Investigation of Biopharmaceutical Properties of Some Taxa Belonging to Meliaceae and Phyllantaceae Family

Özet
Bu çalışmada, doğal kökenli yeni bir farmasötik bileşenin belirlenmesi amacıyla Meliaceae
(Carapa procera ve Pseudocedrela kotschyi) ve Phyllantaceae (Phyllanthus reticulatus ve Uapaca
togoensis) familyalarına ait dört türün yaprak ve gövde kabuğu kısımları incelenmiştir. Ekstraktların (su
ve methanol) toplam biyoaktif içerikleri, antioksidan aktiviteleri, enzim inhibitör özellikleri (diyabet,
nörodejenerasyon ve cilt bozukluğu ile ilgili iki enzim ile ilişkili) ve antimikrobiyal aktivitelerinin tümü
incelendi (sekiz mantar ve bakteri suşu). Daha sonra her bitkinin biyoaktif içeriği ve biyoaktivitesi, çok
değişkenli bir analiz kullanılarak, bitki bölümlerinin ve ekstraksiyon çözücülerinin bir etkisi olup
olmadığını görmek için incelendi. Önemli derecede, P. reticulatus yapraklarının metanol ekstraktı, diğer
özütler arasında en yüksek flavanol konsantrasyonu sergiledi. Buna karşılık en düşük toplam fenolik
içerik ise, P. reticulatus’un metanolik yaprak ekstraktı ve U. togoensis’in su ve metanolik yaprak
ekstraktlarından elde edilmiştir. Ayrıca, test edilen özler geniş etkili antibakteriyel özellikler sergilemiştir;
Örneğin, U. togoensis yapraklarının su özütü dikkat çekici bir anti- Trichoderma viride, antiMicrococcus flavus, anti- Listeria monocytogenes, anti- Escherichia coli, anti- Salmonella typhimurium
ve anti- Enterobacter cloacae aktivitesi gösterdi. P. reticulatus’un yapraklarının su ekstraktı güçlü antiPenicillium verrucosum var. cyclopium aktivitesi sergiledi. P. kotschyi yapraklaının metanol ekstraktı
mükemmel anti- Salmonella typhimurium ve anti- Enterobacter cloacae etkisi gösterdi. Genel olarak, tüm
türler arasında en yüksek antioksidan ve enzim inhibitör aktiviteleri metanolik gövde kabuğu ekstraktı ile
elde edilmiştir. Sonuç olarak, değerlendirilen tüm parametreler çalışma kapsamında kullanılan tüm
bitkilerin farklı kısımlarına ve ekstraksiyon çözücüsünün polaritesine bağlı olarak değişkenlik
göstermiştir.
In this study, to identify a new pharmaceutical ingredient of natural origin, we investigated the leaves and stem bark parts of four species belonging of Meliaceae (Carapa procera and Pseudocedrela kotschyi) and Phyllantaceae (Phyllanthus reticulatus and Uapaca togoensis) family. The total bioactive content, antioxidant activity, enzyme inhibitory property (associated to diabetes, neurodegeneration, and skin disorder-related two enzymes) and antimicrobial activity (eight fungal and bacterial strain respectively) of the extracts (water and methanol) were examined. The bioactive content and bioactivity of each plant were then examined using a supervised multivariate analysis to see if plant sections and extraction solvents had an impact. Water leaf extracts of P. reticulatus and P. kotschyi, as well as methanol leaf extracts of P. kotschyi, had a significant level of flavonoid content. Remarkably, methanol extract of P. reticulatus leaves exhibited the highest concentration of flavanol among other extracts. In contrast the lowest total phenolic content was obtained with methanol leaves of P. reticulatus and water and methanol leaves of U. togoensis. In general, the highest antioxidant and enzyme inhibitory activities were obtained with methanol stem bark extract of all species. Further, the tested extracts exhibited widespectrum antibacterial properties; For instance, water leaves extract of U. togoensis showed remarkable anti- Trichoderma viride, anti- Micrococcus flavus, anti- Listeria monocytogenes, anti- Escherichia coli, anti- Salmonella typhimurium and anti- Enterobacter cloacae activity, water leaves extract of P. reticulatus exhibited great anti-P. verrucosum var. cyclopium activity while methanol leaves extract of P. kotschyi showed excellent anti- Salmonella typhimurium and anti- Enterobacter cloacae effect. As a result, all evaluated parameters varied depending on the different parts of all plants used in the study and the polarity of the extraction solvent. In conclusion, the results suggest that C. procera, P. kotschyi, P. reticulatus, and U. togoeinsis may be a promising source of bioactive chemicals with potential pharmaceutical applications.
