Sıçanlarda Deneysel Böbrek İskemi-Reperfüzyonunda Naringin'in Kaspaz-3, IL-1β ve HIF-1α Düzeylerine Etkisi

Özet
İskemi reperfüzyon (I/R) sonrası dokularda mikrovaskuler fonksiyon bozukluğu gelişir.
Mevcut çalışmanın amacı tek taraflı nefrektomi ve böbrek-iskemi reperfüzyonu yapılan sıçanlarda
naringin takviyesinin böbrekte kaspaz-3, IL-1β ve HIF-1α düzeyleri ile böbrek histolojisine olan etkisini
belirlemekti.
Selçuk Üniversitesi Deney Hayvanları Araştırma ve Uygulama Merkezi ile Necmettin Erbakan
Üniversitesi Deney Hayvanları Araştırma ve Uygulama Merkezinden temin edilen 8-12 haftalık Wistar
tipi erkek 40 sıçan üzerinde gerçekleştirildi.
Ratlarda deneysel böbrek iskemi reperfüzyonu ile tek taraflı nefrektomi genel anestezi altında
gerçekleştiridi. Deney grupları şu şekilde oluşturuldu.
1) Kontrol grubu,
2) Sham kontrol + Vehicle grubu.
3) Böbrek iskemi-reperfüzyonu (Renal İ+R) + Vehicle grubu
4) Renal İ+R + Naringin (50 mg/kg/gün) grubu (3 gün uygulama) grubu
5) Renal İ+R + Naringin (100 mg/kg/gün) grubu (3 gün uygulama) grubu.
İskemi reperfüzyon gruplarında sol böbreklerde nefrektomi, sağ böbreklerde ise 45 dakika
iskemi ve takiben 72 saat reperfüzyon gerçekleştirildi. Naringin reperfüzyonun hemen başında, 24 saat
sonra ve 48 saat sonra intraperitoneal olarak uygulandı. Deneylerin bitiminde genel anestezi altındaki
hayvanlarda öncelikle kalpten kan alındıktan sonra servikal dislokasyonla hayvanlar öldürülerek böbrek
doku örnekleri alındı. Dokular kaspaz-3, IL-1β ve HIF-1α yönünden ve ayrıca histolojik olarak
değerlendirildi.
Böbrek dokularında iskemi-reperfüzyon sonucu HIF-1α azalırken kaspaz-3, IL-1β artış
gösterdi. İ/R aynı zamanda böbrek dokusunda hasar oluşturdu. Ancak naringin takviyesi oluşan
bozulmaları belirli oranda düzeltti.
Çalışmanın sonuçları sıçanlarda tek taraflı nefrektomi ve İ/R ile oluşan böbrek hasarında
antiinflamutuvar ve antipoptozis mekanizmaları üzerinden koruyucu etkilere sahip olabileceğini
göstermektedir.
Microvascular dysfunction develops in tissues after ischemia-reperfusion (I/R). The aim of the current study was to determine the effect of naringin supplementation on kidney caspase-3, IL-1β and HIF-1α levels and kidney histology in rats undergoing unilateral nephrectomy and kidney-ischemia reperfusion. The study was carried out on 8-12 weeks old 40 Wistar type male rats obtained from Selçuk University Experimental Animals Research and Application Center and Necmettin Erbakan University Experimental Animals Research and Application Center. Experimental renal ischemia reperfusion and unilateral nephrectomy were performed under general anesthesia in rats. Experimental groups were formed as follows. 1) Control group, 2) Sham control + Vehicle group. 3) Renal ischemia-reperfusion (Renal İ+R) + Vehicle group 4) Renal I+R + Naringin (50 mg/kg/day) group (3 days application) group 5) Renal I+R + Naringin (100 mg/kg/day) group (3 days application) group. Nephrectomy in the left kidneys and the ischemia for 45 minutes and reperfusion in the right kidneys followed by 72 hours of reperfusion. Naringin was administered intraperitoneally at the beginning of the reperfusion, 24 hours, and 48 hours later. At the end of the experiments, first of all, blood was taken from the heart in animals under general anesthesia, and then the animals were killed by cervical dislocation and kidney tissue samples were taken. Tissues were evaluated for caspase-3, IL1β and HIF-1α as well as histologically. As a result of ischemia in kidney tissues, HIF-1α decreased, while caspase-3, IL-1β increased. I/R also caused damage to the kidney tissue. However, naringin supplementation corrected the deterioration to a certain extent. The results of the study show that it may have protective effects in kidney damage due to antiinflammatuar and antipoptosis mechanisms caused by unilateral nephrectomy and I/R in rats.
