| dc.description.abstract | Bu çalısmanın amacı matriks metalloproteinaz (MMP)-1 -1607 polimorfizminin periodontal hastalık ve diseti olugu sıvısı (DOS) MMP-1 ve MMP doku inhibitörü (TIMP)-1 düzeyleri arasındaki iliskiyi arastırmaktı. Çalısmaya toplam 180 birey dahil edildi. Çalısmaya dahil edilen bireylerde kardiyovasküler hastalıklar, diabet, immün sistem hastalıkları yoktu veya insan immun yetmezlik virüsü ?Human immunodeficiency virus? (HIV), Hepatit A, B veya C virusleri tasımıyorlardı. Çalısmaya dahil olan bireylerin sondlama cep derinlikleri (SCD), klinik atasman kaybı (KAK), plak indeksi (Pİ), gingival indeksi (Gİ), dis mobilitesi ölçüldü. Alveoler kemik kaybı (AKK) yüzdeleri ise panoramik radyograflardan schei cetveli yardımı ile ölçüldü. Bireylerin sigara kullanım miktarları da degerlendirildi. Diseti olugu sıvısı MMP- 1 ve TIMP-1 düzeyleri ELISA kiti yardımıyla belirlendi. Matriks metalloproteinaz-1 -1607 polimorfizmi polimeraz zincir reaksiyonu ?polimerase chain reaction? (PCR) ve kesim uzunlugu polimorfizmi ?restriction fragment length polymorphism? (RFLP) analizi ile degerlendirildi. Bu çalısmada 1G alleline ait frekanslarda saglıklı bireylerde %46.2, orta siddette periodontitiste %48.8, ve siddetli periodontitiste %43 olarak tespit edilmistir. 2G ?allel?line ait frekanslar da saglıklı bireylerde %53.8, orta siddette periodontitiste %51.2, ve siddetli periodontitiste %57 olarak tespit edildi. Bu arastırmanın sınırları içerisinde sigara içen ve içmeyen bireyler arasında DOS MMP- 1 ve TIMP-1 düzeyleri önemli bir fark göstermedi ancak sigara içmeyen bireylerde KAK'nın ve SCD'nin MMP-1 total miktarı ile iliskisi anlamlıydı, ancak bu durum sigara içenlerde önemli degildi. Bu çalısmada saglıklı ve periodontitisli bireyler arasında MMP-1 genotip dagılımının benzer oldugu ve polimorfizmin periodontal hastalık siddeti ve DOS MMP-1 ve TIMP-1 düzeyleri ile iliski göstermedigi bulunmustur. | en_US |
| dc.description.abstract | The aim of this study was to evaluate the relations among periodontal disease, matrix metalloproteinase (MMP)-1 -1607 polymorphism and gingival crevicular fluid (GCF) MMP-1 and tissue inhibitor of MMP (TIMP)-1 levels. A total of 180 subjects were included in this study. Subjects had no history of cardiovascular diseases, diabetes mellitus, immunogenic disorders, or carrying Human immunodeficiency virus (HIV), Hepatitis A, B or C viruses. Clinical parameters including probing depth (PD), clinical attachment loss (CAL), plaque indeks (PI), gingival indeks (GI), tooth mobility were measured. Alveolar bone loss (ABL) was measured by means of schei ruller on panogramic radiographs. Cigarette smoking status of the subjects were also assesed. Levels of GCF MMP-1 and TIMP-1 were analyzed with ELISA. Polymorphism of MMP-1 - 1607 was determined with polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) analysis. In this study, 1G allele was observed at a frequency of %46.2 at healthy subjects, %48.8 at mild to moderate periodontitis subjects and %43 at severe periodontitis subjects. The 2G allele was observed at a frequency of of %53.8 at healthy subjects, %51.2 at mild to moderate periodontitis subjects and %57 at severe periodontitis subjects. Within the limits of this study the GCF MMP-1 and TIMP-1 levels were not different significiantly between smokers and non smokers but among non smokers the CAL and PD values were correlated with MMP-1 total amounts. There were no corralation between clinical parameters and biochemical findings in the smokers group. In this study the MMP-1 -1607 genotype distribution was similar in both healthy and periodontitis groups and it did not seem to be associated with susceptibility to periodontitis and GCF MMP-1 and TIMP-1 levels. | en_US |
