Özet
Bu çalışmada, Diyarbakır Devegeçidi Köprüsü civarından, 2007 yılı haziran ayında toplanan tatlısu salyangozu Melanopsis praemorsa'nın total vücut lipitleri; ince tabaka kromatografi ile fraksiyonlandı. Salyangozun, total vücut lipitleri, fosfolipit ve nötral lipit fraksiyonundaki yağ asitleri ile besinini oluşturan tatlısu alglerinin yağ asitleri gaz kromatografi ve gaz kromatografi-kütle spektrometresi (GC-MS) ile analizlendi. Analizlerde, doymuş yağ asitlerinden C10:0, C12:0, C13:0, C14:0, C15:0, C16:0, C17:0, C18:0; tekli doymamış yağ asitlerinden C16:1?7, C18:1?9, C20:1?9 ve çoklu doymamış yağ asitlerinden C18:2?6, C18:3?3, C20:2?6, C20:4?6, C20:5?3 ve C22:2?6 asitler saptandı. Tek karbonlu ve C20:2?6, C20:4?6, C20:5?3 ve C22:2?6 gibi 20 karbonlu çoklu doymamış yağ asitleri, gaz kromatografi-kütle spektrometre ile doğrulandı. Lipit fraksiyonlarında bu bileşenlerin yüzde değerleri karşılaştırıldı. Yüzde dağılımda en çok C16:0, C18:1?9 ve C18:2?6 asitler tespit edildi. Fosfolipit, nötral, total ve besin (alg) lipitlerinin yüzde içeriğinde bazı farklar tespit edildi.
In this study, body lipids of a freshwater snail Melanopsis praemorsa collected from Devegeçidi Bridge in Diyarbakır in June 2007, were fractionated by thin layer chromatography (TLC). Total lipids, fractionated lipids such as phospholipids and neutral lipids of whole snail and algae as diets were analysed by capillary gas chromatography and gas chromatography-mass spectrometry. In the analysis, saturated fatty acids such as C10:0, C12:0, C13:0, C14:0, C15:0, C16:0, C17:0, C18:0; monounsaturated fatty acids such as C16:1?7, C18:1?9, C20:1?9 and polyunsaturated fatty acids such as C18:2?6, C18:3?3, C20:2?6, C20:4?6, C20:5?3, C22:2?6 asids were found. Odd-numbered fatty acids and C20 polyunsaturated fatty acids such as C20:2?6, C20:4?6, C20:5?3 and C22:2?6 were confirmed by gas chromatography-mass spectrometry. The percentages of these components in lipid fractions were compared with each other. In the percentages, C16:0, C18:1?9 and C18:2?6 acids were the most abundant fatty acids in the freshwater snail. Some differences were found in the percentages of phospholipid, neutral, total and algae lipids.
