Öğe Akkaraman Kuzularda Defaunasyonun Uçucu Yağ Asitleri Üzerindeki Etkisi(Selçuk Üniversitesi, 1988 Ocak) Kocabatmaz, Mehmet; Aksoylar, Mehmet Yaşar; Durgun, Zafer; Eksen, MursayettinTwo groups of Akkaraman lambs (faunated and defaunated lambs) were used to investigate the effect of the presence or absence of rumen ciliate protozoa on pH, volatile fatty acids of rumen content and weight gain. The lambs were fed with milk until 1.5 months of age. Then, all animals were given chopped sainfoin hay and concentrate diet for the duration of the experiment. For the purpose of defaunation of the 2nd group lambs, 0.3-0.8 % solution of Dioctyl Sodium Sulfosuccinate was administrated via stomach tube. Rumen samples were taken from the rumen of lambs before feeding and at 3, 6 hr. after feeding and pH values and volatile fatty acids levels were measured. pH values were found to be highest before feeding, decreased at the 3rd hr. and became lowest at the 6th. after feeding for each animals. Acetic acid, propionic acid, iso-butiric acid and total volatile fatty acid levels in the rumen contents of the faunated lambs were higher than in the defaunated lambs. The faunated lambs grew faster that the defaunated lambs. The average of body weights of the faunated lambs was 4.6 kg more than the faunated lambs It was, therefore, concluted that the rumen ciliate protozoa are essential for the metabolism and growth of young lambs.Öğe Ankara Keçi?leri?ni?n Rumen ve Kan Metaboli?tleri? Üzeri?nde Fosforun Etki?si?(Selçuk Üniversitesi, 1992 Ocak) Kocabatmaz, Mehmet; Eksen, Mursayettin; Durgun, ZaferIn this investigation, three Angora Goats of 3 years old and with the permanent rumen cannula were used. The goats were fed with alfalfa hay (40%) and the pelleted white beet pulp (60%). The first animal was used as a control and 1 g and 2.4 g phosphorus were infused via rumen cannula into rumen of the second and third animals respectively. Rumen content and blood samples were taken before feeding and at the 3rd and 6th hour after feeding with 7 days intervals. The pH values of rumen contents were highest before feeding and generally lowest at 6th hour after feeding for all animals. At the each sampling time, the average values of protozoa counts and percentage of Isotricha intestinalis, Isotricha prostoma and Dasytricha ruminantium for second and third animals to which phosphorus (Ig and 2.4 g respectively) infused were highest than first animal but the percentage of Entodinium minimum and Entodinium caudatum showed decrements for second and third animals. On the other hand, the highest NH3 - N values of rumen contents were determined for the second animal at each sampwling time but same values were lowest for first animal. Before feeding, urea-N of blood was highest and showed gradually decrements after feeding for first animal. The same values were found to be lowest before feeding, and showed increments for second animal at other sampling times. All of the total protein values generally were equal for all animals. The glucose values of blood sera belongig to the third animal were found higher than the other animals only the lower values of glucose at the before feeding showed gradually incaremets after feeding for all animals. In addition before of the effect of phosphate infusion, inorganic phosphorus values of rumen contents and blood sera of the second and third animals were higher than the first animal at each sampling time.Öğe Ankara Keçi?leri?ni?n Rumeni?ndeki? Si?li?alı Protozoonların Geli?şmesi?nde Farklı Rasyonların Etki?si?(Selçuk Üniversitesi, 1988 Ocak) Kocabatmaz, Mehmet; Eksen, Mursayettin; Durgun, ZaferIn this investigation pH values, protozoa counts, protozoa volumes and protozoa species were determined in the rumen contents of Angora goats fed with different rations. For this purpose 4 animals with 2 years old and with permanent rumen cannula were used. Feeding the animals was performed according to 4 x 4 Latin Square desing. Each animal was fed with four different types of rations (I, II, III, IV); alfalfa straw (100 %), alfalfa straw + concentrate mixture (80: 20, 60 40, 40: 60) respectively. pH value of rumen content was found to be highest (7.22) before feeding, but dropped to 6.21 at the 6 th hour and incereased again (6.42) at the 10 th hour following the feeding. The protozoa counts and their volumes of rumen contents showed a paralelism to the declination in pH values. Total 10 protozooon species were identified, but "Entodinium minimum" was predominant at each sampling time. The differences between pH values and protozoa counts were generally found to be higly significant (p<0,01). The protozoon counts and their volumes were higher for the animals fed IV th ration than for the animals having the other rations.Öğe Ankara Keçi?si?ni?n Rumen Sıvısı ve Kanında Bazı Metaboli?tleri?n Değerleri?(Selçuk Üniversitesi, 1988 Ocak) Kocabatmaz, Mehmet; Eksen, Mursayettin; Durgun, ZaferThree Angora Goats of 2-2.5 years old, with permanent rumen cannula were used in this experiment. Animals were fed with three different types of rations with concentrate mixture and alfalfa straw; 20:80, 40: 60, 60:40 respectively. In the investigation; pH Inorganic P and NH3-N values in the rumen fluid and NH3N in the blood and Urea-N, total protein, glucose, Inorganic P levels in the blood sera vere determined before feeding, at the 4 and 8th hour after feeding for each animal. At the 4 th hour after feeding the pH values in the rumen fluid and NH3-N of the blood were both decreased. The NH3-N values of the blood at the 8th hour after feeding and NH3-N levels of the rumen fluid at the 4th hour after feeding were highest. Inorganic P values in the blood sera were gradually increased but Inorganic P levels in the rumen fluid were decreased at the sampling times. No notable changes were observed in each sampling, as far as Urea-N, total protein and glucose levels of the blood sera are concerned.Öğe Aseti?k Asi?ti?n Koyunlarda Rumen pH'sı, Uçucu Yağ Asi?tleri? ve Protozoonlar Üzeri?ndeki? Etki?leri?(Selçuk Üniversitesi, 1995 Nisan) Keçeci, Tufan; Kocabatmaz, Mehmet; Kadak, RamazanThis study was conducted to investigate the effect of ruminal infusion of acetic acid on the ruminal pH, volatile fatty acids (VFA) and protozoa in the sheep. Two fistulated sheep were infused through rumen fistula with 500 ml water (control) or acetic acid (0.62 gr/kg body weight) before feeding for 4 consecutive days. Samples of rumen contents were collected from each sheep before and 1 h after, infusion through the ruminal fistula. Acetic acid infusion has caused a significant drop in the ruminal pH. Furthermore, there was a reducing effect of acetic acid on number of ruminal protozoa. Especially, the Holotrich protozoa were the most susceptible, Whereas Entodinia were the least sensitive to the reducing effect of acetic acid. While the proportions of Holotrich species decreased, the proportions of Entodinium species were generally found to be increase after acetic acid supplementation into the rumen of sheep. Compared with the values of same parameters in the sheep given water, acetic acid proportions and total VFA levels were determined to be higher, but the proportions of propionic and butyric acids were found lower levels in the sheep given acetic acid.Öğe Büyümekte Olan Erkek ve Di?şi? Thirty five male and 35 female, 1 to 8 week-aged Japanese quails were used in this study. Haematological examination were carried out in 5 male and 5 female quails at 1 st, 2 nd, 3 rd 4 th, 5 th, 6 th and 8 th week of ages The red blood cell (RBC) counts, haemoglobine amounts (Hb) and hematocrit values (PCV)) in both groups increased significantly with increased age (P<0.05). The RBC, Hb and PCV were found higher in male than female in all the age periods. The RBC was significantly higher in males (3.65x 106) than females (3.28x 106) at 8 week of age (P<0.05). The white blood cell (WBC) and thrombocyte counts did not show evidently changes, whereas WBC in females (2428x 103) were found slightly higher than males (21-26 x 10³) at all weeks. In the percentage of leucocyte types were not determined any differences between sexes during eight weeks and the lymphocyte percentages (57-70%) were drastically higher than heterophils (24-36 %) in both groups. There was no differences between sexes in respect to the blood cell sizes where as RBC sizes were bigger in females than males at either 1 st and 8 th weeks. On the other hand, in sizes of thrombocyte and leucocyte types in both groups were not determined differences between 1 st and 8 th weeks, but RBC sizes at 8 th week in both groups decreased significantly as compared with 1st week (P<0.05). Bundan dolayı, çalışmada Türkiye'de ekonomik değeri olan beş tatlı su balığı türünün (Gökkuşağı alabalığı, Yayın, Tatlı Su Kefali Aynalı ve Pullu Sazan) lökosit formülü ve lökosit tiplerinin indentifikasyonları yapıldı. Lökositlerin ayırımında ve oranlarının belirlenmesinde May Grünwald - Giemsa karışık boyama yöntemi kullanıldı. Lökositlerin çekirdekleri normal olarak hazırlanan Giemsa boyası ile yeteri kadar boyanmadıklarından, boya hazırlanırken her ml distile suya 1.5 damla Giemsa ana mahlül ilave edildi. Her lökosit tipinin ortalama yüzdeleri ile ayırımları balık türleri için ayrı ayrı yapıldı. After 10 th day of 16 day period, experimental group received daily 50µl of onion oil with 200 g of ground meat additon to cholesterol-supplemented diet whereas control group continued feeding cholesterol-suplemented diet and 200 g of ground meat for six days. At the 5 th. and 6 th. days of period of onion oil administration (15 th. and 16 th. days of total period). Etki?si?(Selçuk Üniversitesi, 1995 Nisan) Keçeci, Tufan; Kocabatmaz, MehmetIn this study, 50 white leghorn cocks which were at the same age, similar body weight and healthy were used. The animals were divided into 5 groups as Control, Group ST, Group AA, Group ASA, and Group ASA?. The groups were placed into different coops. In the coops, while it was 40 dm2 floor area to control animals for one bird, it was 10 dm2 floor area to the animals in the other groups for one bird. During the experiment, while ascorbic acid was added in the ration of the animals in the Group AA, acetylsalicylic acid was added in the ration of the animals in the Group ASA? and Group ASA? at the different dose levels. Because of the increase in the ratio of circulating heterophils to lymphocytes (H/L), in the animals except control ones were determined to occure stress. The mean of red blood cell (RBC), haemoglobin amount (Hb), hematocrit values, the values of erytrocyte sedimentation rate (ESR) at Ist, 2nd, 24th hours, thrombocytes and white blood cell (WBC) count, proportions of heterophiles, eosinophiles, basophiles, monocytes and lymphocytes, H/L ratio in control animals were determined to be 2. 899x100/mm³. 8. 24 gr/ dl, 28. 30 %, 2.56 %, 1. 58 %, 65. 84 % and 0. 43 respectively. In the animals of Group ST, the mean of lymphocyte, eosinophil, basophil proportions and ESR at 2nd, 24th hours decreased (54. 90 %, 0. 64 %, 1. 20 %, 46. 12 mm and 113.76 mm respectively). However, the mean of H/L ratio, heterophil proportions, RBC and WBC counts, Hb and he matocrit values increased (0. 77, 41. 38 %, 3. 448x106/mm3, 36. 58x103/mm³, 10. 56 gr/dl and 32. 26 % respectively). The mean of H/L ratio, lymphocytes, eosinophil, basophil, heterophil and monocytes proportions, RBCWBC and thrombocytes counts, Hb, hematocrit values, ESR at 1st, 2nd and 24th hours in the animals those were given ascorbic acid were found to be 0. 51, 63. 22 %, 0. 92 %, 2. 04 %, 32. 08 %, 1. 88 %, 1. 151x106/mm³, 3300x103/mm³, 0. 4664x105/mm3, 9. 59 gr/dl, 30. 22 %, 24. 78mm, 47. 34mm and 115. 46 mm respectively. On the other hand, in the cocks of Group ASA?, the same values were determined to be 0. 61, 59. 72 %, 0. 80 %, 1. 64 %35. 82 %, 2.02 %, 3. 332x106/mm³, 35, 74x103/mm3, 0, 4696x105/mm3, 10. 18 gr/dl, 31, 92 %, 24. 64mm, 46. 66mm and 113. 52mm respectively. Whereas, the same values in the animals of Group ASA? were found to be 0. 70, 57.00 %, 0.72 %, 1.84 %, 38. 72 %, 1. 72 %, 3. 386x105/mm3, 36. 52x103/mm3, 0. 4688x105/mm³, 10. 30 gr/dl, 32. 02 %24. 63 mm, 46. 98 mm and 114. 74 mm respectively. Based on results, it was concluded that some haematological parameters was influenced by stress and ascobic acid was more effective agent than acetylsalicylic acid on stress in poultry.Öğe Farklı Rasyonların Ankara Keçilerinin Rumen pH'si ve Uçucu Yağ Asitleri Üzerindeki Etkisi(Selçuk Üniversitesi, 1989 Ocak) Kocabatmaz, Mehmet; Aksoylar, Mehmet Yaşar; Durgun, Zafer; Eksen, MursayettinThree Angora goats of 2 years old and with permanent rumen cannula were used in the experiments. Feeding the animals were performed according to 3 * 3 Latin square desing with 30 day periods. The alfalfa straw and concentrate mixture were fed in the proportions of 80 20, 60/40 and 40 60 in treatments I, II and III respectively. The rations were given once a day at the 3% level of each goat's body weight. The ruminal samples were taken before feeding at 4 th and 8 th hours after feeding in the remaining twelve days of each period pH values of rumen content for all of 3 rations were found to be highest (7.15, 7.25, 7.25) before feeding, decreased (5.95, 6.17, 6.32) at the 4 th hour and in- creased (6.03, 6.22, 6.46) at the 8 th hour after feeding. While the pH values of rumen contents were decreasing acetic acid, propionic acid, butyric acid and total volatile fatty acid increased at 4 th hour after feeding. The difference between the acetic acid values of treatment II and III was found significant (p < 0.05) at 4 and 8 th hours after feeding. Where as the difference between butyric acid values of treatment II and III was found highly significant (p < 0.01) at all sampling times. All values of volatile fatty acids were higher for treatment III than the other treatments at the each sampling time.Öğe Farklı yaş ve cinsiyetteki Isviçre esmeri sığırlarda kan serumu tiroid hormonları ile bazı hematolojik değerler(1995) Keçeci, Tufan; Kocabatmaz, MehmetBu çalışmada, farklı yaş ve cinsiyetteki ile gebe olan İsviçre Esmeri sığırlarda, kan serumu tiroid hormonlarının düzeyleri ile bazı hematolojik değerlerin belirlenmesi planlandı. Bu amaçla, 42 sığırdan alınan kan örneklerinde; kan serumu total tiroksin (TT4) ve triiyodotironin (TT3) düzeyleri, alyuvar sayısı, hemoglobin miktarı, hematokrit değer, 1., 2. ve 24. saatlerdeki alyuvarların sedimentasyon hızı değerleri, ortalama alyuvar hacmi, ortalamaa alyuvar hemoglobini, ortalama alyuvar hemoglobin derişimi, akyuvar sayısı, lenfosit, nötrofil, monosit, eozinofil ve bazofil oranları belirlendi. Dişi ve erkek buzağılarda; ortalama kan serumu TT4 (sırasıyla 5.59 ug/dl ve 5.18 ug/dl ) ve TT3 (118.48 ng/ di ve 110.57 ng/dl) düzeyleri, alyuvar sayısı (8.50xl06 /mm3 ve 8.42xlO6/mm3), hemoglobin miktarı (12.83 gr/dl ve 12.50 gr/dl), hematokrit değer (%38.67 ve %37.67), lenfosit (%70.17 ve %69.33) ve monosit (%7.33 ve %6.17) oranlan düve ve tosunların aynı değerlerinden (sırasıyla 4.31 J.g/dl ve 3.98 ug/dl, 97.12 ng/dl ve 92.17 ng/dl, 7.48xlO6/mm3 ve 7.77xlO6/mm3, 11.50 gr/dl ve 11.83 gr/dl, %33.83 ve %35.17, %64.17 ve %63.00, %5.17 ve %5.33) daha yüksek j miktarlardaydı. Gebe olmayan kurudaki yetişkin ineklerin ve boğaların aynı değerler' ise ' sırasıyla 4.17 ig/dl ve 3.74 (o.g/ di, 94.42 ng/dl ve 90.72 ng/dl, 6.80xl06/mm3 ve 7.25xl06/ mm3, 10.83 gr/dl ve 11.50 gr/dl, %32.50 ve %34.50, %63.17ve %61.00, %4.00 ve %3.83), düve ve tosunların parametrelerinden daha az miktarlardaydı. Ancak, dişi (%20.67) ve erkek (%22.33) buzağılann nötrofil oranları düve (%28.33) ve tosunlar (%29.00) ile gebe olmayan kurudaki yatişkin ineklerin (%30.00) ve boğaların (%32.00) nötrofil oranlarından daha az miktarlardaydı. Gebe ineklerin kan serumu TT4 (5.55 xg/dl) ve TT3 (118.40 ng/dl) düzeylerinin, akyuvar sayılarının (7.85x103 /mm3) ve monosit oranlarının (%5.50) gebe olmayan kurudaki hayvanların aynı değerlerinden (ortalama akyuvar sayısı 7.80xl03 /mm3) daha fazla miktarlarda olduğu belirlendi. Denemeye alınan hayvanların diğer parametreleri arasında herhangi bir farklılık yoktu. Eighty, one day old Japanese quail chicks were used in this experiment. They were randomly distributed in four groups, each including 20 chicks. The chicks in group I(control), II, III and IV were fed control ration, the ration containing 1, 0,1 and 0.01% seaweed extract, respectively until they reached 5 weeks of age. The animals were offered the rations and the water ad libitum. At 1 st, 2 nd and 3 rd weeks of age, 5 chicks of each group were randomly slaughtered for blood withdrawal from each chicks. At the and of the first week, the erythrocyte count, hemoglobine amount, haematocrit value, plasma glucose and total protein levels in group II fed ration caontaining %1 seaweed extract were found higher than other groups altough it was no differences among all groups (P>0.05). At the termination of the fifth week, erythocyte count, hemoglobine amount and haemotocrit value in group II were determined signifacantly higher than group I and IV (P<0.05). At that time the plasma glucose and total protein levels were found obviously higher in group II as compared to other groups and plasma glucose level in group II was significantly different than group I and IV (P<0.05) During the whole experiment, it was not dtermined striking changes in respect to leucocyte types, leucocyte and trombocyte counts in all groups.Öğe Hi?poti?roi?di?zmi?n Kan Üre Azotu, Total Protein, Gli?koz ve Total Kolesterol Düzeyleri? The animals were divided into 5 groups as Control, Group 1-A, Group 1-B, Group 2-A and Group 2-8 During the first 3.5 month, hypothyroidism was induced except the control animals all the other by administration of thiourea at the different dose levels. Then, thiourea administration was lasted and potassium iodide (KI) was given to the animals in the Group 1-B and Group 2-B for 3.5 month. In the animals which were given thiourea, because of hypothyroidism, urea-N and total cholesterol levels in the blood serum increased (25.58 mg/dl and 83.78 mg/dl respectively). However, glucose levels in the blood serum decreased (40.34 mg/dl). While the parameters of the animals those were given KI, reached at the levels of control ones (mean blood serum urea-N 17.40 mg/dl, total cholesterol 55.84 mg/dl, glucose 59.00 mg/dl), it was determined that, in the lams not given KI, the levels of those parameters were different values in proportion to the control ones (mean blood serum urea-N 23.57 mg/dl, total cholesterol 78.48 mg/dl, glucose 41.25 mg/dl). Because of hypothyroidism, body weight gain, ruminal pH, ruminal protozoa count and ruminal total volatile fatty acids /eve/s decreased. However. ruminal and blood ammonia nitrogen /eve/s and ruminal bacteria counts ine· reased. The parameters of the anima/s those were given KI, reached at the /eve/s of control ones. In the anima/s not given KI, the increase of those parameters were at lower /eve/s. At the end of the experiment, the mean body weight of control anima/s was 6S kg and the anima/s those were given KI at the end of the hypothyroidism was S6 kg. Where as, it was rather lower (4S kg) in the hypothyroidism induced but not given KI ani ma/s. The animals were divided into 3 groups as Control, Group ST (Stress) and Group AA (Ascorbic acid). Food and water were given ad libitum. Each group was placed into different compartment. It was 40 dm² foreach bird off loor area for the control animals. But, in the experimental groups, the floor area was 10 dm2 for each bird. During the experiment, ascorbic acid (vitamin C) was added to food of the animals in Grofp AA. (150 mg/kg) The increased levels of plasma corticosterone is experimental groups indicated to occur stress. In the cocks of Control Group; mean blood serum total thyroxine (TT4), total triiodothyronine ((TT3), glucose, total cholesterol, total protein levels and plasma corticosterone levels were found to be 2.78 µg/dl, 0.71 ng/dl, 185.63 mg/dl, 115.35 mg/dl, 4.17 gr/dl and 0.96 ng/ ml respectively. The levels of serum TT3 (0.45ng/ml) and total cholesterol (106.62mg/dl) in the animals of Group ST decreased. However, plasma corticosterone (1.57ng/ml) and serum glucose levels (207.32mg/dl) were increased. Mean serum TT3, glucose, total cholesterol and plasma corticosterone levels in the animals those were given ascorbic acid were determined to be 0.53ng/ml, 191.64mg/dl, 111.54mg/dl and 1.02ng/ml respectively. The levels of serum TT4 and total protein amongst the groups were not significantly differences. Based on results, it was concluded that some blood metabolites were significantly influenced by stress.Öğe I?shalli? As soon as the calves arrived to clinic the blood samples were taken from the jugular vein in order to determine the following blood parameters: packet cell volume (hematocrit %), leukocyte number, differential leukocyte count %, sedimentation rate, serum urea, Na, K, Inorganic P and total protein. Especially, the increase in PCV (Hematocrit %). leukocyte number and serum urea were much pronounced in the blood of calves. To decide about prognosis and the treatment of the calves, these measurerements are taken into consideration. The analysis of these blood parameters was carried out before the treatment and during the treatment period. The treatment of animals consisted of subcutaneous infusion of a isolayt solution mixed with hyaluornidaes (50-150 ml isolayt + 15-45 I.U. hyaluronidase per kg of body weight). On the other hand, the application of the effective antibiotics were administered orally (chloramphenicol; 30 mg/kg). and intramuscular (Gentamycine; 5 mg) kg) based on the results of an antibiogram carried out on facces. Three calves died but the other 47 calves were returned to their health.
