    Anti-diabetic and anti-hyperlipidemic properties of Capparis spinosa L.: In vivo and in vitro evaluation of its nutraceutical potential

    In this study, the nutraceutical potential of Capparis spinosa L for the treatment of hyperglycemic states has been thoroughly investigated. A series of in vivo and in vitro tests have been conducted on fresh leaf, buds and salty buds (24 h desalted) processed to dry powder. 60% MeOH/H2O extracts were obtained for HPLC analysis and for alpha-amylase and alpha-glucosidase inhibition tests. To estimate the in vivo anti-diabetic effect, dry powders of C spinosa leaf and buds were orally administered to streptozocin-induced diabetic rats over a period of 28 days. At the end of the experiment, animals were sacrificed, blood taken for assessment of lipid profile and liver/kidney biochemistry while section of the pancreas, liver and kidneys were processed for general histology. Results showed that the regular administration of C. spinosa leaf or buds normalized all the biochemical parameters and reversed the liver/kidney injury with variable degrees of organ protection.     Asphodeline Cilicica Tuzlaci: From the Plant to Its Most Active Part Extract and Its Broad Bioactive Properties

    This study investigated into the polyphenolic content, antioxidant and enzyme (a-amylase, a-glucosidase, tyrosinase, acetylcholinesterase [AChE], and butyrylcholinesterase [BChE]) inhibitory activities of 12 extracts of the endemic Asphodeline cilicica (AC) Tuzlaci, from Turkey. The total phenolic content for the methanol and acetone extracts of AC roots were 56.52 and 52.96 mg GAEs/g extract, respectively. Both the acetone and methanol extracts of AC roots exhibited an important antioxidant capacity. Likewise, the acetone and methanol extracts of AC roots revealed to be the most potent scavengers of DPPH radical (58.25 and 58.78 mg TEs/g extract). Acetone extract of AC roots inhibited a-amylase showing an acarbose equivalent value (mmol ACAEs/g extract) of 0.27, while against a-glucosidase the value recorded was 2.30. The acetone and methanol extracts of the different plant parts of AC inhibited AChE and BChE, while no/poor activity was noted for the water extracts. In the present study, extracts of AC showed inhibitory activity against key enzymes involved in diabetes and neurodegenerative complications, thereby advocating for the need for in-depth studies geared towards the discovery of pharmacophores from this endemic plant.     Biological and chemical insights of Morina persica L.: A source of bioactive compounds with multifunctional properties

    Morina persica L. has weak rosy smell flowers that together with aerial parts are infused or decocted and used as a functional tea. It is a representative of the Anatolian traditional medicine and is used for the treatment of cold. Biological and chemical fingerprints of different extracts (methanolic, acetone and water) of M. persica were investigated. Antioxidant, enzyme inhibitory, antimicrobial, cytotoxic and mutagenic/antimutagenic effects were evaluated for biological profiling. Rutin, chlorogenic and rosmarinic acids were detected as dominant compounds. Generally, the methanolic and acetone extracts had strong antioxidant, enzyme inhibitory, antimicrobial and antimutagenic effects. Additionally, all extracts showed remarkable inhibitory effects on HeLa cells at 100 mu g/mL. Furthermore, possible interactions between rutin, the dominant phenolic compound in M. persica and the tested enzymes were assessed by molecular docking. Results indicate that M. persica could be considered as a natural source of high-valued functional ingredients for further use in healthful formulations. (C) 2016 Elsevier Ltd. All rights reserved.

    Chemical composition and biological activities of extracts from three Salvia species: S. blepharochlaena, S. euphratica var. leiocalycina, and S. verticillata subsp amasiaca

    The genus Salvia has recently attracted great attention due to its notable biological activities. Within this context, in this study, the chemical characterization and biological effects of three extracts (dichloromethane (DCM), methanol (MeOH), and water) from three Salvia species (S. blepharochlaena (SB), S. euphratica var. leiocalycina (SE), and S. verticillata subsp. atnasica (SV)) were assessed. For the chemical characterization, the qualitative and quantitative analysis of phenolic components in the methanol extracts was carried out by high-performance liquid chromatography with electrospray ionization mass spectrometry detection (HPLC-UV-ESI-MS). Total phenolic, flavonoid and phenolic acid contents were also studied. Concerning the biological effects, antioxidant (DPPH and ABTS free radical scavenging; ferric (FRAP) and cupric (CUPRAC) reducing power; phosphomo-lybdenum, and metal chelating assays), enzyme inhibitory (cholinesterase, tyrosinase, amylase, glucosidase, lipase, and elastase) and cytotoxic effects (A-549 and MCF-7 cell lines) were evaluated. After the evaluation of the phytochemical profile by HPLC-UV-ESI-MSn, it was observed that the main compound in the analyzed extracts was rosmarinic acid, which was present at high concentrations, particularly in SV, which presented rosmarinic acid levels higher than the usual levels found in other Salvia species or related plants. Generally, the SV-water extract presented the strongest antioxidant abilities with higher levels of total bioactive compounds. However, the studied DCM extracts had higher enzyme inhibitory potentials compared with MeOH and water extracts. SE-DCM exerted the most potent cytotoxic effects, followed by SB-water and SB-MeOH extracts.

    Cytotoxic and Enzyme Inhibitory Potential of Two Potentilla species (P-speciosa L. and P-reptans Willd.) and Their Chemical Composition

    In this work, the biological and chemical fingerprints of three extracts (ethyl acetate, methanol, and water) from two Potentilla species (Potentilla reptans and P. speciosa) were investigated. Antioxidant, enzyme inhibitory, and cytotoxic activities were performed for the biological fingerprint. For the chemical characterization, total bioactive components, and individual phenolic components were determined using photometric and HPLC methods, respectively. The main identified phenolic compounds in these extracts were rutin and catechin. Methanol and water extracts contained the highest total phenolic and flavonoid content. The results of antioxidant assays showed that methanol and water extracts displayed higher antioxidant activity compared to the ethyl acetate extract. Generally, methanol and water extracts exhibited higher biological activities correlated with higher levels the bioactive components. For P. speciosa, the methanol extract exhibited the highest enzyme inhibitory activity (except BChE inhibitory activity). P. reptans exhibited also high antiproliferative activity against MCF-7 cells whilst P. speciosa had weak to moderate activity against both of A549 and MCF-7 cell lines. The results suggest that Potentilla species could be potential candidates for developing new phyto-pharmaceuticals and functional ingredients.

    Enzymatic assays and molecular modeling studies of Schisandra chinensis lignans and phenolics from fruit and leaf extracts

    Considerable interest has been shown in natural sources and their compounds in developing new therapeutically agents for different diseases. In this framework, investigations performed on this topic play a central role for human health and drug development process. Schisandra chinensis (Turcz.) Baill is a medicinal and edible plant showing highly advantageous bioactivity and nutritional value. The main bioactive compounds from its fruits are lignans, derivatives of dibenzocyclooctadiene whereas concerning its leaves, phenolic acids, and flavonoids are dominant. The purpose of this study was to investigate the enzyme inhibitory potential on selected carbohydrate hydrolases, cholinesterases, and tyrosinase of extracts from fruits and leaves of Schisandra in relation with their main bioactive compounds. Furthermore, the interactions between dominant compounds (schisandrol A, schisandrol B, schisandrin B, and cinnamic acid) from extracts and selected enzymes were investigated by molecular modeling and molecular dynamic studies in order to explain at a molecular level our findings.

    Euphorbia denticulata Lam.: A promising source of phyto-pharmaceuticals for the development of novel functional formulations

    In this study, Methanolic extracts of Euphorbia denticulata parts (flowers, leaf, stem, and mix of aerial parts) were assessed for a panoply of bioactivities. Inhibitory potential against key enzymes involved in diabetes (alpha-glucosidase and alpha-amylase), obesity (pancreatic lipase), neurodegenerative diseases (cholinesterases), and hyperpigmentation (tyrosinase) was evaluated. The antioxidant and antibacterial properties were also assessed. The total phenolic, flavonoid, and phytochemical profile were established using HPLC/DAD and molecular modelling studies on specific target compounds were performed in silico. The flower extract was found to be rich in phenolics and flavonoids, (60.11 +/- 1.40 mgGAE/g and 42.04 +/- 0.16 mgRE/g respectively), which tend to correlate with the high radical scavenging activity of this extract (120.34 +/- 3.33 mgTE/g and 165.42 +/- 2.16 mgTE/g for DPPH and ABTS respectively). Catechin, epicatechin, gallic acid, p-OH-Benzoic acid, rosmarinic acid, and epigallocatechin gallate, found in significant abundance in the extracts were assessed using molecular modelling with the aim to study their docking properties on a set of six enzymes used in this study. The extracts were moderately effective with MIC values ranging between 1.56 to 6.25 mg/ml, but potent growth inhibitors of MRSA strains. Results amassed herein can be used as a stimulus for further studies geared towards the development of novel phyto-pharmaceuticals. (C) 2016 Elsevier Masson SAS. All rights reserved.

    Functional constituents of six wild edible Silene species: A focus on their phytochemical profiles and bioactive properties

    Six wild species (S. alba, S. conoidea, S. dichotoma, S. italica, S. supina, and S. vulgaris) from the Silene genera were tested for potential anti-enzymatic (acetyl cholinesterase (AChE), butyryl cholinesterase (BChE), tyrosinase, alpha-amylase, and alpha-glucosidase), antimicrobial (16 microbial strains), and antioxidant activity. An ultra-high performance liquid chromatography-electrospray ionization-tandem mass spectrometry method was used for phytochemical determination. Quinic acid, malic acid, protocatechuic acid, p-coumaric acid, and hesperidin were common in the six Silene species. All extracts showed higher antibacterial effects compared to streptomycin and ampicillin (except S. dichotoma). Antifungal agents, bifonazole (MIC 0.10-0.20 mg/mL and MFC 0.20-0.30 mg/mL) and ketoconazole (MIC 0.15-2.30 mg/mL and MFC 0.20-3.50 mg/mL) showed lower activity than the investigated Silene species extracts. S. alba inhibited AChE (2.00 mg GALAE/g extract) and BChE (1.0 mg GALAE/g extract). The results showed metal chelating potential ranging from 12 to 19 mg EDTAE/g extract, with S. conoidea being the most active, and S. supina the least. S. dichotoma showed the highest reducing potential against both cupric (154 mg TE/g extract for CUPRAC) and ferric (102 mg TE/g extract for FRAP) ions. Overall, Silene species could be considered as emerging interesting functional foods and sources of nutraceuticals with applications in the management of different diseases.

    Functional constituents of wild and cultivated Goji (L-barbarum L.) leaves: phytochemical characterization, biological profile, and computational studies

    Goji (Lycium barbarum L.) leaves are emphasized as a functional tea or as dietary supplements. The phenolic compound profile, antioxidant, enzyme inhibitory, antimicrobial, and antimutagenic activities of leaf extracts from two selected cultivars in comparison with wild-growing plants have been evaluated. HPLCDAD/ ESI-ToF-MS analysis revealed the presence of phenolic acids and flavonoids with chlorogenic acid and rutin being the dominant compounds in the cultivated plants, whereas rutin and kaempeferol-3-O-rutinoside for wild growing ones. In particular, cv. Erma contained the highest amount of chlorogenic acid and showed a strong tyrosinase-inhibitory effect. Staphylococcus aureus, Listeria monocytogenes, and Penicillium funiculosum were the most sensitive strains when exposed to extracts from cultivated plants. Antimutagenic activity was evaluated by Ames' test. The tested extracts provided high protection against mutagenicity induced by 2-anthramine (2-AA) to Salmonella typhimurium strains TA 98 and TA 100 (max. inhibition (%) 88% and 74.2%, respectively). Overall, Goji leaves are a rich source of bioactive compounds with functional properties that need further risk/benefit evaluation when used in foods or health-promoting formulations.

    Identification of phenolic components via LC-MS analysis and biological activities of two centaurea species: c. drabifolia subsp drabifolia and c. iycopifolia

    The Centaurea genus has great potential in traditional systems and has attracted much interest in the design of novel drug formulations. The present study was focused on the chemical fingerprints and biological properties of Centaurea drabifolia subsp. drabifolia and Centaurea lycopifolia extracts. Spectrophotometri c and LC-MS techniques were used to establish the chemical profiles of the studied extracts. Enzyme inhibitory potential was assessed against key enzymes linked to global health problems, namely neurodegenerative diseases (acetylcholinesterase), pigmentation (tyrosinase), and diabetes (alpha-amylase and alpha-glucosidase). The antimicrobial propensities of the extract were evaluated against 16 bacterial and fungal strains using the microdilution method. The antioxidant abilities were assessed using DPPH and ABTS radical scavenging, ferric, and cupric reducing powers, phosphomolybdenum, and ferrous metal chelation. The total phenolic compounds varied from 18.33 to 32.84 nigGAE/gextract.Total flavonoid content of the extracts were in the range of 2.88-22.39 mgRE/g extract. Methanol and water extracts showed stronger antioxidant abilities compared to the ethyl acetate extracts. However, the latter extracts were most efficient towards the target enzymes (except for tyrosinase). The water extracts also exerted considerable antimicrobial effects. Findings from the present work tend to support the idea that C. drabifolia subsp. drabifolia and C lycopifolia may be utilized as effective bio-resources for designing novel health promoting products or ingredients. It is anticipated that results amassed from this still will open new avenues for research and contribute towards establishing primary data on these species for designing novel phytopharmaceuticals.     Insight into the biological properties and phytochemical composition of Ballota macrodonta Boiss. et Balansa, - an endemic medicinal plant from Turkey

    Ballota macrodonta Boiss. et Balansa., is a traditionally used endemic medicinal plant used to manage a plethora of diseases in Turkey. Nonetheless, few studies have endeavoured to highlight its therapeutic potential and its phytochemical profile. We hypothesized that B. macrodonta would possess multi-pharmacological propensities and bioactive compounds that would justify its use as a folk remedy. Aerial part of B. macrodonta was assessed for its antioxidant, antimicrobial, and enzymatic inhibitory potential using different extraction solvents. The total bioactive components determination and compound identification using High-Performance Liquid Chromatography with Diode-Array Detection (HPLC-DAD) method were used to correlate the biological properties. HPLC-DAD profile showed the presence of 19 components (e.g. hydroxybenzoic acid, catechin hydrate, vanillic acid, caffeic acid, syringic acid, epicatechin, coumaric acid, ferulic acid, luteolin-7-O-glucoside, rutin, and quercetin). The water and methanolic extracts showed high total phenolic and flavonoid contents, respectively. The overall antioxidant capacity of the water extract was higher than the one of the methanol extract. The extracts showed inhibitory activity against cholinesterases, tyrosinase, alpha-amylase and alpha-glucosidase. The extracts also presented antimicrobial activity against eight Gram positive/negative bacteria (MIC - 0.05-0.15 mg/mL) and eight fungi (MIC - 0.0125-0.20 mg/mL). The presence of active biomolecules with multi-pharmacological properties makes B. macrodonta a potential source of health-promoting compounds that can be exploited as a novel medicinal herbal product.

    Lathyrus Aureus and Lathyrus Pratensis: Characterization of Phytochemical Profiles by Liquid Chromatography-Mass Spectrometry, and Evaluation of Their Enzyme Inhibitory and Antioxidant Activities

    Different wild plants commonly used in folk medicine, such as different species from the genus Lathyrus, may represent new sources of biologically active compounds. Hence, a study of the composition and (bio)chemical behaviour of extracts from these plants may provide valuable information. To evaluate the phytochemical profile, and the enzyme inhibition and antioxidant activities of the aerial parts of L. pratensis and L. aureus, extracts from both plants were analyzed by high-performance liquid chromatography with electrospray ionization mass spectrometric detection (HPLC-ESI-MSn). The in vitro antioxidant activity (phosphomolybdenum, beta-carotene bleaching, DPPH, ABTS, FRAP, CUPRAC and metal chelating) and enzyme inhibitory activity (acetyl cholinesterase, butyrylcholinesterase, tyrosinase, alpha-amylase and alpha-glucosidase) were also investigated for these Lathyrus species. Flavonoids and saponins were the main groups of compounds detected in the extracts from both plants. Generally, the methanol and water extracts presented remarkable antioxidant and enzyme inhibitory effects; all the observed results are critically discussed. The content of organic compounds and the antioxidant and enzyme assays suggest that these plants may be further used in phytopharmaceutical or food industry applications.

    Medicinal plants and natural products used in cataract management

    Cataract is the leading reason of blindness worldwide and is defined by the presence of any lens opacities or loss of transparency. The most common symptoms of cataract are impaired vision, decreased contrast sensitivity, color disturbance, and glare. Oxidative stress is among the main mechanisms involved in the development of age-related cataract. Surgery through phacoemulsification and intraocular lens implantation is the most effective method for cataract treatment, however, there are chances of serious complications and irreversible loss of vision associated with the surgery. Natural compounds consisting of antioxidant or anti-inflammatory secondary metabolites can serve as potential leads for anticataract agents. In this review, we tried to document medicinal plants and plant-based natural products used for cataract treatment worldwide, which are gathered from available ethnopharmacological/ethnobotanical data. We have extensively explored a number of recognized databases like Scifinder, PubMed, Science Direct, Google Scholar, and Scopus by using keywords and phrases such as "cataract", "blindness", "traditional medicine", "ethnopharmacology", "ethnobotany", "herbs", "medicinal plants", or other relevant terms, and summarized the plants/phytoconstituents that are evaluated in different models of cataract and also tabulated 44 plants that are traditionally used in cataract in various folklore medical practices. Moreover, we also categorized the plants according to scientific studies carried out in different cataract models with their mechanisms of action.

    Phenolic compounds and biological effects of edible Rumex scutatus and Pseudosempervivum sempervivum: potential sources of natural agents with health benefits

    The present study outlines a chemical characterization and further effects beneficial to health of edible Rumex scutatus and Pseudosempervivum sempervivum, in addition to presenting the antioxidant, enzyme inhibitory effects and antimicrobial properties of different extracts. The phenolic compounds composition of the extracts was assessed by RP-HPLC-DAD, outlining benzoic acid and rutin as major constituents in P. sempervivum and rutin and hesperidin in R. scutatus. Moreover, further biological effects were tested on key enzymes involved in diabetes mellitus, Alzheimer's disease and skin melanogenesis revealing an important tyrosinase inhibitory effect of Pseudosempervivum water extract. Moreover, both species possessed antimicrobial properties towards bacteria and fungi relevant to public health. Accordingly, we find that R. scutatus and P. sempervivum can be considered as novel functional foods because they are rich sources of biologically active compounds that provide health benefits.

    Phenolic profile and bioactivities of Sideritis perfoliata L.: The plant, its most active extract, and its broad biological properties

    Sideritis, also named "ironwort," "mountain tea," or "shepherd's tea," is a genus of flowering plants used as herbal medicine in traditional Mediterranean-area medicine systems, and these plants are generally consumed as a herbal tea. Its use as herbal tea and in traditional herbal medicine is quite popular. There are currently few studies on Sideritis perfoliata L., and only one reports the use of a liquid chromatography coupled to diode array detection and electrospray ionization tandem mass spectrometry (LC-DAD-ESI-MSn) profile and the content of phenolic compounds without considering a possible correlation with its biological activities. This paper aims to investigate the antioxidant activities by means of several different biological/biochemical assays (radical scavenging, reducing power, ferrous ion chelating, and total antioxidant by phosphomolybdenum and beta-carotene bleaching methods) as well as analyze the enzyme inhibitory activities (against AChE (acetylcholinesterase), BChE (butyrylcholinesterase), tyrosinase, alpha-glucosidase, and alpha-amylase) as well as the total phenolics, flavonoids, and condensed tannins. The reported results on Sideritis perfoliata highlighted that methanol and water extracts generally showed higher radical scavenging and reducing power activities. A similar trend could be observed for phosphomolybdenum and ferrous ion chelating activities. Methanol extracts showed lower activity only for the beta-carotene bleaching assay.

    Shedding light on the biological and chemical fingerprints of three Achillea species (A-biebersteinii, A-millefolium and A-teretifolia)

    Representatives of the Achillea genus are widely used as foods or nutraceuticals. Considering the increasing demand for herbal dietary supplements with health promoting effects, the objective of this research was to evaluate the biological and chemical profiles of different extracts (ethyl acetate, methanol and water) obtained from three Achillea species (A. biebersteinii, A. millefolium and A. teretifolia). The antioxidant (free radical scavenging (DPPH and ABTS), reducing power (CUPRAC and FRAP), metal chelating and phosphomolybdenum), enzyme inhibitory (anti-cholinesterase, anti-tyrosinase, anti-amylase and anti-glucosidase) and antimicrobial (antibacterial and antifungal) effects were assessed to investigate their biological profiles. Moreover, the total phenolic and flavonoid contents were determined and LC-MS analysis was performed for the chemical profile of the investigated extracts. The LC-MS analysis revealed the presence of several caffeoylquinic acids in these extracts. Generally, the methanol and water extracts exhibited stronger antioxidant abilities, which correlated with the higher levels of phenolic compounds when compared to the ethyl acetate extracts. In addition, the best antimicrobial activities were obtained for the ethyl acetate and methanolic extracts. However, the ethyl acetate extract had remarkable enzyme inhibitory potential. On the basis of our results, Achillea species may be promoted as promising sources of natural agents and used for the development of nutraceuticals or functional food ingredients.
