Öğe Abomasum Deplasmanlı Sütçü Sığırlarda Bazı Antioksidan Vitamin Düzeyleri(2003) Irmak, Kemal; Haliloğlu, SeyfullahBu çalışmada, abomasum deplasmanlı sığırlarda serum vitamin C, P-karoten ve vitamin A düzeyleri araştırılmıştır. Bu araştırmada, 3- 8 yaşlı, erken laktasyonda (7 - 12 gün), Sağa Abomasumun Deplasmanlı 10 (RDA'lı grup), Sola Abomasumun Deplasmanlı 10 (LDA'lı grup) ve kinik olarak sağlıklı 10 (Kontrol Grubu) olmak üzere toplam 30 adet Holstein sütçü sığır materyal olarak kullanıldı. Abomasumun Deplasmanlı (AD'li grup) hayvanlar kliniğe getirildiklerinde ortalama 3 günlük hastaydılar ve cerrahi operasyonundan sonra 6 adedi öldü. Kontrol grubuna göre; Vitamin C, AD'li grupta önemli düzeyde (p0.05) yüksek; Vitamin A ise, RDA, LDA ve AD'li gruplarda önemli düzeylerde (sırasıyla p0.05, p0.01, p0.01) düşük olarak tespit edildi. Furthenııore, phagocytic and microbicidal activity of PMNl were not statistically changed by these combinations.Öğe Broylerlerde Yeme İlave Edilen Sodyum Aliminosilikat (Zeolit) Bileşiğinin Kan Plazması Mineral Kompozisyonuna Etkisi(1998) Kurtoğlu, Firuze; Başpınar, Nuri; Haliloğlu, SeyfullahAraştırma, broylerlerde yeme farklı düzeylerde Zeolit ilavesinin kan plazması Ca, Pi, Na, K, Cl ve Zn değerlerine etkisinin tespiti amacı ile yapıldı. 51 gün süren denemede Peterson x Avian hibrit broyler civcivler yemlerine katılan Zeolit miktarına bağlı olarak 5 ayrı gruba ayrıldılar. Deneme süresince hayvanlar broyler besleme programına uygun olarak 1-21, 22-41, 42-51. günler arasında 3 farklı rasyonla beslendiler. Deneme, sonunda her bir gruptan 8 er hayvanın kanları heparinize tüplere alınarak 2500 rpm de 4 C de 10 dakika santrifüj edildi ve plazmalar elde edildi. Kan plazması Ca, Pi, Na, Cl, K ve Zn değerleri yönünden kontrol ve deneme grupları arasında karşılaştırılarak değerlendirildi. Sonuç olarak aliminosilikatların Ca ve P değerleri başta olmak üzere mineral ve iz elementler üzerinde farklı etkiler ortaya çıkardığı ve bu etkilerin hayvanların verim özelliklerini etkileyebileceği sonucuna varılmıştır.Öğe Development of an Enzyme Immunoassay for the Determination of Ovine Growth Hormone in Plasma(Scientific Technical Research Council Turkey, 2000) Serpek, Behiç; Haliloğlu, SeyfullahAbstract: A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 µl of plasma and the calibration graph (90% relative binding at 0.38±0.02 ng ml and 50% relative binding at 3.80±0.18 ng ml) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 100.03%, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, OLH, OTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.Öğe Effect of Dietary Vitamin A and Zinc on Egg Yield and Some Blood Parameters of Laying Hens(SCIENTIFIC TECHNICAL RESEARCH COUNCIL TURKEY, 2001) Kaya, Şule; Umucalılar, H. Derya; Haliloğlu, Seyfullah; İpek, HüdaiThe purpose of the present study was to examine the effects of vitamin A and zinc interaction in laying hens fed on diets supplemented with two different levels of vitamin A (0, 10.000 IU/kg) in combination with five different levels of zinc (0, 25, 50, 100, 200 mg/kg) on egg yield, plasma and yolk concentration of zinc, and the levels of plasma triglyceride and phosphorus for 12 weeks. Egg production, egg weight, feed intake, body weight and feed efficiency did not differ among the groups. Plasma and yolk zinc concentrations were affected only by zinc supplementation. An interaction between vitamin A and zinc had an effect and the independent effect of vitamin A supplementation increased the plasma triglyceride levels of laying hens. The level of phosphorus in plasma was altered by zinc supplementation and an interaction between zinc and vitamin A. Since the positive relationship between plasma zinc and egg production was observed (r=0.279 p<0.0 1), it is suggested that plasma zinc may be an indicator for the estimation of egg production or hen performance.Öğe The effect of endodontic therapy on periapical exudate neutrophil elastase and prostaglandin-E-2 levels(ELSEVIER SCIENCE INC, 2005) Alptekin, Nilgün Özlem; Arı, Hale; Haliloğlu, Seyfullah; Alptekin, Tuncay; Serpek, Behiç; Ataoğlu, TamerThis study aimed to investigate periapical exudate neutrophil elastase (NE) and prostaglandin E-2 (PGE(2)) levels and their relationships with clinical symptoms, and to determine the changes of their levels following first treatment visit. Periapical exudate samples were collected from the canals of 34 nonvital single-rooted teeth at two sequential treatment visits. Periapical exudate NE and PGE(2) levels were found to be higher in the presence of clinical symptoms (pus discharge, swelling) (p < 0.05). The canals of teeth with larger periapical radiolucent area (>= 1 cm) contained more PGE(2) levels than with smaller ones (< 1 cm) (p < 0.05). Periapical exudate NE levels were significantly correlated with PGE2 levels (p < 0.05), and their levels at first treatment visit did not change following root canal therapy (p > 0.05). The periapical exudate NE and PGE(2) levels may regulate periapical disease expression, but the results of this study were unable to reveal this association.Öğe The effects of alpha-tocopherol on serum Il-1, Il-4 and Il-6 levels of rats with experimental periodontitis and type I diabetes(2013) Hatipoğlu, Mükerrem; Alptekin, Nilgün Özlem; Haliloğlu, SeyfullahAmaç: Epidemiyolojik ve klinik çalışmalar periodontal hastalığın diyabeti etkileyebileceğini göstermiştir, diğer taraftan periodontitis diyabetin komplikasyonlarından birisidir. Bu çalışmanın hedefi, deneysel periodontitis ve streptozotosin (STZ) ile diyabet oluşturulan/ oluşturulmayan sıçanlarda -tokoferol uygulamasının serum sitokin düzeyleri üzerine etkilerini araştırmaktır. Gereç ve Yöntemler: Toplam kırk adet erkek Sprague Dawley sıçana anestezi uygulandı ve mandi- bular sağ birinci molar dişlere subgingival olarak 3/0 ipek sütur bağlandı. Bu hayvanlar, sağlıklı grup (Grup I) ve tek seferde 50mg/kg STZ enjekte edilen diyabe- tik grup (Grup II) olmak üzere iki gruba ayrıldı. Bu gruplarda serum fizyolojik (Grup IA, Grup IIA) ve - tokoferol (40mg/kg/gün) enjekte edilen alt gruplara (Grup IB, Grup IIB) ayrıldı. Ligatür bağlandıktan 21 gün sonra sıçanlar kurban edildi. Aynı zaman aralı- ğında serum IL-1, IL-4 ve IL-6 konsantrasyonlarının (pg/ml) ticari ELISA kitleri kullanılarak ölçülebilmesi için kan örnekleri alındı. İstatistiksel kıyaslamalar için iki-yönlü varyans analizi kullanıldı p0.05 anlamlılık düzeyi kabul edildi. Bulgular: Verilerin analizi sonunda, gruplar arasında serum IL-1, IL-4 ve IL-6 konsantrasyonları istatistik- sel olarak farklı bulunmadı (p0.05). Sonuçlar: Elde edilen sonuçlara göre -tokoferol’ün deneysel periodontitisli, STZ ile diyabet oluşturulmuş/oluşturulmamış sıçanlarda serum IL-1, IL-4 ve IL-6 konsantrasyonlarını değiştirmediği belirlenmiştir.Öğe Effects of Defaunation and Dietary Nitrogen Source on Sodium, Potassium, Iron and Zinc in the Rumen Fluid, Plasma and Wool of Lambs(Scientific Technical Research Council Turkey-Tubitak, 2006) Özdemir, Mehmet; Çınar, Miyase; Haliloğlu, Seyfullah; Eryavuz, AbdullahThis study was conducted to investigate the effects of defaunation, and the addition of urea and sulfur in the replacement of plant protein (PP) sources on concentrations of Na, K, Zn and Fe in the ruminal fluid, blood plasma and wool of lambs. Forty male Ramlic lambs, 2.70 +/- 0.30 months of age, and weighing approximately 16.44 +/- 0.41 kg, were used. Lambs were divided into 4 groups as follows: faunate + control diet (FC), defaunate + control diet (DC), faunate + experimental diet (FE), and defaunate + experimental diet (DE). The control diet contained plant protein as a N source, whereas the experimental diet was supplemented with urea and sulfur, both at 3% of BW per day. The forage portion of the diet was 350 g of alfalfa hay per lamb per day. Diets were fed twice daily in equal amounts. Defaunation, or feeding a diet supplemented with urea and sulfur in place of plant protein sources, had no effect on ruminal fluid Na concentrations but there was an interaction on Na concentration in the rumen fluid. Ruminal K concentration was lower (P < 0.021) in the experimental diet versus the control diet, while it was not affected by defaunation. There was an interaction effect on Zn concentration in rumen fluid. Plasma Na, K and Zn concentrations were not affected by treatments, whereas defaunation significantly decreased (P < 0.048) plasma Fe concentration. Defaunation or N source had no effect on Na, K, Zn and Fe concentrations in wool. We concluded that both defaunation and feeding a diet supplemented with urea and sulfur in place of plant protein sources did not affect blood plasma, except for Fe, and wool Na, K, Zn and Fe concentrations despite differences in their ruminal concentration.Öğe Effects of testosterone undecanoate treatment on serum biochemical parameters in New Zealand rabbits(Selçuk Üniversitesi, 2024) Alan, Beyza Suvarıklı; Camgöz, Avni; Dayan, Mustafa Orhun; Haliloğlu, Seyfullah; Karakaya, NidaAmaç: Çeşitli hastalıkları tedavi etmek için kullanılan anabolik androjenik steroidler (AAS) aynı zamanda sporcuların sportif performansın iyileştirilmesi amacıyla kullandıkları doping maddeleridir. Bu çalışmanın amacı, uzun süreli Testosteron Undecanoate (TU) uygulanan Yeni Zelanda tavşanlarında bazı serum biyokimyasal parametrelere etkilerini değerlendirmektir. Gereç ve Yöntem: Sunulan çalışmada 5-6 haftalık 21 adet erkek Yeni Zelanda tavşanı, Kontrol, Hint yağı ve Hint yağında çözündürülen TU grubu olmak üzere 3 gruba ayrıldı. Hint yağı ve Hint yağında çözündürülmüş TU (10 mg/kg) haftada 5 gün olmak üzere 6 hafta boyunca 0.2 ml olarak ilgili gruplara kas içi olarak uygulandı. Bulgular: Toplam 22 biyokimyasal parametrenin değerlendirildiği çalışmanın sonuçları, aspartat aminotransferaz, amilaz, trigliserit, kolesterol, total protein, kreatinin, ürik asit, fosfat, potasyum, sodyum, kalsiyum ve glukoz düzeylerinin TU grubunda kontrol ve hint yağı gruplarına göre istatistiksel olarak anlamlı derecede yüksek olduğunu ve yüksek yoğunluklu lipoprotein düzeylerinin diğer gruplarda kontrol grubuna göre anlamlı derecede düşük olduğunu göstermiştir. Öneri: Sonuç olarak Yeni Zelanda tavşanlarında TU uygulamasının serum biyokimyasını olumsuz yönde etkileyebileceği ve farklı testosteron analogları ile multidisipliner çalışmaların yapılması gerektiği ifade edilebilir.Öğe Effects of Zinc and Vitamin a Supplements on Plasma Levels of Thyroid Hormones, Cholesterol, Glucose and Egg Yolk Cholesterol of Laying Hens(Elsevier Sci Ltd, 2001) Kaya, S.; Keçeci, Tufan; Haliloğlu, SeyfullahThe effects of zinc and vitamin A supplementation to the diet on some blood metabolites were evaluated in Hisex brown laying hens from 56 weeks to 68 weeks of age. A total of 130 birds were divided into two main groups according to vitamin A treatment (0 and 3.44 mg retinyl acetate kg(-1) feed, respectively). each consisting of 65 hens. Hens in both of the main groups were then, divided into five zinc treatment groups (0, 25, 50, 100 and 200 mg zinc kg diet(-1) respectively) of 13 hens each. It was observed that plasma T-4.T-3 and total cholesterol levels were affected by only zinc supplementation. While 100 and 200 mg Zn kg(-1) decreased plasma T-4 level compared to control value. plasma T-3 level was reduced by 100 mg Zn kg(-1) compared to groups fed less Zn. Adding 50 and 200 mg Zn kg(-1) to the diet increased plasma total cholesterol level in the birds compared to other groups. Vitamin A, zinc. and their interaction did not influence the concentration of plasma high-density lipoprotein (HDL)-cholesterol, glucose and egg yolk cholesterol in laying hens.Öğe The Enzyme Activity of Alkaline Phosphatase in Gingival Crevicular Fluid of Smokers and Non-Smokers with Chronic Periodontitis(2006) Erdemir, Ebru; Duran, İsmet; Haliloğlu, SeyfullahAmaç: Bu çalışmanın amacı sigara kullanımının kronik periodontitisli hastalarda klinik parametreler ile DOS ALP enzim aktivitesi üzerine etkisinin değerlendirilmesidir. Gereç ve Yöntem: Araştırma, sigara içen 22 hasta, hiç sigara içmemiş 19 hasta olmak üzere toplam 41 gönüllü hasta üzerinde yürütüldü. Bütün hastalardan gingival indeks (Gİ), plak indeksi (Pİ), sondalama cep derinliği (SCD), klinik ataşman kaybı (KAK) ve sondalamada kanama (SK) dahil klinik parametreler ve alkalen fosfataz (ALP) enzim aktivitesi değerlendirmeleri için dişeti oluğu sıvısı (DOS) örneklemeleri yapıldı. Bütün ölçümler 3. ve 6. aylarda tekrarlandı. Bulgular: 3. ve 6. aylardaki ölçümlerde sigara içmeyen grupta, Gİ ve SK ortalama değerleri istatistiksel olarak anlamlı bir şekilde daha yüksek bulunmuştur (P0.05). DOS ALP total aktivitesinin sigara içmeyenlerde 6. ayda istatistiksel olarak anlamlı bir şekilde daha yüksek olduğu ve sigara içen grupta ise DOS ALP total aktivitesinin 6. aya doğru azaldığı görülmüştür. Sonuç: Bu araştırmada sigara kullanımının DOS ALP total seviyesini azalttığı sonucuna varılmıştır.Öğe Feeding Diets Supplemented With Zinc and Vitamin a in Laying Hens: Effects on Histopathological Findings and Tissue Mineral Contents(W B Saunders Co Ltd, 2002) Kaya, Şule; Ortatatlı, Mustafa; Haliloğlu, SeyfullahThe effects of different dietary levels of zinc and vitamin A on tissue mineral accumulation and histopathological alterations were examined in laying hens. For this purpose, 130 Hisex brown laying hens aged 56 weeks were divided into two main groups. Each group of hens were fed on diets supplemented with two different levels of vitamin A (010 000 IU kg(-1)). Then, laying hens in both of the main groups were subdivided into five treatment groups given different levels of zinc (0, 25, 50, 100, and 200 mg kg(-1)) for 12 weeks. Dietary additions of zinc and/or vitamin A did not significantly affect the zinc concentrations of the pancreas, liver, gizzard, thyroid, and copper concentrations of the pancreas, gizzard, and thyroid. The group fed the highest zinc (200 mg kg(-1)) in the diet had twofold iron concentration in their thyroid tissue compared to the concentration of the control group receiving no extra zinc. Histopathologically, there was slight oedema, degranulation, and vacuolation in exocrine cells and some fibrotic areas in the pancreas and reduced follicular size. Follicular cell hypertrophy and hyperplasia occurred in the thyroids, and gizzard erosions were seen in some cases.Öğe Gebe ve gebe olmayan holstein ırkı ineklerde plazma vitamin a ve beta-karoten düzeyleri(2003) Haliloğlu, Seyfullah; Semacan, Ahmet; Başpınar, Nuri; Doğruer, GökhanBu çalışma, tohumlama sonrası gebe ve gebe olmayan ineklerde, plazma p-karoten ve vitamin A düzeylerindeki değişimlerin belirlenmesi amacıyla, tek doğum yapmış, aynı bakım ve besleme şartlarındaki 20 adet Holstein ırkı inek üzerinde gerçekleştirildi. Vaginal, rektal ve ultrasonografik muayeneler yapılarak reprodüktif problemleri olmadığı belirlenen ineklerin östrüs senkronizasyonları yapıldı ve tohumlandılar. Tohumlama sonrası 0-23. günler arasında her bir inekten toplam 11 kez kan örnekleri alınarak, spektrofotometrik olarak plazma p-karoten ve vitamin A düzeyleri belirlendi. Tohumlama sonrası 25.günde ineklerin ultrasonografik gebelik muayeneleri yapılarak gebe (n11) ve gebe olmayanlar (n9) olmak üzere iki gruba ayrıldılar. Çalışmada gebe ineklerde p-karoten ve vitamin A düzeyleri arasında pozitif korelasyon gözlenirken, gebe olmayanlarda ilişkiye rastlanmamış, ayrıca gebe ve gebe olmayanlar arasında ortalama plazma p-karoten düzeyleri açısından anlamlı farklılıklar gözlenirken, vitamin A düzeylerinde fark bulanamamıştır. Sonuç olarak; özellikle kapalı ahır besisi yapılan ineklerde fertilite oranının artırılması amacıyla p-karotence zengin beslemenin yararlı olacağı kanısına varılmıştır.Öğe Holstein Irkı Gebe İneklerin Plaza ve Ovaryum Vitamin C, Progesteron ve 17 Beta-Östradiol Düzeyleri Arasındaki İlişkiler(2002) Haliloğlu, Seyfullah; Serpek, Behiç; Başpınar, Nuri; Erdem, Hüseyin; Bulut, ZaferThis study was performed to determine the relationship between the corpus luteum weight/diameter, follicular diameter and the levels of vitamin C, progesterone and oestradiol 17-b of the plasma, corpus luteum and follicular fluid of pregnant Holstein cows in a slaughterhouse. Progesterone and oestradiol 17-b concentrations were determined by the microtitration plate EIA method. Vitamin C levels were determined by spectrophotometer. Although there was a positive correlation between follicular fluid oestradiol 17-b levels and the follicle diameter in the follicular fluid, in contrast to oestradiol 17-b levels, small follicles have higher progesterone content than large follicles. This negative correlation was due to the fact that progesterone is the precursor of oestradiol 17-b. There was no correlation between corpus luteum progesterone levels and corpus luteum weight/diameter. However, a positive correlation between corpus luteum progesterone levels and the corpus luteum weight/diameter was observed; therefore, we suggest that this situation resulted from placenta-originated progesterone synthesis in pregnancy. Furthermore, levels of corpus luteum vitamin C were found to be higher and we concluded that this increase resulted from the effect of vitamin C on steroid and collagen biosynthesis. Nevertheless, negative correlations between corpus luteum vitamin C concentrations and corpus luteum weight/diameter were seen, and hence we suggest that this may be the result of the inhibitory effect of vitamin C on PGF2a synthesis.Öğe I?nvi?tro Soğan Yağı Uygulamasının Plazma Kolesterol ve Gli?koz Düzeyleri? ile Bazı Hematoloji?k Parametreler Üzeri?ne(Selçuk Üniversitesi, 1995 Nisan) Durgun, Zafer; Keskin, Ercan; Tiftik, Ali Muhtar; Keçeci, Tufan; Haliloğlu, SeyfullahIn this study, ten dogs, 6-8 months aged, were used. All dogs were fed normal diet plus cholesterol powder for ten days. At the end of the period, the means plasma cholesterol levels increased from 64. 60 to 230. 20 mg/ dl. Hypercholesterolemia didn't cause deformation in erytrocytes shapes. Then, ten blood samples taken from all the dogs were divided equally into two groups as control and experimental. Two hours after as in vitro 0. 2 ?l of onion oil addition to each 10 ml of blood sample in experimental group, plasma cholesterol levels in experimental and control groups decreased from 238. 20, 180. 40 and 171. 60 mg/dl respectively. Plasma glucose levels in experimental group decreased significatly as compared to control group after onion oil administration (P<0.05). After in vitro onion oil administration to experimental group, erytrocytes shapes showed deformation and crenation due to Heinz body formation resulting obviously falling in erytrocytes count, haemoglobine amount and haematocrit values in comparison with control group. Consequently, in this investigation, it was determined that onion oil had no effect on plasma cholesterol levels whereas reduced plasma glucose levels and had haemolytic effect on erytrocytes.Öğe Interleukin-1 receptor antagonist levels in gingival crevicular fluid and serum in nonsmoking women with preterm low birth weight and intrauterine growth retardation(Dental Investigations Society, 2015) Kayar, Nezahat Arzu; Alptekin, Nilgün Özlem; Haliloğlu, SeyfullahObjective: The aim of this study was to evaluate interleukin (IL)-1 ? and IL-1 receptor antagonist (IL-1ra) levels in gingival crevicular fluid (GCF) and serum (S) in nonsmoking women with normal birth (NB), preterm low birth weight (PLBW), and intra-uterine growth retardation (IUGR). Materials and Methods: In this unmatched case-control study design, 64 women with NB, 45 women with PLBW, and 47 women with IUGR were recruited within 24 h delivery. Clinical periodontal parameters were recorded. IL-1 ? and IL-1ra levels in GCF (pg/30 s) and serum (pg/ml) of were evaluated using commercial enzyme immunoassay and ELISA kits. Results: Greater pocket depth and clinical attachment loss were observed in PLBW and IUGR women than in NB women (P < 0.05). The total amounts of IL-1ra and IL-? of GCF were higher levels in NB women than PLBW and IUGR women (P < 0.05). The lowest total amount of IL-1ra of GCF was found in IUGR women (P < 0.05). The concentrations of IL-1ra in serum samples were not statistically significant for any of the study groups (P > 0.05). A total of 42 endosseous root-form dental implants of 14 patients were clinically examined by modified Plaque index (PI), modified Gingival index (Gl) and probing depth (PD). Smoking habits of the patients were recorded. PICF of implants were collected by Periopaper strips and IL-1beta, TNF-alpha levels were determined by enzyme-linked immunosorbent assay (ELISA). NE was analyzed with a neutrophil specific chromogenic substrate, N-methoxysuccinyl-Ala-Ala-Pro-Val-p-nitroanilide. The cytokine and enzyme levels in PICF were expressed as total amount/activity and as concentrations. NE activity in PICF significantly correlated with GI and PD, and IL-1beta levels with GI and PICF volume (P < 0.05). The correlations were stronger when the PICF levels were expressed as total IL-1beta amount and as total NE activity. The implants with inflamed gingiva (GI > 1) had higher levels of IL-1beta and NE activity than implants with noninflamed or slightly inflamed gingiva (GI less than or equal to 1) (P < 0.05). Total NE activity in implants with deep pockets (PD > 3 mm) was greater than the implants with shallow pockets (PD :5 3 mm) (P < 0.05). The implants of smoker patients had significantly lower PICF NE activity and IL-1beta levels, and significantly higher TNF-alpha levels than the implants of nonsmokers (P < 0.05). The findings of the present study indicate that NE activity and IL-1beta levels in PICF may be used to measure implant health status of patients who do not smoke.Öğe Koyunlarda Plazma Vitamin C ve Seruloplazmin Düzeyleriyle Eksojen Vitamin C Uygulamalarının Döl Verimi Üzerine Etkileri(Scientific Technical Research Council Turkey, 2000) Haliloğlu, Seyfullah; Serpek, BehiçThe effects of vitamin C on progesterone and oestradiol 17 beta synthesis, and fertility rate were studied in Merino sheep. A total of 60 sheep, aged between 2 and 7 years, of which 30 had low, and the other 30 had high plasma vitamin C levels were used. Each low and high vitamin C group was divided into 2 subgroups. Each subgroup had equal numbers of sheep. Subgroups 2 and 4 served as controls in the low and high vitamin C groups, respectively. 500 mg of vitamin C was injected intramuscularly into the subgroups 1 and 3 in the low and high vitamin C level groups, respectively. Plasma vitamin C and ceruloplasmin levels were determined using spectrophotometric methods. Plasma progesterone and oestradiol 17 beta levels were determined using the microtitration enzyme immunoassay technique. Although vitamin C levels were low during the sexual cycles in all groups, there was an increase with the start of pregnancy. On the other hand, the changes in the plasma ceruloplasmin levels were irregular. The low and high oestradiol 17 beta levels of the low and high vitamin C groups showed the importance of vitamin C in steroid hormone synthesis. There was a positive correlation between ascorbic acid levels and body weight of the mother sheep and new-born. The vitamin C and progesterone increases were observed in the sheep pregnant with twins. The aim of this study was to evaluate the length of sampling time and sequential sampling of GCF neutrophil elastase (NE) enzyme levels by using intracrevicular and orifice methods. Material and methods: Twenty adults (mean age of 41.8 years, ranged 31-60 years, 18 males and 2 females) with chronic periodontitis were enrolled and all completed the 3-d study. GCF was collected by both intracrevicular and intrasulcular methods, 720 samples of GCF were collected. In first, second and third day, the length of sampling time in seconds (s) and order were '5- 10-30-s'; '10- 30- 5-s' and '30- 5- 10-s,' respectively. GCF elastase levels were determined by hydrolysis of neutrophil specific substrate N-methoxysuccinyl-Ala-Ala-Pro-Val-p-nitroanilide. Results: NE activity (mu U) and NE activity/volume (mu U/mu l) were significantly different for order of sampling (p < .05), but not for the length of sampling time (p>.05). Conclusions: Within the limits of this study, the choice of sampling technique in GCF-profile studies seems to be a critical decision as it has the potential to affect the GCF volume and NE activity.
