dc.description.abstractEffectiveness of the various stalning methods to detect abnormal sperm types and rates in fresh and frozen·thawed dog semen were studied. Semen samples were collected by digital manipulation. Abnormal sperm rates ollresh semen samples were deıermined lollowing Ihe fixaıion in Hanrock solution or in the smears stained by eosinnigrosin, hematoksilen-eosln and Caserette stains. Morphological examinalion ol the semen sanyles were also con· ducted lollowing Ireezing and thawing in Tris-lructose extender in 0.25 ml of French straw. Analyses ol lhe data shawed Ihat there is no slatistic difference (p>O.OS) among the fixation and stain groups to determine abnormal sperm types and rales of fresh or Irozen·thawed dog semen. As a conclusion; in addition lo Haneock solution. dilleren! sla· ining methods can be used to delect morphologically abnormal sperm in Iresh and frozen-thawed dog semen.en_US
dc.description.abstractBu çalışmada, köpeklerde taze ve donmuş-çözünmüş spermada anormal spermatozoon oranı ve tiplerinin tespit edilmesinde larldı Hancock solusyonu, Eosin·nigrosin, Hematoksilen·eosin ve Caserett boyaları kullanılarak anormal spematozoon oran· lan belirlendi. Ejakülatın geri kalan kısmı Tris-Frukloz sulandırıcısı kullanılarak 0.25 ml'lik payeller içerisinde don· duruldu. Payeller çözdürüldüklen sonra taze spermada anormal spermatozoon oranını tespit etmek amacı ile yapılan tüm muayeneler aynı yöntemler kullanılarak tekrar edildi. Anormal spermaıozoon oranlarının tespiti için kullanılan yöntemler arasında Istatistiki lark gözlenmedi (p>0.05). Sonuç olarak; taze ve donmuş-çôzünmüş spermada anormal spermatozoon oranlannın belirlenebilmesi Için Hancock solusyonu yanında farklı boyama yönıemlerinin de başarıyla kullanılabileceği kanısına varıldı.en_US
