Özet
Bu çalışma, ratlarda metabolik sendrom (MetS) oluşumunun ardından uygulanan yüzme egzersizinin plazma irisin, adropin, insülin ve leptin hormonları ile biyokimyasal parametrelere etkilerinin araştırılması amacıyla gerçekleştirildi. Çalışmaya başlamadan önce canlı ağırlıkları belirlenen 70 adet erkek Wistar Albino ratın 6 tanesi sakrafiye edilerek 0. gün biyokimyasal parametreleri saptandı. Geriye kalan 64 adet rat kontrol ve metabolik sendrom grubu (MetS) olmak üzere 2 gruba ayrıldı. Deneme süresince standart rat yemi ile beslenen ratlardan MetS grubunun içme sularına %20 fruktoz ilavesi yapıldı. Yapılan kontrollerin sonunda ATP III kriterlerine göre (artan glikoz, vücut kitle indeksi, LDL, total kolesterol, trigliserid ile azalan HDL) MetS oluşumu incelendi. MetS oluşumunun ardından, kontrol ve MetS grupları egzersiz yapan ve yapmayan olmak üzere iki alt gruba ayrıldı. Yüzme egzersizi günde 30 dakika süre ile 4 hafta boyunca gerçekleştirildi. Egzersiz programının sonunda tekrar canlı ağırlıkları belirlenen tüm gruplardaki ratlar sakrafiye edilerek, plazma irisin, adropin, insülin ve leptin düzeyleri EIA yöntemi ile serum glikoz, total kolesterol, HDL, LDL, trigliserid, AST, ALT, GGT ve LDH düzeyleri otoanalizör kullanılarak spektrofotometrik yöntemlerle belirlendi. Metabolik sendrom grubunun biyokimyasal parametreleri incelendiğinde; glikoz, total kolesterol, LDL, trigliserid düzeylerinin arttığı (p<0,05), HDL'nin ise azaldığı belirlendi. Aynı şekilde insülin, leptin (p<0,05) ve irisin düzeylerinde artış, adropin (p<0,05) düzeylerinde ise düşüşler gözlendi. Egzersiz protokolünün sonunda ise metabolik sendromlu ratlarda egzersizin glikoz düzeylerini düşürdüğü belirlendi (p<0,05). Ayrıca aynı grupta insülin, irisin, leptin ve adropin düzeylerinde istatiksel olarak anlamlı olmayan düşüşler gözlendi. Bunun yanında egzersizin birçok biyokimyasal parametrede artış ve düşüşlere yol açtığı ancak istatistiki olarak önemli olmadığı görüldü. Egzersiz yapan MetS grubunda, egzersiz ile birlikte vücut kitle indeksindeki azalmaya paralel olarak leptin ve irisin düzeylerinde düşüş gözlendi. Sonuç olarak özellikle irisin ve adropin gibi oldukça yeni hormonların metabolik sendrom oluşum mekanizması ve tedavisinde etkilerinin araştırılması amacıyla farklı tip egzersizlerin yanında egzersiz sürelerinin de değişkenlik gösterdiği yeni çalışmaların yapılması gereğini ortaya koymaktadır.
This study was performed to investigate the effects of swimming exercise on plasma insulin, leptin, irisin, adropin levels and biochemical parameters in rats with Metabolic syndrome. 70 male Wistar Albino weighed at the start of the study, 6 of them were euthanized to 0. day biochemical parameters were determined. The remaining 64 rats were divided into 2 groups; control group and the metabolic syndrome (MetS). Control group fed with standard chow diet, MetS group fed with 20% fructose in drinking water. Development of MetS was evaluated according to the criteria of ATP III (increased glucose, body mass index, LDL, total cholesterol, triglyceride and decreased HDL). After the formation of MetS, control and MetS groups were divided into two sub-groups including exercise and non-exercise. Swimming exercise was performed for 30 minutes a day for 4 weeks. After exercise program, the rats in all groups were weighed and euthanized and then their plasma hormone levels were determined by EIA method, their biochemical parameters (glucose, total cholesterol, HDL, LDL, triglyceride, AST, ALT, GGT, LDH levels) were determined by spectrofotometric method. In metabolic syndrome group, increased glucose, total cholesterol, LDL, trigliserid (p<0,05) and decreased HDL were determined. The same way an increase was observed in leptin, insulin (p<0,05) and irisin levels, while declines in adropin levels (p<0,05). At the end of the exercise protocol, it was determined that exercise reduces glucose level in rats with MetS (p<0,05). Also, a slight decrease in insulin, irisin, leptin and adropin levels were observed. Although plasma leptin and irisin levels increased on metabolic syndrome group, body mass index, leptin and irisin levels decreased on metabolic syndrome exercise (MetSEG) groups. In conclusions, it is suggested that new studies in which the durations of exercise in addition to different types of exercises change need to be done in order to the effects of new hormones especially such as irisin and adropin in metabolic syndrome and its treatment.
