Kedi ve köpeklerde helicobacter pylori' nin dot-immunobinding assay ve polimeraz zincir reaksiyonu ile teşhisi

Özet
Bu çalışma ile kedi ve köpeklerin mide mukoza örneklerinde Helicobacter py/or/nin kültür, mikroskopi, direk üreaz testi, Dot-lmmunobinding Assay (DIA) ve Polimeraz Zincir Reaksiyonu (PZR) ile teşhisi amaçlandı. Bu amaçla endoskop yardımıyla 41 köpeğin antrum ve aynı köpeklerin 40'ının korpus ve kardiasından biyopsi örneği, nekropsi ile 9 köpek ve 2 kedi midesinin antrum, korpus ve kardiasından mukoza örnekleri alındı. Ayrıca DIA testi için 39 köpekten kan örnekleri alındı. Kedi ve köpeklerden alınan 154 mide mukoza örneğinden H. pylori veya diğer Helicobacter türleri izole edilemedi. Ancak örneklerin 59'u (% 38) direk üreaz testi ile Helicobacter türleri yönünden pozitif bulundu. DIA ile incelenen 39 köpek serum örneğinin tümü Helicobacter spp. antikorlar! yönünden pozitif bulundu. PZR ile incelenen 148 köpek ve 6 kedi mide mukoza örneğinden, sadece bir köpeğe ait mide korpusunda H. pylori pozitifliği saptandı. Sonuç olarak bu çalışma ile, kedi ve köpeklerde gastrik Helicobacter spp. türlerinin bulunabileceği, ancak bunların çoğunlukla kültüre edilemeyen Helicobacter türleri olabileceği kanısına varıldı.
By this study, the presence of H. pylori in the gastric mucosa samples of cats and dogs were investigated by culture, microscopy, direct urease test, Dot-lmmunobinding Assay (DIA) and Polymerase Chain Reaction (PCR). For this reason, biopsy samples from antrum of 41 dogs and from corpus and cardia of 40 dogs by endoscopy and from antrum, corpus, cardia of 9 dogs and 2 cats by necropsy were taken three times. Blood samples from the 39 dogs were taken for serological examination by DIA test. Neither H. pylori nor any other Helicobacter spp. could be isolated from the animals used in the study. Positivity for Helicobacter spp. by the direct urease test were found in 59 (38 %) of 154 gastric mucosa samples. All serum samples of dogs were found to be positive by DIA. Mucosa samples of the dogs (n148) and cats (n6) were analysed using CAM-2 and CAM-4 primers by PCR, and produced only one positive result from a dog corpus. In conclusion, this study reports that the gastric Helicobacter spp. which can be strains that cannot be cultured found in the stomach of dogs and cats.
