Çalışmanın sonunda, yaşadıkları her dönemde insanoğlunun adak adamaya ihtiyaç duyduğu sonucuna ulaşılmış ve tarihleme yapılmaya çalışılmıştır.Öğe Antimicrobial Resistance and Plasmid Typing of Coagulase Positive Staphylococcus Species Isolated from Bovine Mastitis(Selçuk Üniversitesi Fen-Edebiyat Fakültesi, 2004) Açık, Leyla; Çelebi, Ayten; Arslan, Emine; Uçan, Uçkun SaitIn this study, a number of 50 Coagulase positive Staphylococcus isolates has been isolated and identified using conventional bacteriological methods from bovine mastitis. Of the total isolates 40 was identified as Staphylococcus aureus and 10 was identified as Staphylococcus intermedius. This study indicates the increasing significance of S.intermedius as the causative agents of bovine subclinical mastitis. All the Staphylococcus strains were examined for their susceptibilities to antibiotics and plasmid profiles. Plasmid profiling demonstrated that 38 of 40 S.aureus and 9 of 10 S.intermedius isolates contained plasmid. Molecular weight of plasmids varied from 1.1 to 19 kb. The highest resistance to chematerapotics used was observed to penicillin. This study suggests that plasmid profile analysis does not appear to be a suitable method for typing S.aureus and S.intermedius.Öğe Antioxidant, Antimicrobial Activity And Dna Protective Effect Of Mespilus Germanica (L)(Selçuk Üniversitesi, 2023 Eylül) Taner Saraçoğlu, Hatice; Arslan, Nursena; Arslan, Emine; Zengin, Gökhan; Gülbahçe Mutlu, ElifAim: Medlar (M. germanica) is a durable plant which have hard fruits. In this study, it was aimed to determine the antioxidant, antimicrobial activity and DNA protective effects of both fruit and seed extracts of medlar. Method: Antioxidant capacities of extracts of medlar was determined by using the DPPH method, Folin-Ciocaltaeu method was used for total phenolic content and the antimicrobial effect of the extract against 16 bacteria was investigated. Findings: As a result of the study, the possible protective effect of medlar fruit extracts obtained at different concentrations against DNA damage was determined. In addition, it was revealed that the total phenolic contents and radical scavenging activities of the seed extracts were higher than the fruits extracts. While the medlar seed extract used in the study did not show any effect against bacteria, the fruit extract of this plant had various effects on the bacteria used. In addition, it was determined that the medlar fruit extract had a protective effect in DNA+UV, DNA+H2O2 and DNA+UV+H2O2 applications. Results: According to the results obtained, it was observed that M. germanica could provide protection against oxidative stress.Öğe The application of ISSR-PCR to determine the genetic relationship and genetic diversity between narrow leaved bluegrass (Poa angustifolia) and rough bluegrass (Poa trivialis) accessions(TUBITAK SCIENTIFIC & TECHNICAL RESEARCH COUNCIL TURKEY, 2011) Arslan, Emine; Tamkoc, AhmetThe present study aimed to determine the genetic relationship between Poa angustifolia and Poa trivialis as well as intraspecies genetic diversity using inter simple sequence repeat (ISSR) markers. ISSR amplification was evaluated for its applicability to narrow leaved bluegrass and rough bluegrass accessions identification. A total of 401 bands, 363 of which were polymorphic (90.52%), were generated using selected 20 primers. Genetic similarity between accessions belonging to Poa species was estimated using Nei's genetic similarity and UPGMA (unweighted pair group method with arithmetic) cluster analysis was used to construct a dendrogram. UBC836 and M6 primers were observed to be the most suitable for distinguishing between species. Because interspecies differences are especially important in plant breeding, M5 and UBC812 primers were ideal in distinction of P. trivialis and P angustifolia accessions, respectively. Primer UBC856 was determined to be ideal in discrimination both intraspecies and interspecies. The ISSR fingerprinting technique used was confirmed to be a reproducible and sensitive tool for the identification of Poa accessions and for determination of the correct starting material for plant breeding.Öğe Association of Polymorphisms in TCF7L2 Gene with Gastric Cancer Risk: A Preliminary Study in Turkish(DERMAN MEDICAL PUBL, 2015) Kaya, Dudu Erkoc; Arikoglu, Hilal; Sumen, Incilay Celik; Avci, Ebru; Ata, Ozlen; Arslan, EmineAim: The Transcription factor 7-like 2 (TCF7L2) gene, located on chromosome 10q25.3, encodes a transcription factor, which contains a high mobility group box, demonstrated in association with many cancer types. This factor is a critical part of Wnt/beta-catenin signaling pathway that plays key roles in regulation of embryonic development and homeostasis in mature tissues. It is known that the constant activation of Wnt/beta-catenin signaling pathway can cause cancer development. In this study, it is aimed to reveal the association between rs7903146C>T and rs12255372G/T single nucleotide polymorphisms in TCF7L2 gene, the key effector of Wnt/beta-catenin signaling pathway, and gastric cancer and to determine associated risk alleles. Material and Method: In our study, polymorphisms in TCF7L2 gene were genotyped using PCR-PFLP technique in 38 patients with gastric cancer and 48 healthy individuals. The obtained data were statistically analyzed and p<0.06 was accepted significant in all assessments. Results: No significant association was determined between rs7903146C>T and disease (p> 0.05) while 122553720>T polymorphism was associated with the disease under additive model (OR: 0.366 195% Cl: 0.135-0.989] p=0.047). Discussion: This is the first study to examine the association between TCF7L2 gene and gastric cancer risk in Turkish population and suggests that rs12255372C>T could be a potential indicator for gastric cancer. On the other hand, further studies are required which will be carry out in more increased number of samples in a wider population.Öğe Biological activities and DNA interactions of Amanita ovoidea(TAYLOR & FRANCIS LTD, 2015) Dogan, Hasan Huseyin; Arslan, EmineContext: Amanita ovoidea (Bull.) Link (Amanitaceae) is a well-known species due to its pleasant aroma and flavor since ancient times in the worldwide. This species is also known in Turkey and people consume it extensively. Objective: To evaluate medicinal importance of A. ovoidea for human health, to explain the effect of mushroom extracts on bacterial DNA, and to find preventive role on bacterial disease. Materials and methods: Chloroform, acetone, and methanol extracts of A. ovoidea were tested for the antimicrobial activities against four Gram-positive bacteria, five Gram-negative bacteria, and yeast using a micro-dilution method. In addition, DNA binding, DNA cleavage activity, and restriction enzyme digestion of the methanol extract of A. ovoidea were examined at different concentrations (40.000-78.125 mu g/mL). Results: The highest minimum inhibitory concentration (MIC) value observed against the test micro-organisms was with the chloroform extract (MIC 19.5 mg/mL concentration) against Candida albicans. Other highest antimicrobial effects observed against the test micro-organisms were with the methanol extracts against Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Streptococcus pyogenes, Candida albicans, Klebsiella pneumoniae, Proteus vulgaris, and Salmonella enteritidis (MICs, 78 mu g/mL concentrations). All concentrations reduced the mobility of plasmid DNA. BamHI and HindIII targeted specially to supercoils and cut them. Amanita ovoidea extract prevented cutting with HindIII by binding especially to the AA region in open circular DNA. Discussion and conclusion: Present results demonstrated that A. ovoidea has excellent antimicrobial and antifungal activities by its DNA interaction activity on pBR322.Öğe C-banded karyotype and nucleolar organizer regions (nors) of marsh frog, rana ridibunda (ranidae: Anura) in central anatolia(KAFKAS UNIV, VETERINER FAKULTESI DERGISI, 2010) Arslan, Emine; Arslan, Atilla; Gulbahce, ElifKaryotype, C-banding and nucleolar organizer regions (NORs) characteristics of nine Rana ridibunda samples collected from Konya (Meram, Beysehir, Hadim) province were examined. The diploid number of chromosomes (2n) and the fundamental number of chromosome arms (FN) were determined as 26 and 52, respectively. All of the chromosomes of this species have centromeric constitutive heterochromatin (C-band). Nucleolar organizer regions (secondary constriction) were determined on the long arm of the no. 8 submetacentric chromosome by using silver-nitrate staining technique.Öğe C-heterochromatin and NORs distribution in karyotypes of three vespertilionid bat species from Turkey(WALTER DE GRUYTER GMBH, 2015) Arslan, Atilla; Zima, Jan; Albayrak, İrfan; Yorulmaz, Tarkan; Arslan, EmineThe chromosomal banding analysis of the karyotypes of Turkish populations of Eptesicus serotinus, Nyctalus lasiopterus and Barbastellus barbastellus was performed with the use of C-banding and Ag-NOR staining. The results obtained in E. serotinus and N. lasiopterus were congruent with previous data reported from other regions. The karyotype of E. serotinus (2n = 50, NF = 52) contained a moderate amount of centromeric C-heterochromatin and a single NOR was localized in an acrocentric autosomal pairs. The karyotype of N. lasiopterus (2n = 42, NF = 54) contained a higher amount of centromeric C-heterochromatin and the NORs were localized in two autosomal pairs. The karyotype of B. barbastellus was standard in its general characteristics (2n = 32, NF = 54, low amount of C-heterochromatin) but the NOR was localized in only one acrocentric autosomal pair. In studies from other regions, the NORs were recognized in all five acrocentric autosomal pairs of the complement of B. barbastellus.Öğe Characterisation of coagulase positive Staphylococcus species isolated from bovine mastitis using protein and plasmid patterns(SCIENTIFIC TECHNICAL RESEARCH COUNCIL TURKEY-TUBITAK, 2009) Arslan, Emine; Çelebi, Ayten; Açık, Leyla; Uçan, Uçkun SaitThe objective of this work was to study the protein patterns, plasmid profiles, and antibiotic susceptibility of Staphylococcus intermedius and Staphylococcus aureus isolates originating from mastitic mammary glands of dairy cattle in different parts of Konya province. A total of 114 Staphylococcus species were isolated and identified by conventional bacteriological methods from bovine mastitis. Of the total isolates 77 were identified as S. aureus and 37 as S. intermedius. Intra- and inter-species diversities in the coagulase-positive staphylococci were investigated by analysis of whole-cell protein profiles using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Plasmid profiling also demonstrated that 75 S. aureus isolates and 36 S. intermedius isolates contained plasmid. In addition, 88.3% of S. aureus and 59.4% of S. intermedius isolates were resistant to penicillin. Sixty-six of the 77 S. aureus isolates were also resistant to amoxicillin+clavulanic acid (85.7%). The corresponding number for S. intermedius was 17 (45.9%). Only 1 S. aureus isolate was resistant to danofloxacin. One of each of the Staphylococcus isolates was resistant to methicillin. Results from the study showed that the susceptibility of S. intermedius isolates to antibiotics used widely in mastitis therapy is a matter of concern.Öğe Characterization of Consolida Sf Gray (Ranunculaceae) Taxa in Turkey by Seed Storage Protein Electrophoresis(Walter De Gruyter Gmbh, 2010) Ertuğrul, Kuddisi; Arslan, Emine; Tugay, OsmanObjective: This research aimed to investigate the genetic relations of Consolida S. F. Gray species collected in Turkey using seed storage proteins. This study has become significant as the taxonomic status of genus Consolida changes often. Methods: Mature seeds of 22 Consolida species were collected. Total protein were isolated from seeds collected for each species. Sodium Dodecyl Sulphate-Polycarylamide Gel Electrophoresis was performed by a standard method on a vertical slab gel. After electrophoresis, the protein bands were visualized by staining with Coomassie Brillant Blue G-250.The polymorphic bands were scored visually as present (1) or absent (0). Genetic similarity among taxa was estimated basing on Nei's homology. Results: In all, 74 polypeptide bands of different sizes ranging from 25 to 140 kDa were observed in the 22 taxa of the genus Consolida. Cluster analysis was performed using the unweighted pair-group method and arithmetic averages in Bio1D++ computer program. According to similarity coefficients, the genus Consolida was separated into the main two groups. It was discovered that the first group consisted of the species of Consolida with 5-lobed corolla, and in the second main group were the species with 3-lobed corolla. Conclusion: It is concluded that seed storage protein profiles could be useful markers in the studies of genetic relations of Consolida species. The grouping as a result of the dendogram has also supported the relationship in Flora of Turkey.Öğe CHARACTERIZATION OF UROPATHOGENIC ESCHERICHIA COLI STRAINS: ANTIBIOTIC RESISTANCE PATTERNS, DETECTION OF ESBL GENES AND INTERACTIONS BY LYTIC PHAGES(PARLAR SCIENTIFIC PUBLICATIONS (P S P), 2018) Uysal, Ahmet; Gunes, Erdogan; Arslan, Emine; Durak, YusufIn this study, 97 Escherichia coli strains isolated from urinary tract infections were evaluated in terms of phenotypic diversity, susceptibility to sixteen antibiotics, and extended spectrum of beta lactamase (ESBL) characteristics. The bacteriophage interactions was used to identify the relations of the E.coli isolates. Results showed that 21 of the 76 typable strains revealed 19 different phage types. Phage types of 21 strains could not be determined. Antibiotic susceptibility tests, performed against sixteen antibiotics, showed that although the isolates revealed 44 different types of resistance profiles, the strains showed the greatest resistance to cephalothin (54.6%), followed by tetracycline (53.6%), nalidixic acid (44.3%), and aztreonam and ofloxacin (29.8%). There was no resistance to amikacin and meropenem, and all strains were susceptible to meropenem. ESBL detection conducted by double disc synergy and multiplex polymerase chain reaction methods showed that 13 of 97 (13.4%) strains were ESBL-producing. Of the 13 ESBL-producing isolates, two carried bla(CTX-M), six bla(TEM), five bla(OXA), and one bla(TEM) and bla(SHV) at same time. We concluded that lytic phages could not be successful for typing the strains phenotypically. The monitoring of antimicrobial resistance levels and antimicrobial usage in humans should be an integral part of the prevention and control of antimicrobial resistance.Öğe Determination of Some Metal Levels in Muscle Tissue of Nine Fish Species from Beysehir Lake, Turkey(CENTRAL FISHERIES RESEARCH INST, 2012) Ozparlak, Haluk; Arslan, Gulsin; Arslan, EmineConcentrations of some metals were determined in muscle tissues of nine fish species from Beysehir Lake, Turkey by using atomic absorption spectrophotometer. These cytotypes are largely indistinguishable morphologically, are mainly allopatric and their taxonomic ranking is contradictory. We established a cytochrome b phylogeny for 15 cytotypes belonging to all three species recognized on morphological grounds (morphospecies): N. leucodon, N. xanthodon and N. ehrenbergi. Phylogenetic reconstructions yielded two highly divergent groups which are in agreement with the current division into two subgenera (Nannospalax and Mesospalax). The former comprised samples from south-eastern Turkey, Israel and Egypt (the morphospecies N. ehrenbergi). Basal dichotomy within Mesospalax remained unresolved and the putative sister position of N. leucodon against the two lineages of N. xanthodon was not supported in our analysis. Net divergences between sister cytotypes were low (< 2.0%) and two N. leucodon cytotypes were not even reciprocally monophyletic. Among the three morphospecies, the genetic diversity was lowest in N. leucodon (2.4% +/- 0.3%), highest in N. xanthodon (8.8% +/- 0.7%) and intermediate in N. ehrenbergi (5.0% +/- 0.5%). Our results show that associations between genetic and chromosomal variation are not widespread and common in mole rats, and therefore refute the generalization of a cytotype-equals-species approach. The variability of seed storage proteins was analysed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). As many as 72 bands were scored and a dendrogram was constructed using UPGMA (unweighted pair group method with arithmetic mean). The dendrogram of the electrophoretic protein profiles of seeds showed 2 main clusters. While the first cluster with further subclusters included Onobrychis species, the second cluster included Sartoria and Hedysarum species present in 2 separate subclusters. The results showed that Sartoria and Hedysarum are closer to each other than they are to Onobrychis. It is also suggested that Sartoria hedysaroides should be included in the genus Hedysarum. Additionally, it is concluded that seed storage protein profiles could be useful markers in studies of genetic diversity, genetic relationships, and classification of adapted cultivars.Öğe Genotyping of Staphylococcus aureus Strains Isolated from Bovine Mastitis in Turkey by using ERIC-PCR Method(ZOOLOGICAL SOC PAKISTAN, 2016) Arslan, Emine; Mutlu, Elif GulbahceMastitis is an intramammary infection that leads to important economic losses in dairy industry such as increasing the amount of waste milk by affecting the quality and quantity of the milk, removing cows from herd or sending them for slaughter early. Staphylococcus aureus has an important place in the fight against mastitis. In this study, it is aimed to genotyping of the 98 S. aureus isolates that were isolated from bovine mastitis by the polymerase chain reaction of enterobacterial repetitive intergenic consensus (ERIC-PCR). ERIC primers generated DNA products in sizes ranging between similar to 8000 bp and 250 bp. According to the Nei homology, a dendrograme was obtained by the assessment of 28 bands in total. ERIC-PCR grouped 98 S. aureus isolates into this clusters (I-VI) showing 64 ERIC genotypes. This method, which enables the S. aureus strains to be described and classified successfully, could be used in forming a database by formulation of genotypes, determining the origin of the epidemic, studies of vaccine development and developing effective protection strategies against mastitis since it is easy, fast and reliable. At the same time, this study would contribute to the comprehension of S. aureus epidemiology and ecology in dairy herds.
