Methods: We cared for the animals in the Pharmacology Department of the Medical School of Selcuk University in 2004. We used 30, New Zealand, 7-day-old male rabbits in the experiment. We anesthetized all the animals with xylazine hydrochloric acid and ketamine. We divided the rabbits equally into 3 groups. In group 1 (n=10) (sham group), we observed baseline levels, and did not apply ischemia. In group 2 (n=10) (untreated group) we produced cerebral ischemia by clamping the bilateral common carotid arteries for 60 minutes, and in group 3 (n=10), we administered deferoxamine (DFO) 50 mg/kg intravenously immediately after opening the clamps. Results: The Na+-K(+)ATPase activity increased after DFO treatment (p=0.045). Conclusion: We conclude that Na+-K(+)ATPase activity in cortical brain tissue was higher in DFO-treated rabbits compared with untreated animals after ischemia.Öğe Effects of the Trans-9 18:1 Octadecenoic Acid Isomer on Rat Liver Membrane Na+/K+ ATPase Enzyme Activity Plasma Lipoproteins(2010) Kaya, Ayşe; Topçu, Cemile; Gürbilek, Mehmet; Aköz, Mehmet; Ünaldı, M.Purpose: The consumption of trans-fatty acids is associated with increasing coronary heart disease risk. When oils are partially hydrogenised to margarine form, trans isomers of fatty acids are formed. In this study we investigated the effects of dietary trans-9 18:1 octadecenoic acid isomer on liver cell membrane Na+/K+ ATPase enzyme activity, fat composition and lipid metabolism. Method and Material: This study was conducted on two groups of rats, study and control. Study group was given 50 mg/day trans-9 18:1 octadecenoic acid isomers for ten days. Fatty acid composition of serum and liver tissue was analyzed using gas chromatography Results: Study group showed trans-9 18:1 fatty acid presence while control group did not, thus trans-9 18:1 octadecenoic acid isomer is not a naturally occurring compound. The meaningful increase of C18:2 6n fatty acid and meaningful decrease of C20:4 6n in study group show that trans-9 18:1 octadecenoic acid isomers affected desaturation activity and prevented C18:2 6n fatty acid from turning into C20:4 6n. Rats fed with trans-9 18:1 octadecenoic acid showed decreased HDL and HDL2 cholesterol levels. No change was observed in HDL3, LDL, total cholesterol, triglyceride and Na+/K+ ATPase activity. Our findings combined with current literature allow us to conclude; sine TFAs change serum HDL and HDL2 cholesterol levels, nutrition's containing TFA may increase the risk of heart disease. 2010 © TurkJBiochem.com.Öğe The effects of trans-9 18:1 octadecenoic acid isomer on levels of sICAM-1, sVICAM-1 and IGF-1(2011) Aköz, Mehmet; Kocabaş, Rahim; Topçu, Cemile; Gürbilek, MehmetObjective: The effects of trans-9 18:1 octadecenoic acid isomer from the fatty acid isomers taken with diet on soluble cell adhesion molecules (sICAM-1: Soluble intercellular adhesion molecule-1 and svCAM-1: Soluble vascular cell adhesion molecule-1) and insulin like growth factor-1 (IGF-1) levels were investigated on rats. Method: The rats which were fed for two weeks by the same diet were separated into experimental and control groups. Fifty mg/day trans-9 18:1 octadecenoic acid isomer was added to the diet of experimental group for 10 days. Following the completion of special diet, fatty acid composition in both groups was studied by gas chromatography analysis. Results: Trans-9 18:1 fatty acid level in the experimental group was higher than control group (p<0.01). The soluble cell adhesion molecules (sICAM-1 and svCAM-1) increased (p<0.01) and IGF-1 levels decreased (p<0.05) in rats fed with trans-9 18:1 octadecenoic acid. Conclusion: Our results indicate that, high levels of dietary trans fatty acid isomers are a factor in the increasing of cell adhesion molecules (sICAM-1, svCAM-1) levels and decreasing of IGF-1 levels. Hence, measurement of these parameters may be beneficial in patients with coronary artery disease and atherosclerosis, as well as in other pathological cases. We believe that these parameters may also be useful for early diagnosis.Öğe The Investigation of Relationship Between Na+/k +atpase Enzyme Activity, Atp1a1 Gene Polymorphism and C-Peptide in Type 2 Diabetic Patients With Neuropathy(2009) Topçu, Cemile; Gürbilek, Mehmet; Aköz, Mehmet; Çora, TülinObjectives: Na+/K+-ATPase is encoded by various genes, of which the ATP1A1 gene is expressed predominantly in peripheral nerves and in erythrocytes. We investigated whether the ATP1A1 polymorphism is associated with Na+/K+-ATPase activity modifications in healthy subjects and in type 2 diabetic patients with polyneuropathy. We also tested whether C-peptide level could influence the relationship between ATP1A1 polymorphism and Na+/K+-ATPase activity. Methods: Na+/K +-ATPase polymorphism has been determined by using polymerasechain reactions and restriction fragment length polymorphism methods. The erythrocyte membrane Na+/K+-ATPase activity measurement was performed with modified Kitao-Hattori method. C-peptide measurements were performed using competitive chemiluminesence enzyme immunoassay method. Results: Na +/K+-ATPase activity was significantly lower in diabetic polyneuropathic patients than in healthy subjects. Correlation between C-peptide levels and Na+/K+-ATPase activity was not detected. All diabetic patients with polyneuropathy and healthy controls were homozygous for the unrestricted allele. Conclusion: Oxidative stress which develops in diabetes causes lipid peroxidation in the membrane and consequently Na +/K+-ATPase activity decreases. Low enzymatic activity leads to decrease in nerve conduction velocity. Therefore, we believe that better regulation of diabetes can abrogate complications and prevent the development of neuropathy.Öğe Tip 2 diyabetik nöropatili hastalarda eritrosit zarı Na / K ATPaz enzim aktivitesi, ATP1A1 gen polimorfizmi ile C-peptid arasındaki ilişkinin araştırılması(2009) Topçu, Cemile; Gürbilek, Mehmet; Aköz, Mehmet; Çora, TülinAmaç: Na/K-ATPaz çeşitli genler tarafından kodlanır, bunlardan ATP1A1 geni pe-riferal sinirlerde ve eritrositlerde baskın olarak eksprese edilir. Bu çalışmada Tip 2 di-yabetik polinöropatili hastalarda ve sağlıklı bireylerde ATP1A1 gen polimorfizminin, Na/K-ATPaz enzim aktivitesindeki modifikasyonlarla ilişkisi olup olmadığı araştırıl- dı. Aynı zamanda C-peptid seviyelerinin ATP1A1 gen polimorfizmi ve Na /K -ATPaz enzim aktivitesi arasındaki ilişkiye etkisi araştırıldı. Metot: Na/K -ATPaz gen polimorfizmi, polimeraz zincir reaksiyonu ve restriksiyon fragmenti uzunluk polimorfizmi yöntemleriyle belirlendi. Eritrosit membranı Na /K - ATPaz aktivitesi modifiye edilmiş Kitao-Hattori metodu ile C-peptid düzeyleri kemilu-minesans enzim immunoassay metodu ile ölçüldü. Bulgular: Na /K -ATPaz aktivitesi diyabetik nöropatili hastalarda sağlıklı kontrollere göre düşük bulundu. Na /K -ATPaz aktivitesi ve C-peptid seviyesi arasında bir korelasyon bulunamadı. Tüm diyabetik polinöropatili hastalar ve sağlıklı kontrol grubu, kesilmemiş allel için homozigot olarak bulundu. Bu hastalarda ATP1A1 gen polimorfizminin bulunmaması nöropatiye yatkınlığın olmadığını düşündürmektedir. Sonuç: Diyabette oksidatif stres membranda lipid peroksidasyonuna sebep olmakta ve bunun sonucunda Na /K -ATPaz aktivitesi düşmektedir. Düşük enzimatik aktivite sinir iletim hızının azalmasına neden olmaktadır.
