Koyunlarında Bazı Kan Parametreleri?ni?n Araştırılması(Selçuk Üniversitesi, 1997 Ocak) Bildik, Ayşegül; Yur, Fatmagül; Belge, Ferda; Değer, Yeter; Dede, SemihaIn this study, 15 Hamdani sheep and 4 rams belong to Research Center of University of Yüzüncü Yıl were used as research material. Using spectrophotometric tecnique the whole blood sample and plasma were analysed for the determination of the levels of glutathione, ceruloplasmin, Cu, Ca and P. In addition, haematocrit was determinated by using microhaematocrit technique. Animals having over 25mg/dl glutathione were designated as high glutathione type (GSHH). Values of GSHH, GSHh, ceruloplasmin, Cu, Ca, P and haematocrit in Hamdani sheep were 30.8 1.7 mg/dl, 11.88 ± 2.2 mg/dl, 12.68 ± 0.87 mg/dl, 75 +23 (g/dl, 6.71 ± 0.4 mg/dl, 14.74 ± 0.63 mg/dl, % 33 10.37, respectively. These values in Hamdani rams also were 32.8 mg/dl, 22 ± 0.25 mg/dl, 12 ± 2.9 mg/dl, 91 ± 40 (g/udl, 9.45 ± 2.1 mg/dl, 5.69 ± 1.7 mg/dl and % 36.7 ± 1.3, respectively. Statisticaly positive correlation was found between Cu and ceruloplasmin concentration of plasma (p<0.05).Öğe In vitro evaluation of glutathione implementation on oxidative DNA damage and oxidant status in high glucose conditions(Selçuk Üniversitesi, 2022) Yur, Fatmagül; Dede, Semiha; Çetin, Sedat; Taşpınar, Mehmet; Usta, AyşeAim: This study aimed to show the effects of glutathione, recognized by its antioxidant specialties, on the potential DNA damage (8-hydroxy-2- deoxyguanosine) and the antioxidant system changes upon its implementation in BHK-21 cells cultured with high glucose. Materials and Methods: BHK-21 cell line was regularly surpassed in vitro conditions (5% FBS, 10% horse serum, 1% L-Glutamine, 1% penicillin/ streptomycin in RPMI 1640 medium, and 5% CO2 and 95% humidity and 37oC) incubated. The control group determined glucose's IC50 value based on the viability tests executed on MTT cells. Cells were seeded in plates as each would have 2x106 cells. The control, the test, and the crossbreed test(glucose; (285 mM), glutathione (250 µM)) groups were prepared. After 24 hours of incubation, trypsinized cells were designed for analysis through vitrification. In the lysate of the cell culture that was procured, Oxidative DNA damage, TAS, TSO, and OSI were measured by the spectrophotometric system with ELISA. Results: It was observed that 8-OHdG levels increased significantly with glucose application. Moreover, the increase in the HG+GSH group was more significant when compared to the control group (p?0.05). No difference with the control group was found only in the groupwhereGSHwas applied.As forTAS,whereas any differencewas observedinGSHusedgroups,theincreaseintheHG+GSHgroupwassignificantcompared to the control group (p?0.05). that were the same as the control group. TOS and OSI considerablyincreasedinHG+GSHimplementedgroupsastothecontrolgroup(p?0.05). Conclusion: According to the results, no protective impacts of glutathione at the cellular level in the doses mentioned above were observed on high-dose glucose implemented cells. On the other hand, it was revealed that the applied amounts of glutathione in the process did not cause any toxic effects.Öğe Suplemental Olarak Çi?nko Uygulanan Tavuklarda Ni?tri?k Oksi?t Oksi?dasyon Ürünleri? ve Antioksidan Vitamin Düzeyleri?(Selçuk Üniversitesi, 2001 Mart) Dede, Semiha; Değer, Yeter; Kahraman, Tahir; Dönmez, NurcanThe aim of this study is to determine of the effect of supplemental zinc on the concentration of the oxidation products of nitric oxide and antioxidant vitamins in chicken serum. Chicken were divided into three experimental groups and one control group, each group containing seven chicken. The experimental groups were treated supplement zinc (125, 500, 1000 ppm). The concentration of nitric oxide oxidation products and antioxidant vitamins in the serum of zinc treated chicken were analysed with spectrophotometrically. Nitrate (ppm), nitrite (ppm). vitamin E (mg/dl), vitamin C (mg/dl) concentrations of experimental groups 1., 2., 3. and control group were found; 6.16, 6.41, 13.16, 7.38, 1.09, 1.20, 1.30, 1.78; 0.121, 0.326, 0.247, 0.313; 0.927, 1.682, 1.204, 1.323 respectively. Nitrite levels of control groups was found higher than experimental groups 1,and 2 (p<0.05). Nitrate levels were not signifficantly differentes between groups. The vitamin E concentration of 1 experimental group was lower than other groups (p<0.05). The vitamin C concentration of 2 experimental group was higher than 1 and 3 experimental groups (p<0.05). In conclusion; the concentration of nitric ide oxid products in serum of zinc I chicken were not differentes between groups, but the antioxidant vitamins levels were increased. These results support the idea that zinc has an important rol in the metabolism as an antioxidant trace element in chicken.Öğe Van Bölgesi Su Kaynaklarında Ağır Metal Kirlilik Düzeyleri(Selçuk Üniversitesi, 2007) Alemdar, Süleyman; Ağaoğlu, Sema; Alişarlı, Mustafa; Dede, SemihaBu çalışmada, Van merkez ve ilçelerinde (Erciş, Özalp, Saray, Muradiye, Çaldıran, Gürpınar, Gevaş ve Edremit) bulunan kuyu, dere, kaynak/çeşme, musluk ve depo sularında ağır metal (Fe, Cu, Zn, Mn, Cd, Cr, Ni ve Co) düzeyleri araştırıldı. Bu amaçla ilkbahar, yaz, sonbahar ve kış dönemlerinde 100 farklı kaynaktan sağlanan toplam 366 örnek materyal olarak kullanıldı. Araştırma bulgularına göre; incelenen su kaynaklarının tamamında Mn ve Cd değerleri, merkez ve ilçelerdeki kuyu ve kaynak/çeşme sularında Fe, merkezdeki kuyu sularında ise Zn düzeyi İçme ve Kullanma Suları Standardi'nda bildirilen kriterlere uygun bulunmadı. Ayrıca, merkez ve ilçelerdeki kaynak/çeşme sularında Fe, Mn, Cd ve Ni; musluk sularında Mn, Cd ve Ni; depo sularında Mn ve Cd, ilçelerdeki depo sularında ise Ni düzeyi ilgili yönetmelikte bildirilen değerlere uygunluk göstermedi.Öğe Van Bölgesi? Bu amaçla ilkbahar, yaz, sonbahar ve kış dönemlerinde 100 farklı kaynaktan sağlanan toplam 366 örnek materyal olarak kullanıldı. Örneklerde pH, renk, bulanıklık, sıcaklık, toplam sertlik, sodyum, potasyum, kalsiyum, magnezyum, klorür, elektriksel iletkenlik, karbondioksit, karbonat, bikarbonat, sülfat, fosfat ve organik madde analizleri yapıldı. Araştırma bulgularına göre; merkez ve ilçelerdeki kaynak/çeşme suları ile ilçelerdeki depo sularında belirlenen bulanıklık düzeyleri ilgili yönetmelikte bildirilen kriterlere uygun bulunmadı. Ayrıca, merkez ve ilçelerdeki kuyu sularında klorür, renk, bulanıklık, elektriksel iletkenlik ve sülfat, kaynak/çeşme sularında renk ve sülfat, derelerde sertlik; merkezdeki derelerde magnezyum ve renk, kaynak/çeşme sularında klorür ve bulanıklık, ilçelerdeki kaynak/çeşme sularında ise elektriksel iletkenlik düzeyi İçme ve Kullanma Sulan Standardi'nda bildirilen sınır değerlerden daha yüksek bulundu. İncelenen su kaynaklarında tespit edilen karbondioksit düzeyleri su kalite kriterlerine uygun bulunmadı.
