazı hemostatik parametre (D-dimer, PAI-1, Faktör
VIII, t-PA, TAT, ATIII, aPTT, PT ve fibrinojen) düzeyleri üzerine diyabetik sıçanlarda olumlu
etkisinin olmasının yanı sıra E grubundaki sıçanlarda olumsuz etki oluşturmaması bakımından dikkate
değer bir yöntem olduğu gözlenmektedir.
In this study, it was aimed to determine the effects of regular exercise on some hemostatic parameters in rats with experimental diabetes mellitus with streptozotocin (STZ). In the study, 36 adult healthy Wistar Albino rats with similar live weights were used. Before the start of the experiment, the rats were divided into 4 groups as Control (C), Diabetes (D), Exercise (M), and Exercise+Diabetes (ED). All groups were fed with standard rat chow throughout the experiment. In the D and ED groups, 60 mg/kg STZ was administered as a single dose by intraperitoneal injection. E and ED groups were given 45 minutes of daily exercise at 20 m/min on the treadmill for 4 weeks. In the data obtained from the subjects at the end of the experiment, it was determined that the insulin values in the D and ED groups were significantly lower than the control groups (p<0.05). However, although the insulin level in the ED group was lower than the C and E groups, it was found to be significantly higher than the data in the D group (p<0.05). It was observed that serum fibrinogen, D-Dimer, PAI-1, Factor VIII, t-PA and TAT levels in group D showed a significant increase compared to group C (p<0.05). On the other hand, PT levels were significantly lower (p<0.05) and ATIII and aPTT levels were numerically decreased (p>0.05) in D group compared to C group. In addition, Factor VIII and TAT levels were significantly higher in rats that were only exercised (group E) compared to group C (p<0.05). In group E, other parameters (fibrinogen, D-Dimer, PAI-1, t-PA, ATIII, aPTT, PT) were found to be similar to the data obtained from group C (p>0.05). In addition, serum PAI-1 and Factor VIII levels were found to be significantly decreased in the ED group, which was created to determine the protective effectiveness of exercise, compared to group D (p<0.05). It was observed that the values of the mentioned parameters obtained from the ED groups, except for insulin and PT values in the E group, tended to approach or were similar to the C and E groups, except for PAI-1, Factor VIII and PT in the C group. As a result, exercise, which is considered as an alternative treatment method in experimental diabetes-induced rats in this study; In addition to having a positive effect on insulin and some hemostatic parameter (D-dimer, PAI-1, Factor VIII, t-PA, TAT, ATIII, aPTT, PT and fibrinogen) levels in diabetic rats, it was notable in that it did not have a negative effect on rats in group E. method is observed.
