Öğe Behçet hastalarında asimetrik dimetilarginin düzeyleri(2014) Öztürk, Bahadır; Vatansev, Hüsamettin; Taner, Alpaslan; Yılmaz, Sema; Dağlı, Mehmet; Ünlü, AliAmaç: Behçet Hastalığı (BH), etiyolojisi tam olarak anlaşılmamış, kronik, inflamatuvar multisistemik bir vaskülittir. Nitrik Oksit (NO), endotel hücrelerinde Nitrik Oksit Sentaz (NOS) enzimi tarafından sentezlenen, vasküler sistem için önemi olan bir moleküldür. Asimetrik dimetilarjinin (ADMA) NOS'ın endojen inhibitörüdür. Bu çalışmada BH'larında ADMA düzeyleri ve ataklarla ilgisinin araştırılması amaçlandı.Metod: 34 Behçet hastası ve 34 sağlıklı kontrol çalışmaya dâhil edildi. Serum ADMA, arjinin ve sitrülin düzeyleri HPLC yöntemi ile saptandı. ESR ve CRP düzeyleri rutin yöntemlerle analiz edildi.Bulgular: Literatür bilgilerine ters olarak Behçet hastalarında ADMA düzeyleri kontrol grubuna göre düşük bulundu. Bunun aksine Arjinin ve Sitrülin düzeyleri Behçet hastalarında anlamlı derecede yüksek bulundu. Hasta grubunda Arjinin/ADMA oranları yüksek bulundu. ADMA ve CRP düzeyleri arasında ters bir korelasyon vardı. Arjinin değerleri de sitrülin ve CRP düzeyleri ile koreleydi. Sonuç: BH da genç yaşta atakların sayısının fazla olmasının sebeblerinden birisi düşük ADMA düzeyleri olabilir. Our aim in this study was to contribute to further studies by investigating the biochemical and hematological profile of Wistar rats raised under standard local conditions at the Selcuk University Experimental Medicine Research and Application Center (SUDAM), Konya, Turkey. Materials and Methods: The study included 1 (male=15, female=15), 3 (male=15, female=15), 6 (male=15, female=15), and 12-month-old (male=15, female=15) Wistar rats living their normal lives in the Animal House of SUDAM. Selected biochemical and hematological parameters were measured with the Beckman AU 5800 and Beckman Coulter LH-780 analyzers, respectively. Results: Urea, creatinine, ALT, albumin, globulin, RBC, hemoglobin, hematocrit, and MCH levels differed significantly between rats according to months (p<0.05). When the selected biochemical and hematological parameters were compared according to gender, it was found that glucose, urea, ALP, amylase, albumin, total protein, CK, cholesterol, triglyceride, sodium, potassium, RBC, hematocrit, MCH, platelet, WBC, neutrophil, monocyte, eosinophil, basophil levels showed a statistically significant difference (p<0.05). Conclusion: This study proved that biochemical and hematological parameters of Wistar rats may alter with factors such as age, gender, environment, and nutrition.Öğe Comparative effects of atorvastatin 80 mg and rosuvastatin 40 mg on the levels of serum endocan, chemerin, and galectin-3 in patients with acute myocardial infarction(TURKISH SOC CARDIOLOGY, 2019) Tunçez, Abdullah; Altunkeser, Bülent Behlül; Öztürk, Bahadır; Ateş, Muhammed Salih; Tezcan, Hüseyin; Aydoğan, Canan; Kırık, Emre Can; Yalçın, Ulvi; Aygül, Nazif; Demir, Kenan; Akyürek, FikretObjective: Endocan, chemerin, and galectin-3 are discrete biomarkers associated with cardiovascular diseases and acting through different pathophysiological pathways. The aim of this study is to investigate and compare the effects of high doses of atorvastatin and rosuvastatin on serum endocan, chemerin, and galectin-3 levels in patients with acute myocardial infarction (AMI). Methods: Sixty-three patients with AMI were randomized to receive atorvastatin (80 mg/day) or rosuvastatin (40 mg/day) after percutaneous revascularization. Serum levels of endocan, chemerin, and galectin-3 were evaluated at baseline and after 4-week therapy. Results: Endocan levels were not decreased statistically significantly with atorvastatin 80 mg, but rosuvastatin 40 mg markedly decreased the levels of endocan according to baseline [from 110.27 (86.03-143.69) pg/mL to 99.22 (78.30-122.87) pg/mL with atorvastatin 80 mg and from 110.73 (77.28-165.22) pg/mL to 93.40 (70.48-115.13) pg/mL with rosuvastatin 40 mg, p=0.242 for atorvastatin 80 mg and p=0.014 for rosuvastatin 40 mg]. Chemerin levels significantly decreased in both groups according to baseline [from 264.90 (196.00-525.95) ng/mL to 135.00 (105.95-225.65) ng/mL with atorvastatin 80 mg and from 309.95 (168.87-701.27) ng/mL to 121.25 (86.60-212.65) ng/mL with rosuvastatin 40 mg, p<0.001, respectively, for both groups]. Galectin-3 levels did not change markedly with atorvastatin 80 mg, but they decreased with rosuvastatin 40 mg [from 17.00 (13.10-22.25) ng/mL to 19.30 (15.25-23.45) ng/mL with atorvastatin 80 mg, p=0.721, and from 18.25 (12.82-23.82) ng/mL to 16.60 (10.60-20.15) ng/mL with rosuvastatin 40 mg, p=0.074]. There were no significant between-group differences in terms of absolute and percentage changes of endocan, chemerin, and galectin-3 at 4 weeks. In this study, we compared two automated immunoassay methods, the Abbott Architect 25-OH Vitamin D assay and the Roche Cobas Vitamin D total assay, with the liquid chromatography-tandem mass spectrometry (LC-MS/MS). Materials and methods: One hundred venous blood samples were randomly selected from routine vitamin D tests. Two of the serum aliquots were analyzed at the Abbott Architect i2000 and the Roche Cobas 6000's module e601 in our laboratory within the same day. The other serum aliquots were analyzed at the LC-MS/MS in different laboratory. Passing-Bablok regression analysis and Bland-Altman plot were used to compare methods. Inter-rater agreement was analyzed using kappa (kappa) analysis. Results: The Roche assay showed acceptable agreement with the LC-MS/ MS based on Passing-Bablok analysis (intercept: -5.23 nmol/L, 95% CI: -8.73 to 0.19; slope: 0.97, 95% CI: 0.77 to 1.15). The Abbott assay showed proportional (slope: 0.77, 95% CI: 0.67 to 0.85) and constant differences (intercept: 17.08 nmol/L; 95% CI: 12.98 to 21.39). A mean bias of 15.1% was observed for the Abbott and a mean bias of -14.1% was observed for the Roche based on the Bland-Altman plots. We found strong to nearly perfect agreement in vitamin D status between the immunoassays and LC-MS/MS. (kappa: 0.83 for Abbott, kappa: 0.93 for Roche) using kappa analysis. Conclusion: Both immunoassays demonstrated acceptable performance, but the Roche Cobas assay demonstrated better performance than the Abbott Architect in the studied samples.Öğe Comparison of electrocautery and plasmablade on ischemia and seroma formation after modified radical mastectomy for locally advanced breast cancer(PAGEPRESS PUBL, 2017) Alptekin, Hüsnü; Yılmaz, Hüseyin; Öztürk, Bahadır; Ece, İlhan; Kafalı, Mehmet Ertuğrul; Acar, FahrettinThe aim of this study was to compare postoperative drainage volumes and IMA levels in patients who underwent modified radical mastectomy (MRM) with using PlasmaBlade (PB) or electrocautery (EC). A total of 36 patients who underwent MRM with PB or EC in our clinic between August 2012 to February 2013 were enrolled. Number of removed and positive lymph nodes, duration of drainage and total drainage volume was recorded. Seroma formation after drain removal and number of aspirations were also recorded. Serum ischemia modified albumine (IMA) levels were analysed before surgery, 1 hour and 24 hour after surgery. In total, 36 patients were treated with MRM in the study period. Of the 36 patients, 16 underwent MRM with PB, and 20 underwent MRM with EC. The patients demographics were similar in both groups. The mean drainage volume and seroma formation were significantly higher in the PB group when compared with EC group (P<0.05). Number of aspirations due to the seroma were also high in PB group. The total aspiration volume of seroma was not different in both groups. IMA levels 24 hours after surgery in the PB group was significantly higher than EC group. There was no statistical significance between the groups for IMA levels at 1st hour. PB is a monopolar energy device and is associated with increased levels of ischemia. This situation resulted with an increased volume of total axillary drainage and elevated risk of seroma formation.Öğe Effect of pinealectomy on homocysteine levels in ovariectomized rats(MAGHIRA & MAAS PUBLICATIONS, 2007) Öztekin, Esma; Öztürk, Bahadır; Moğulkoç, Rasim; Baltacı, Abdülkerim Kasım; Tiftik, Ali MuhtarOBJECTIVE: Hyperhomocysteinemia plays a role in obstetric and gynecological diseases and such diseases are seen as a consequence of an impaired methionine cycle. In the present study, it was aimed to determine the effect of estradiol and progesterone on homocysteine levels in pinealectomized and ovariectomized rats. MATERIALS AND METHODS: The study was carried out on 24 adult Spraque-Dawley female rats. Rats were divided into 4 groups as follows: Group 1: Sham ovariectomy group (Sham-Ovx), Group 2: Ovariectomized and sham pinealectomized group (Ovx-Sham-Px), Group 3: Ovariectomized and pinealectomized group (Ovx-Px), Group 4: Group which were supplemented with estradiol and progesterone after ovariectomy and pinealectomy (Ovx-Px-E+P). Serum homocysteine levels were determined after experimental period. RESULTS: Homocysteine levels in Group 3 were higher than those in Groups 1, 2 and 4 (p<0.05). Homocysteine levels in Groups 2 and 4 were higher than those in Group 1 (p<0.05). Homocysteine levels in Groups 2 and 4 were not different. CONCLUSION: The findings of the present study demonstrate that ovariectomy lead to increases homocysteine levels. Pinealectomy in addition to ovariectomy increases homocysteine levels. However, administration of estradiol and progesterone following pinealectomy prevents the increase in homocysteine levels in ovariectomized rats.Öğe The Effect of Prophylactic Melatonin Administration on Reperfusion Damage in Experimental Testis Ischemia-reperfusion(Maghira & Maas Publications, 2003) Öztürk, Ahmet; Baltacı, Abdulkerim Kasım; Moğulkoç, Rasim; Öztürk, BahadırAIM: Torsion of testis, which is a urologic emergency case, is generally treated by surgical detorsion procedure. However, the resulting reperfusion and both ipsilateral and contralateral testis damage caused thereby are important problems. This study aims at investigating the administration of prophylactic melatonin in order to reduce free radical damage that is caused due to reperfusion after experimental testis torsion-detorsion procedure. MATERIAL AND METHOD: The rats used in the study were allocated to four groups, each containing 10 rats. Rats in Group I had six hours of torsion (ischemia) followed by orchiectomy. Rats in Group II had six hours of torsion, then received melatonin (10 mg/kg/IM) and after that had detorsion (reperfusion). Rats in Group Ill had detorsion after six hours of torsion and serum physiologic administration (the same volume as melatonin). Torsion and/or detorsion procedures were not applied in Group IV (control). Ipsilateral, contralateral testis and plasma Malondialdehyde (MDA) levels were determined in all groups. RESULTS: Tissue and plasma MDA levels in the group which had detorsion were found to be significantly higher than those in the group that had orchiectomy only (p < 0.02). Ipsilateral and contralateral testis MDA levels were identified to be significantly lower in the group receiving prophylactic melatonin in comparison to the group receiving serum physiologic (p < 0. 02). Plasma and contralateral MDA levels correlated positively with MDA levels identified in ipsilateral testis in all groups (rs= +0.89, n=40, p=0.000). CONCLUSION: In cases where testis torsion is identified, administration of melatonin just before torsion may reduce local and systemic free radical damage.Öğe The effects of chrysin in an experimental model of acute pancreatitis(Edizioni Luigi Pozzi, 2020) Alptekin, Hüsnü; Çalışır, Akın; Öztürk, Bahadır; Uğurlu, Ceyhan; Yormaz, Serdar; Ece, İlhanPURPOSE: This experimental study was conducted to evaluate the possible effects of orally administered chrysin on acute pancreatitis. MATERIAL AND METHOD: Twenty four rats were procured. The animals were randomly divided into four groups. In Group I, only vehicle solution (5% dimethylsulfoksid) was administered, and in Group II, chrysin dissolved in the vehicle solution was administered for six days. In Group III and Group IV cerulein was administered to induce acute pancreatitis. In Group III, only vehicle solution was administered, and in Group IV, chrysin dissolved in the vehicle solution was administered orally for six days. Blood samples were analyzed and the pancreatic tissue specimens were evaluated for histopathological examination. RESULTS: Group III and Group IV, exhibited markedly higher levels of serum WBC, amylase, and lipase, compared with Groups I and II. In the pancreatitis induced groups, CRP and TOS values were found to be significantly higher. In Group II and Group IV, TAS values were significantly higher. The highest calculated OSI values were observed in Group III. Group IV OSI values were significantly lower than those in Group III and even in Group I. Noticeable histopathological changes were identified in the pancreatitis induced Groups III and IV. Compared with Group III, the extent and severity of pancreatic injuries were markedly lower in Group IV. CONCLUSION: Chrysin application reduced oxidative stress and histopathological parameters. Estrogen is considered a nongenetic factor influencing homocysteine metabolism (Pines et al., 1997) Homocysteine levels can be a risk factor for arteriosclerosis and osteoporosis (El-Swefy et al., 2002). El-Swefy et al., (loc. cit) found that high homocysteine levels in ovariectomized rats decreased with estradiol and folic acid supplementation. Methods: A total of 34 Wistar albino male rats were randomly divided into 5 groups as: Group C (control, n= 6), Group S (sham; underwent only left scrotal exploration, n= 7), Group TD (torsion and detorsion; 6 h of ischemia and 7 days of reperfusion, n= 7), Group TD/M (TD + Melatonin; 6 h of ischemia and 7 days of reperfusion and 7 days of 17 mg/kg intraperitoneal melatonin per day, n = 7), group TD/Col (TD + Colchicine; 6 h of ischemia and 7 days of reperfusion and 7 days of 1 mg/kg oral colchicine per day, n = 7). Histopathologic evaluation of seminiferous tubule deterioration was performed by Johnsen's scoring system. Total antioxidant status (TAS), total oxidant status (TOS), IL-6, TNF alpha levels were analyzed in each group. Results: The histopathologic scores, total antioxidant status (TAS), total oxidant status (TOS), IL-6, TNF alpha levels in groups C and TD/Col were significantly lower than groups TD and TD/M (P < .001). Conclusion: Our study results revealed that colchicine reduced testicular ischemia-reperfusion injury in experimental rat testis torsion model. Although detorsion of testis is crucial for the preserving the testicular viability, antioxidant and anti-inflammatory treatment modalities like colchicine might help to reduce ischemia-reperfusion injury in detorsed testis. Oral glukoz tolerans testi sonuçlarına göre 57 gebeye gestasyonel diabet mellitus tanısı konuldu. 85 sağlıklı gebe ise kontrol grubuna dahil edildi. Serum analizi için tüm kan örnekleri bir gecelik açlık sonrası oral glukoz tolerans testi başlangıcında sabah 7:00 ve 8:00 saatleri arasında alındı.Bulgular: Gestasyonel diabet mellitus grubunda serum prokalsitonin düzeyleri kontrol grubuna göre anlamlı olarak yüksek bulundu. Serum prokalsitonin düzeyi ile açlık glukozu ve oral glukoz tolerans testi 60. dakika glukoz düzeyleri arasında anlamlı pozitif korelasyon bulundu.Sonuç: Bu çalışmada gestasyonel diabet mellitus’lu gebelerde serum prokalsitonin düzeylerinin sağlıklı gebelere göre anlamlı olarak yüksek olduğu ilk kez gösterilmiştir. Bu sonuç kronik düşük-dereceli inflamasyonun gestasyonel diabet mellitus patogenezinde önemli role sahip olduğu bilgisini desteklemektedir. Various studies have revealed that added sugar consumption may be involved in the pathogenesis of cardiovascular, metabolic, and neurocognitive disorders by triggering subclinical inflammation. The imbalance in the kynurenine pathway metabolites may be associated with inflammation and oxidative stress. This study aims to investigate the effect of high-fructose corn syrup-55 (HFCS-55), invert sugar and sucrose intervention on the kynurenine pathway metabolite levels (tryptophan, kynurenine, 3-hydroxykynurenine, 3-hydroxyanthranilic acid, quinolinic acid, and kynurenic acid) in Wistar rats. Material and Methods: Twenty-four Wistar male rats (8-12 weeks old, weighing 300-350 g) were included in the study. After one week of conditioning, the animals were randomly divided into four groups: chow diet and tap water (control, n = 6), chow diet and tap water including 10% HFCS-55 (55% sucrose, 45% glucose), chow diet and tap water including 10% sucrose, chow diet and tap water including 10% invert sugar (33% sucrose, 66% glucose and fructose). At the end of the 3-month experimental period, serum kynurenine metabolites levels were measured by tandem mass spectrometry. Results: Serum kynurenine levels and kynurenine/tryptophan ratio were significantly higher (p<0.05) and serum kynurenic acid levels were significantly lower (p<0.05) in rats fed with HFCS, sucrose and invert sugar compared to the control group. Conclusion: Our findings suggest that consumption of added sugar may lead to an imbalance in the kynurenine pathway metabolites. Patients with diagnosed ACS (n = 100) were included in the study group. Control subjects (n = 100) were selected from healthy volunteers in the same age range. Venous blood samples were obtained to evaluate the levels of Se, Zn, Cu, and TnI (on admission for the ACS group). Serum Se, Zn, and Cu levels were significantly less in the ACS group compared to the control group (p < 0.001, p < 0.01, and p < 0.001, respectively). The serum TnI level was significantly greater in the ACS group compared to the control group (p < 0.05). The serum Se level in the ACS group correlated significantly with the TnI level (r = -0.211, p = 0.035). These results indicate that Se, Zn, and Cu deficiencies may be risk factors for ACS, and a decreased serum Se level in patients with ACS might reflect the degree of myocardial necrosis.Öğe Malign-nonmalign plevral efüzyon ayırıcı tanısında IGF ve IGFBP’lerin rolü(2004) Gederet, Yavuz T.; Öztürk, Bahadır; Karagözoğlu, Emrah; Gök, Mehmet; Tiftik, A. MuhtarAmaç: IGF’ler, Büyüme Hormonunun (GH) periferik etkilerine aracılık eden polipeptidlerdir. IGF’lerin, hücre büyümesi ve proliferasyonunda önemli etkileri vardır. Son yıllarda yapılan çalışmalarda IGF’lerin malign proliferasyonda da rolleri olduğu ortaya çıkmıştır. Çalışmamızda IGF ve bağlayıcı proteinlerin akciğer kanseri ve nonspesifik plöritiste seviyeleri ve malignite ile ilişkisi araştırıldı. Yöntem: Bu çalışmaya değişik nedenli plevral efüzyonu bulunan toplam 40 hasta alındı. Hastalar, konulan tanılara göre akciğer kanseri ve nonspesifik plörezi şeklinde iki gruba ayrıldı. Her bir grupta 20 hasta vardı. Bu hastalardan alınan serum ve plevral sıvı örneklerinde IGF I-II, IGFBP I-II analizleri yapıldı. Bulgular: Serumda IGFBP-I ve IGFBP-II, malign grupta anlamlı olmasa da yüksek iken, plevral sıvıda IGFBP-I düşük, IGFBP-II ise yüksek bulundu. Serum IGF-II ve plevral sıvı IGF-II seviyesi malign grupta anlamlı şekilde yüksek bulundu. Plevral sıvı IGF-I seviyesi malign grupta anlamlı şekilde yüksek bulundu. Sonuç: IGF’lerin etkilerine aracılık eden bağlayıcı proteinlerin malignitede yükseldiğini, plevral sıvı değerleri açısından da farklılık gösterdiğini tespit ettik. Materials and methods: The serum PAPP-A, IMA, procalcitonin, and troponin I levels were measured in 100 patients with acute coronary syndrome admitted to the emergency department and 100 healthy control subjects. Results: Patients with acute coronary syndrome had significantly greater mean serum PAPP-A (patients, 10 +/- 10 mIU/L; control subjects, 6 +/- 10 mIU/L; P <= 0.001), procalcitonin (patients, 2 +/- 10 mu g/L; control subjects, 0.4 +/- 2 mu g/L; P <= 0.001), and troponin I levels (patients, 6 +/- 8 mu g/L; control subjects, 0.2 +/- 0.3 mu g/L; P <= 0.001) than control subjects. There was no difference in mean IMA levels between patients and control subjects. There were no significant correlations between PAPP-A levels and IMA, procalcitonin, or troponin I levels in patients with acute coronary syndrome. Conclusion: The PAPP-A, procalcitonin, and troponin I levels were increased in patients with acute coronary syndrome. Therefore, elevated PAPP-A and procalcitonin levels, in addition to troponin I levels, may be useful markers of myocardial injury on admission to the emergency department.Öğe Romatoid artritli hastalarda asimetrik dimetilarjinin ve arjinin düzeyleri(2013) Vatansev, Hüsamettin; Öztürk, Bahadır; Yılmaz, Sema; Sivrikaya, Abdullah; Dağlı, Mehmet; Kıyıcı, Aysel; Ünlü, AliAmaç: Romatoid artrit (RA) ataklar ile seyreden kronik inflamatuar bir hastalıktır ve sıklıkla erken ateroskleroz gelişimi ile birliktedir. RA hastalarında erken ateroskleroz gelişimini hızlandıracak birçok faktör bulunmaktadır. Yüksek serum asimetrik dimetil arjinin (ADMA) düzeyleri endotel bütünlüğünü ve fonksiyonunu bozarak ateroskleroz gelişimini hızlandıran bağımsız bir faktör olarak kabul edilmektedir. Çalışmamızda RA hastalarında ADMA, arjinin ve sitrülin düzeylerini, hastalık aktivasyonundaki olası rolü içinde aktivite parametreleri ile ilişkisini araştırmayı amaçladık. Yöntemler: RA tanısı almış ve romatoloji kliniğinin takibinde olan 92 RA hastası ve bilinen herhangi bir hastalığı olmayan 34 sağlıklı kontrol çalışmaya dâhil edildi. Serum ADMA, arjinin ve sitrülin düzeyleri pre-kolon derivatizasyon-HPLC yöntemi ile saptandı. Rutin takip parametreleri olarak kullanılan eritrosit sedimantasyon hızı ve CRP değerleri hasta dosyasından alındı. Bulgular: ADMA düzeyleri RA hasta grubunda anlamlı derecede artarken, arjinin düzeylerinde azalma saptandı. Sitrülin seviyelerinde anlamlı bir değişikliğe rastlanmadı. Arjinin ve sitrülin arasında anlamlı zayıf bir korelasyon saptanırken, ADMA, arjinin ve sitrülin ile CRP, sedimantasyon hızı, hastalık şiddeti ve süresi arasında önemli bir ilişki saptanmadı. Sonuç: Bulgularımızda yüksek ADMA düzeylerinin azalan arjinin düzeyleri ile birlikte olmasının romatoid artritte artan arjinin-ADMA dönüşümünün bir göstergesi olabileceği yanında, bu hastalarda artan prematür ateroskleroz gelişiminde de rol oynayabileceğini göstermektedir. Yüksek ADMA düzeylerinin NO bağımlı vazodilatasyonu engellemesi nedeni ile prematür ateroskleroz gelişiminde, ADMA'nın rolünün inflamasyondan bağımsız olabileceğini düşündürmektedir
